Toll-like receptors (TLRs) are promising targets for treatment of viral infections, autoimmune diseases, and cancers. Here, two new series of selective small-molecule TLR7 agonists with novel scaffolds and good selectivity over TLR8 are described, some with potencies in the low micromolar range. 8-Hydroxy-1-isobutylchromeno[3,4-d]imidazol-4(1H)-one (26) from the first series was designed and synthesized on the basis of previously described TLR7 antagonist 2, and is shown to be a selective TLR7 agonist (EC50, 1.8 mu M). The second series was based on 2-(trifluoromethyl)quinolin-4-amine and 2-(trifluoromethyl) quinazolin-4-amine scaffolds, which were defined according to our in-house ligand-based virtual screening protocol. Further synthesis of a focused library of analogs, biological evaluation, and docking studies provided systematic exploration of the structure-activity relationships, which indicate that a secondary or tertiary amine with smaller flexible alkyl substituents up to three carbon atoms in length, or bulkier rigid aliphatic rings is required at position 4 on 2-(trifluoromethyl)quinoline/quinazoline scaffold for potent TLR7 agonist activity. The influence of selected TLR7 agonists on cytokine production is also reported showing that N-cyclopropyl-2-(trifluoromethyl)quinazolin-4-amine (46) is able to induce increased levels of IL-6 and IL-8. These data demonstrate successful in-silico definition of novel TLR7 versus TLR8-selective compounds as promising chemical probes for further development of potent small-molecule immunomodulators. Background: Nivolumab was the first immune checkpoint inhibitor approved for previously treated advanced nonesmall cell lung cancer (NSCLC). Before its introduction in the market, nivolumab was made available to NSCLC patients through an expanded access program (EAP). Here we present the Italian cohort of patients with non-squamous NSCLC enrolled in a worldwide nivolumab EAP, with subgroup analyses involving elderly patients, patients with central nervous system (CNS) metastases and patients receiving nivolumab beyond progression. Methods: Pretreated patients with advanced non-squamous NSCLC received nivolumab at 3 mg/kg every 2 weeks up to 24 months. Efficacy data (investigator-assessed tumour response, progression date and survival) and safety data were collected. Findings: 1588 patients were treated across 153 Italian centres. Overall response rate and disease control rate were 18% and 44%, respectively; median overall survival (OS) was 11.3 months (95% CI: 10.2-12.4). Elderly patients (>= 70 n = 522; >= 75 n = 232) achieved outcomes similar to the global study population; patients with CNS metastases (n = 409) had an OS of 8.6 months (95% CI: 6.4-10.8), and a 1-year OS rate of 43%. Nivolumab was administered beyond progression to 276 patients (26%), 57 of whom achieved subsequent disease control; the median OS of patients receiving nivolumab beyond progression was 16.2 months (95% CI: 14.0-18.4), while 1-year OS rate was 62%. Interpretation: To date, this is the largest clinical experience with nivolumab in a real-world setting. 10,10-Bis[4-(4-aminophenoxy)-3-methylphenyl]-9(10H)-anthrone, as a new aromatic diamine, was synthesized from 9(10H)-anthrone by three-step procedure. Low-temperature polycondensation of this diamine with various aromatic dicarboxylic acid dichlorides produced a series of new cardo poly(ether amide)s with moderate to high inherent viscosities of 0.86-1.43 dL g(-1)while the weight-average and number-average molecular weights were in the range of 48,600-59,800 and 29,800-35,600 g mol(-1), respectively. All the polymers were readily soluble in polar solvents such asN,N-dimethylacetamide (DMAc),N,N-dimethylformamide,N-methyl-2-pyrrolidinone, dimethyl sulfoxide, and pyridine, and afforded transparent, flexible, and strong films upon casting from DMAc solvent. These polymers had glass transition temperatures of 254-297 degrees C, 10% weight loss temperatures of 493-505 degrees C, and char yields of 61-64% at 800 degrees C in nitrogen. All polymers were amorphous and their films exhibited tensile strength of 80.7-101.4 MPa, elongation at break of 8.0-12.8%, and tensile modulus of 1.9-2.6 GPa. These polymers had low moisture uptake in the range of 2.13-4.02% and high transparency with an ultraviolet-visible absorption cutoff wavelength in the 322-375 nm range.

