BackgroundWith the increasing demand for vegetative oil and the approach of peak seed oil production, it is important to develop new oil production platforms from non-seed tissues. Castor bean (Ricinus communis) is one of the crops for vegetable oil for industrial applications with yield around 1.4 ton oil per hectare produced in seed. The castor caruncle is a non-seed tissue attached to seed.ResultsCaruncle accumulates up to 40% oil by weight in the form of triacylglycerol (TAG), with a highly contrasting fatty acid composition when compared to the seed oil. Biochemical analysis indicated that the caruncle synthesizes TAGs independent of the seed. Such non-seed tissue has provided an excellent resource for understanding the mechanism of oil accumulation in tissues other than seeds. Transcriptome analysis revealed the key members of gene families involved in fatty acid synthesis and TAG assembly in the caruncle. A transient expression assay of these selected genes resulted in a 20-fold increased TAG accumulation in leaves.ConclusionsCastor caruncle utilizes an independent system to synthesize TAGs. Results provide the possibility of exploiting caruncle gene set to engineer oil production in non-seed tissues or microbes.

In vivo animal bioassays are increasingly being supplemented with in vitro assays to serve as the new standard for chemical toxicity tests. Despite this shift, investigators face challenges related to increased reliance on in vitro data. The aim of this study was to deploy a streamlined method to assess the ability of in vitro data to predict similar results as in vivo data by correlating chemical toxicity rankings obtained using Benchmark Doses and Benchmark Dose Lower Limits (BMD(L)s) derived from in vivo and in vitro assays. In vitro and in vivo assay characteristics were assessed for their impact on the predictive ability of in vitro data. Minimum best-fit BMD(L)s were calculated for chemicals using Environmental Protection Agency’s (EPA’s) Benchmark Dose Software (BMDS). Forty-one chemicals met the inclusion criteria of this study. Relative chemical toxicity rankings were assessed through Kappa statistics, Pearson correlations, and/or Ordinary Least Squares (OLS) regressions. Results illustrated likely ability of in vitro data to predict similar results as short-term in vivo data. Further, rankings derived from in vitro cytotoxicity assays, unlike stress response assays, significantly correlated with rankings derived from short-term in vivo assays. These results support the use of in vitro data as a prioritization tool within toxicity testing.

Sulfate radical (SO4 center dot(-)) and hydroxyl radical (center dot OH) generated from persulfate or peroxymonosulfate in AOPs have been widely used in contaminant degradation. Anthracene (ANT) can be decomposed by SO4 center dot(-) and center dot OH. The processes of ANT decomposition were investigated using theoretical calculations in this paper. The initiation reactions of ANT, anthrone, anthraquinone (ATQ) and 1-hydroxylanthraquinone (1-hATQ) by two radicals are studied. The highest free energy barriers of initiation reactions are 22.30 kcal mol(-1) in ATQ + SO4 center dot(-) reaction and 6.84 kcal mol(-1) in ATQ + center dot OH reaction. Comparing the rate constants of initiation reaction through the two radicals at 273-373 K, it can be concluded that SO4 center dot(-) and center dot OH both play important roles on the initiation of ANT and anthrone at lower pH. For ATQ and 1-hATQ, center dot OH is more important than SO4 center dot(-) in the initiation process, which indicates that the indirect influence of SO4 center dot(-) are more significant in the degradation processes of ATQ and 1-hATQ. This study provides theoretical confirmations for the mechanisms of reactions of ANT with SO4 center dot(-) and center dot OH, and evaluates the importance of SO4 center dot(-) and center dot OH according to the reaction rates. The work can give more insight into the degradation of PAHs by radicals.

A combination of DIBAL-H and Cp2ZrCl2 is shown to promote the regioselective cleavage of primary acetates on a broad scope of substrates, ranging from carbohydrates to terpene derivatives, with a high tolerance toward protecting groups and numerous functionalities found in natural products and bioactive compounds. A novel method was used to prepare a magnetic phenylene-based periodic mesoporous organosilica nanocomposite with yolk-shell structure (Fe3O4@YSPMO). The Fe3O4@YSPMO nanomaterial was prepared by using easily accessible pluronic-P123 and cetyltrimethylammonium bromide (CTAB) surfactants under basic conditions. This material was employed for effective immobilization of potassium perruthenate to prepare an Fe3O4@YSPMO@Ru nanocatalyst for the aerobic oxidation of alcohols. The physiochemical properties of the designed Fe3O4@YSPMO@Ru nanocomposite were studied using PXRD, FT-IR, TGA, SEM, TEM, ICP, VSM and XPS analyses. Fe3O4@YSPMO@Ru was effectively employed as a highly recoverable nanocatalyst in the selective aerobic oxidation of alcohols.

Ball milling of calcium carbide (CaC2) enables the reaction of its ethynyl moiety with organic electrophiles. The extensive development and use of new anthropogenic chemicals have inevitably led to their presence in aquatic environments. Surface waters affected by sewage effluents have been exposed to these new substances. In the present study, the occurrence of anthropogenic substances, including pharmaceuticals and industrial chemicals, was investigated in one of the major rivers in Korea, the Nakdong River. Furthermore, seasonal variations in their content were determined via annual monitoring. Through the suspect and non-target screening (SNTS) technique, 58 substances were newly identified in the river and integrated in the quantitative monitoring practice. The results revealed that niflumic acid and melamine exhibited the highest median concentrations, i.e., 320 ng/L and 11,000 ng/L, respectively. The results associated with seasonal change revealed that the concentration of a considerable number of substances increased in winter when the flow rate was low. Conversely, some substances exhibited high concentrations in summer (e.g., polyethylene glycol) and spring (e.g., niflumic acid). This was attributed to the seasonal changes in the consumption, prescriptions, or the application of alternative substances. These changes were also reflected by the risk quotient (RQ) values calculated from the concentration and toxicity values. Pharmaceuticals such as telmisartan and carbamazepine and industrial chemicals such as organophosphorus flame retardants (OPFRs) and melamine accounted for approximately 90% of the total RQ. Major substances prioritized using the production of the RQ value and the detection frequency included OPFRs and telmisartan. It is recommended that these results be reflected in future water quality monitoring plans. (C) 2020 Elsevier Ltd. All rights reserved.

A class of zwitterionic organocatalysts based on an amide anion/iminium cation charge pair has been developed. The zwitterions are easily prepared by reacting aziridines with aminopyridines. They are catalytically applicable to transesterifications and dehydrative esterifications. Mechanistic studies reveal that the amide anion and iminium cation work synergistically in activating the reaction partners, with the iminium cationic moiety interacting with the carbonyl substrates through nonclassical hydrogen bonding. The reaction can be applied to large-scale synthesis of biodiesel under mild conditions.
