A series of novel podophyllotoxin-naphthoquinone compounds 5a-p were synthesized in good yields using microwave-assisted four-component reactions of 2-hydroxy-1,4-naphthoquinone, aromatic benzaldehydes, tetronic acid and ammonium acetate. All the synthesized compounds were fully characterized by spectral data and evaluated for their cytotoxicity activities against KB, HepG2, Lu1, MCF7, and non-cancerous Hek-293 cell lines. Among 16 new compounds screened, compounds 5a, 5d, 5h, and 5k displayed high potent inhibitory activities with IC50 < 40 nM against HepG2 and SK-Lu-1 cell lines, and showed lower toxicity for non-cancerous Hek-293 cell line, demonstrating the potential importance of these compounds in the development of potential anticancer agents. WRKY transcription factors participate in the salt stress responses of various plant species, but the underlying functional mechanisms are not well understood, particularly in grape. In this study, the role of VvWRKY13 in the salt stress response was characterized and the underlying physiological and molecular mechanisms were explored. The results show that VvWRKY13 expression was induced by salt stress, and the VvWRKY13 transcript level in the salt stress-sensitive cultivar was significantly higher than that in the resistant cultivars. Overexpression of VvWRKY13 in Arabidopsis significantly inhibited the growth of transgenic plants under salt stress, indicating that VvWRKY13 has a negative effect on the salt stress response. The photosynthetic rate, contents of proline and soluble sugars, activities of superoxide dismutase (SOD) and catalase (CAT), as well as transcriptional levels of proline and soluble sugar metabolic genes, such as P5CS1, SS1, SS2 and G6PDH, as well as SOD and CAT encoding genes CAT1, CAT2, and Cu/Zn-SOD, all decreased under salt stress, whereas hydrogen peroxide and oxygen-free radical contents increased in transgenic Arabidopsis plants compared with those of wild-type plants. VvWRKY13 suppressed upregulation of SnRK2.3, ABF1, ABF2, ABF3, RD29B, RD29A, and RAB18 induced by salt stress, but showed no significant effect on the transcript levels of SnRK2.2, SnRK2.6, or RD22, indicating that VvWRKY13 affects salt stress tolerance via a specific abscisic acid signaling pathway. Key message VvWRKY13, isolated from grapevine, negatively regulate salt stress tolerance by depressing photosynthetic capacity, inhibiting salt-induced accumulation of osmolytes andup-regulation in antioxidant activity as well as expression of related genes. The oxidation of a series of alkylaromatic compounds with H2O2 catalyzed by an imine-based non-heme iron complex prepared in situ by reaction of 2-picolylaldehyde, 2-picolylamine, and Fe(OTf)(2) in a 2 : 2 : 1 ratio leads to a marked chemoselectivity for aromatic ring hydroxylation over side-chain oxidation. This selectivity is herein investigated in detail. Side-chain/ring oxygenated product ratio was found to increase upon decreasing the bond dissociation energy (BDE) of the benzylic C-H bond in line with expectation. Evidence for competitive reactions leading either to aromatic hydroxylation via electrophilic aromatic substitution or side-chain oxidation via benzylic hydrogen atom abstraction, promoted by a metal-based oxidant, has been provided by kinetic isotope effect analysis.

Background Accurate sentinel lymph node (SLN) staging is essential for both prognosis and treatment in patients with breast cancer. However, the preoperative lymphoscintigraphy may fail to visualize the SLN in some patients. The purpose of this retrospective study was to identify risk factors associated with SLN nonvisualization on lymphoscintigraphy. For this single-center retrospective study, all data of lymphoscintigraphy of SLN procedures from March 2011 to April 2021 were collected and reviewed from the Amsterdam UMC database. Results A total of 1886 SLN procedures were included in this study. The SLN nonvisualization rate was 25.1% on lymphoscintigraphy at 4 h post-injection. The SLN nonvisualization rate decreased to 9.4% after reinjection. Multivariable analysis showed that age >= 70 years (P < 0.001; OR: 2.27; 95% CI: 1.46-3.53), BMI >= 30 kg/m(2) (P = 0.031; OR: 1.48; 95% CI: 1.04-2.12) and nonpalpable tumors (P = 0.004; OR: 1.54; 95% CI: 1.15-2.07) were independent predictors of SLN nonvisualization. Tumor location, brand of radiopharmaceutical, injected dose and volume, experience of preparer and administrator were not associated with SLN nonvisualization. None of the patient, tumor or tracer characteristics were associated with SLN nonvisualization after radiotracer reinjection. Conclusions This study shows that risk factors for SLN nonvisualization in breast cancer patients during preoperative lymphoscintigraphy are age >= 70 years, BMI >= 30 kg/m(2) and nonpalpable tumors. Our results support the notion that SLN lymphoscintigraphy is a very robust technique that does not depend on the experience of the preparer or administrator of the radiotracer.

