The reaction of aromatic aldehydes with malononitrile, ethyl or butyl cyanoacetate and acetylacetone in the presence of NaOH under mild conditions (EtOH, 25 degrees C) led to the formation of new series of (1S,5R,6R,9R)/(1R,5S,6S,9S)-2-amino-6,9-diaryl-7-acetyl-8-methyl-4-oxo-5-cyano-3-azabicyclo[3.3.1]nona-2,7-diene-1-carboxylic acids esters. A plausible mechanism of the cascade reaction was proposed.

Severe acute respiratory syndrome coronavirus (SARS-CoV-2) disease is a global rapidly spreading virus showing very high rates of complications and mortality. Till now, there is no effective specific treatment for the disease. Aloe is a rich source of isolated phytoconstituents that have an enormous range of biological activities. Since there are no available experimental techniques to examine these compounds for antiviral activity against SARS-CoV-2, we employed an in silico approach involving molecular docking, dynamics simulation, and binding free energy calculation using SARS-CoV-2 essential proteins as main protease and spike protein to identify lead compounds from Aloe that may help in novel drug discovery. Results retrieved from docking and molecular dynamics simulation suggested a number of promising inhibitors from Aloe. Root mean square deviation (RMSD) and root mean square fluctuation (RMSF) calculations indicated that compounds 132, 134, and 159 were the best scoring compounds against main protease, while compounds 115, 120, and 131 were the best scoring ones against spike glycoprotein. Compounds 120 and 131 were able to achieve significant stability and binding free energies during molecular dynamics simulation. In addition, the highest scoring compounds were investigated for their pharmacokinetic properties and drug-likeness. The Aloe compounds are promising active phytoconstituents for drug development for SARS-CoV-2.

4-Imidazol-1-yl-butane-1-sulfonic acid (ImBu-SO3H) has been successfully synthetized and fully characterized by FT-IR and high-resolution NMR spectroscopy (H-1, C-13). We report here a C-H homoallylation reaction of aromatic ketones with methylenecyclopropanes (MCPs) only using a catalytic amount of Fe(PMe3)(4). A new magnetic nanocatalyst based on the immobilization of tungstic acid onto new design robust, cost-effective, green, and scalable zirconium-L-aspartate amino acid metal-organic framework (MOF)-grafted L-(+)-tartaric acid stabilized magnetite nanoparticles (Fe3O4/tart-NPs) was synthesized by two successive solvo (hydro)-thermal methods. This catalyst was characterized by Fourier-transform infrared (FT-IR), energy-dispersive X-ray spectroscopy (EDS), X-ray diffraction (XRD), field-emission scanning electron microscopy (FESEM), Brunauer-Emmett-Teller (BET), Barrett-Joyner-Halenda (BJH), zeta potential, Thermogravimetry Analysis-Differential Thermal Analysis (TGA-DTA), and vibrating sample magnetometer (VSM) analyses. This catalyst is outstanding to prepare l-(4-phenyl)-2,4-dihydropyrano[2,3-c] pyrazole derivatives in aqueous media due to the open metal sites, high and steady proton conductivity in the zirconium-MOF, MIP-202(Zr), and MOF and also due to Bronsted acid properties of tungstic acid. This acidic catalyst can easily be extracted by an outward magnetic field after completion of the reaction without any deactivation or selectivity loss. The products were characterized by spectroscopic analysis (FT-IR, H-1-NMR, and C-13-NMR). The optimized reaction conditions and a possible reaction mechanism are outlined.

The oxidation of a series of alkylaromatic compounds with H2O2 catalyzed by an imine-based non-heme iron complex prepared in situ by reaction of 2-picolylaldehyde, 2-picolylamine, and Fe(OTf)(2) in a 2 : 2 : 1 ratio leads to a marked chemoselectivity for aromatic ring hydroxylation over side-chain oxidation. This selectivity is herein investigated in detail. Side-chain/ring oxygenated product ratio was found to increase upon decreasing the bond dissociation energy (BDE) of the benzylic C-H bond in line with expectation. Evidence for competitive reactions leading either to aromatic hydroxylation via electrophilic aromatic substitution or side-chain oxidation via benzylic hydrogen atom abstraction, promoted by a metal-based oxidant, has been provided by kinetic isotope effect analysis.

A novel series of bis Indy-NHC gold complexes have been developed and investigated via a mild Agcarbene transfer route. The obtained complexes were characterized by NMR spectroscopy and X-ray diffraction analysis. The catalytic property of these gold complexes was further evaluated in the oxidation of benzylic. Solvent free synthesis of dihydropyrano[3,2-b]chromenediones was formulated via multicomponent reactions of kojic acid, dimedone and several substituted aromatic aldehydes catalyzed with BCl3. Reactions progressed efficiently and the corresponding heterocyclic products were obtained in good to high yields within short period. The developed protocol is one of the better and efficient alternative methods for the synthesis of dihydropyrano[3,2-b]chromenediones. The simple reaction procedure, easy separation of products, radially available catalyst are certain benefits of this reported protocol.

We report the synthesis of beta-lactams from alpha-ketoacylsilanes and imines, which proceeds via a formal [2+2] photochemical cycloaddition with in situ generation of siloxyketene. This mild and operationally simple reaction proceeds in an atom-economic fashion with broad substrate scope, including aldimines, ketimines, hydrazones, and fused nitrogen heterocycles, affording a variety of important beta-lactams with satisfactory diastereoselectivities in most cases. This reaction also features good functional-group tolerance, facile scalability and product diversification. Experimental and computational studies suggest that alpha-ketoacylsilanes can serve as photochemical precursors by engaging in a 1,3 silicon shift to the distal carbonyl group.