Imitating the structures and behaviors of natural creatures is of great significance to scientists to explore novel materials for practical applications. However, the design and fabrication of biomimetic devices with complex and outstanding performances is still on the way. A bilayer film composed of liquid crystalline networks (LCN) film and hydrochromic aggregation-induced-emission molecule-doped hydrophilic layer is prepared. Under different relative humidity, the composite film can deform and change fluorescence color simultaneously. The influence of the content of the hydrophilic matrix on the fluorescent property and humidity-responsive behaviors of the bilayer film is investigated. Thanks to the mechanical anisotropy provided by uniform-aligned LCN film, different modes of deformation of the bilayer film are achievable, like bending, curling, and twisting. More importantly, due to the independence of the LCN film and the non-mesogenic molecules brought by this bimorph strategy, complex alignment of the LCN film and modification by immiscible molecules are realized in a single LCN actuator. Based on the functional composite film, artificial flowers showing synergistic blooming and shape-changing is prepared. By regulating the molecular alignment of the LC mesogens of the LCN film, the artificial flowers can imitate various blooming behaviors of natural flowers like confederate jasmine and jade lotus.

A total of 22 quinazoline thioether derivatives incorporating a 1,2,4-triazolo[4,3-a]pyridine moiety were designed, synthesized, and evaluated as antimicrobial agents in agriculture. Among these compounds, the chemical structure of compound 61 was further confirmed via single-crystal X-ray diffraction analysis. The bioassay results revealed that some of the compounds possessed noticeable in vitro antibacterial activities against the tested phytopathogenic bacteria. For example, compounds 6b and 6g had EC50 values as low as 10.0 and 24.7 mu g/mL against Xanthomonas axonopodis pv. citri (Xac), respectively, which were significantly better than that of the commercial agrobactericide bismerthiazol (56.9 mu g/mL). Particularly, compound 6b was also found to be capable of suppressing the pathogenic bacterium Xanthomonas oryzae pv. oryzae (Xoo) approximately 12-fold more potent than control bismerthiazol, in terms of their EC 50 values (7.2 versus 89.8 mu g/mL). Importantly, the most active compound 6b turned out to be one with the highest hydrophilicity and the lowest molecular weight within the series. In vivo bioassays further showed the application prospect of 6b as a promising plant bactericide for controlling Xoo. Additionally, in vitro antifungal activities of these compounds were also evaluated at the concentration of 50 mu g/mL. Overall, the present study demonstrated the potential of 1,2,4-triazolo[4,3-a]pyridine-bearing quinazoline thioether derivatives as efficient agricultural antibacterial agents for crop protection.

Flexible fiber-type solid-state supercapacitors (FSCs) are demanded for energy storage of wearable soft electronics. A flexible carbon fiber (CF) with high electrical conductivity and good energy storage ability can act as the current collector and the active material simultaneously. Improving the energy storage ability of the CF without depositing active materials is highly promising to avoid active materials from peeling off, especially under bending conditions. In this study, the CF is treated with acid mixtures containing different HNO3 to H2SO4 ratios. The roughened surface and the production of graphene quantum dots and functional groups are achieved for the acid-treated CF, especially for the CF treated with the acid mixtures. The largely enhanced specific capacitance (C-L) is first obtained for the optimized CF electrode (641.08 mF/cm at 5 mV/s) treated with the acid mixture containing the HNO3 to H2SO4 ratio of 1 to 3. The FSC also shows a maximum energy density of 16.57 mu W h/cm at the power density of 62.50 mu W/cm, as well as a C-L retention of 77% and Coulombic efficiency of 100% in 5000 times of a charge/discharge cycling process. Excellent flexibility is also attained for the FSC with no shape distortion of cyclic voltammetry curves measured under the bending angle of 180 degrees and with 1000 times of repeated bending. This work proposes a simple acid treatment to largely enhance the energy storage ability of pure CF electrodes. The bifunctional substance with current collecting and energy storing abilities is expected to be well developed in the future for applying on numerous efficient flexible energy storage devices.