Under the acyclic diene metathesis (ADMET) reaction condition, the C3-vinyl groups of cinchona alkaloids readily react with each other to form a C-C bond. A novel type of cinchona alkaloid polymers was synthesized from dimeric cinchona squaramides using the Hoveyda-Grubbs’ second-generation catalysts (HG(2)) by means of ADMET reaction. The chiral polymers, containing cinchona squaramide moieties in their main chains, were subsequently employed as catalysts for the enantioselective Michael reaction to give the corresponding chiral adducts in high yields with excellent enantioselectivity and diastereoselectivity. Both enantiomers from the asymmetric Michael reaction were distinctively prepared while using the polymeric catalysts, possessing pseudoenantiomeric structures. The catalysts were readily recovered from the reaction mixture and recycled several times due to the insolubility of the cinchona-based squaramide polymers.

Construction of enantiopure carbohydrate conjugates incorporating heterocyclic fragments is often hampered by the necessity of protection/deprotection protocols and multistep routes. Herein, an operationally simple, environmentally benign and effective method for intermolecular radical hydroacylation of unactivated substrates by employing photo-induced hydrogen atom transfer (HAT) initiation is described. The use of commercially available and inexpensive photoinitiators (Ph2CO and NHPI) makes the process attractive. The olefin hydroacylation protocol applies to a wide array of substrates bearing numerous functional groups and many complex structural units. The reaction proves to be scalable (up to 5 g). Herein, the facile photografting coating method was used to develop a novel metal biochelator film by covalent immobilization of citric acid (CA) onto polypropylene (PP) surface. First, PP film underwent a surface modification by UV-grafting of glycidyl methacrylate (GMA) in the presence of benzophenone (BP). The concentrations of GMA (bifunctional monomer) and BP (photoinitiator) were appropriately optimized to achieve the breath-figure-array structure and brush-like grafting configuration with a high availability of the reactive oxirane ring. Subsequently, CA was immobilized onto the PP-g-GMA surface via the ring-opening reaction to obtain the biochelator film (PP-g-GMA-g-CA). Fourier-transform infrared spectroscopy and solid state C-13 nuclear magnetic resonance spectroscopy elucidated the chemical structure and grafting mechanism of grafted samples. X-ray photoelectron spectroscopy was used to detect the chemical composition of the surface of the grafted samples, by which the grafting density was relatively estimated. In addition, the uniformity of the grafting pattern and morphology of the grafting unit were observed using scanning electron microscopy and atomic force microscopy. Furthermore, the carboxylic acid density of the biochelator film (PP-g-GMA-g-CA) was quantified using a toluidine blue O assay, in which approximately 327 +/- 58 nM carboxylic acid was detected per each film. The metal chelating activity of the film was also investigated using the colorimetric ferrozine assay, by which PP-g-GMA-g-CA film chelated approximately 215 +/- 11 nM of Fe3+. Therefore, this newly developed system can be used as a versatile metal biochelator in a wide range of biomedical, cosmetic, and packaging applications.

Chiral polyureas of cinchona alkaloids were synthesized via repetitive Mizoroki-Heck (MH) coupling reaction. The Pd-catalyzed polycondensation of cinchona urea dimers6and aromatic diiodide7afforded the chiral polyureas (P1-P4). The catalytic activity of the chiral polymers was subsequently investigated. The asymmetric Michael addition of ketoesters to nitroolefins was successfully catalyzed by the polymeric organocatalysts (P1-P4) to give the corresponding Michael adducts with high catalytic activities and excellent enantionselectivities (up to >99% ee).