A series of novel rutaecarpine derivatives were synthesized and subjected to pharmacological evaluation as PDE5 inhibitors. The structure-activity relationships were discussed and their binding conformation and simultaneous interaction mode were further clarified by the molecular docking studies. Among the 25 analogues, compound 8i exhibited most potent PDE5 inhibition with IC50 values about 0.086 mu M. Moreover, it also produced good effects against scopolamine-induced cognitive impairment in vivo. These results might bring significant instruction for further development of potential PDE5 inhibitors derived from rutaecarpine as a good candidate drug for the treatment of Alzheimer’s disease.

In this study, three aqueous extracts (APE, ACE and APE) from aged residual biomixtures in three biopurification systems (BPSs) exposed to pesticides at a pilot scale were found to improve pesticide removal. The addition of ACEs and AVEs to solutions containing the model compound diuron increased removal rates 6- and 17-fold, respectively, as compared to APEs. These extracts also increased the removal of the metabolite 3,4-dichloroaniline, while AVEs, in particular, were found to remove all pesticides within 9 days. Three metabolites less hazardous than 3,4-dichloroaniline were identified by SPME/GC/MS. AVEs, which also enhance linuron removal in liquid media, were found to increase diuron removal 6-fold in BPSs. We observed an increase in the relative abundance of taxa, such as Chloroflexi, Acidobacteria, Gemmatimonadetes, Firmicutes, Deinococcus-Thermus and especially Proteobacteria (10%), in AV biomixtures, as well as an enrichment of gamma-proteobacteria and the actinobacterial genus Dokdonella in AVEs with respect to initial noncontaminated IV biomixture. We demonstrate that extracts containing a pollutant-acclimatized microbiome could be used as part of a bioaugmentation strategy to improve the functioning of on-farm BPSs and contaminated systems. Spain is the first olive oil producer worldwide (representing around 45%), with more than 80% of its agricultural area dedicated to this crop. The goal of this study is to assess the environmental impact of the Spanish virgin olive oil production, attending to the farming and industrial phases. A Life Cycle Assessment (LCA) is conducted for 5 harvests in approximately 4.000 ha of olive grove in Jaen, the largest producer region of Spain. The type of farming is the most representative in Spain: conventional (non-organic), with a medium-low slope, extensive (100-150 trees per ha) and around 60% dryland orchards. The industrial phase is based on 2-phase extraction process and olive pomace valorization. The functional unit (FU) chosen for the comparative analysis is 1 kg of unpacked virgin olive oil under a perspective from cradle to gate. One of the most representative categories, climate change, places the average environmental impact in 2.43 kg CO2 eq/kg, while the range is between 1.93 and 3.00 kg CO2 eq/kg depending on the harvest. Huge differences between values are observed in the farming phase and they are mostly caused by the virgin olive oil yield of every harvest. The carbon sequestration analyses and the impact produced per FU ranges from 43.78% (for 15/16 harvest) to 46.36% (for 17/18). The impact hotspots detected in the farming phase, in terms of climate change potential, are the categories of plant protection products and herbicides (24.11% of the average value) and fertilizers (20.01%). However, the environmental impact of the industrial phase is relatively constant, with a value of 0.547-0.554 kg of CO2 equivalent. The 80.74% is caused by pomace valorization, which translated to the average whole value, represents the 18.56%. (C) 2020 Elsevier Ltd. All rights reserved.
