Unlike the previously studied N-trifyl substituted 1,3,5-triheterocyclohexanes, the preferred conformers of compound 1 and of 1,4-diheterocyclohexanes 2-4 are those with the CF3 group directed outward from the ring, which is caused by intramolecular interactions of the oxygen atoms of the CF3SO2N groups with the equatorial hydrogens in the a-position. B3LYP/6-311+G(d,p) calculations of the energy, geometry and NMR parameters corroborate the experimental data. The calculated Perlin effects for all conformers of compounds 1-4 as well as those measured for the major conformers of compounds 3 and 4 were analyzed by the use of the NBO analysis. Synthesis and metal complexes of a chelating ligand containing thiomorpholine and pyridine: 2,6-bis(thiomorpholinomethyl)pyridine

A chelating ligand, 2,6-bis(thiomorpholinomethyl)pyridine (L), was synthesised. Its palladium(II), copper(II) and nickel(II) complexes of the formula ML(NO3)(2), (M = Pd, Cu and Ni), were prepared and characterised. The molecular structures of the three complexes were determined by X-ray diffraction. In all the compounds prepared the ligand acts in a tridentate fashion using its three N atoms while the two S atoms remain free. The Pd complex possesses a distorted square planar coordination geometry with one of the two nitrate groups coordinating as a unidentate ligand and the other ionic. The Pd-N(pyridine) bond length is 1.857(4) Angstrom, which is believed to be the shortest Pd-N separation ever observed. In the Cu complex, the copper atom is five-coordinated in a distorted square planar arrangement with the two nitrate groups acting as unidentate ligands and occupying the apex and one equatorial position. Eight mononuclear [Ni(sal 4-Phtsc)center dot D] thiosemicarbazonato complexes [sal4-Phtsc = salicylaldehyde 4-phenylthiosemicarbazonato ligand; D = imidazole (1), methylimidazole (2), pyridine (3), 4-aminopyridine (4), 4-methylpyridine (6), morpholine (7), thiomorpholine (8), 2-aminophenol (9)] and one dinuclear {[Ni(sal 4-Phtsc)](2)center dot D} center dot 2DMSO [D = 4,4′-bipyridine (5)] complex have been prepared by adding the corresponding Lewis base to the methanol suspension of the parent complex [Ni(sal 4-Phtsc)(H(2)sal4-Phtsc)]center dot CH3OH. The exchange of the neutral salicylaldehyde 4-phenylthiosemicarbazone (H(2)sal4-Phtsc) ligand in the parent complex for the appropriate Lewis base has been confirmed by IR spectroscopy and powder X-ray diffraction (PXRD) in the solid state. The single-crystal X-ray diffraction of seven complexes 1 and 3-8 confirmed the formation of the complexes with the Ni-II ion, coordinated through O,N,S-donor atoms from the dibasic salicylaldehyde 4-phenylthiosemicarbazonato ligand and endocyclic N-donor atom from the neutral ligand D in the form of a distorted square-planar coordination. NMR spectroscopy in DMF or DMSO and quantum mechanical calculations have been performed in order to explain and compare the stability of the complexes in solution, depending on the polarity of solvents in the context of donor properties and the nucleophilicity of the heterocyclic Lewis base. The single-crystal X-ray data enables a comparison with calculated standard Gibbs energies of binding in the context of crystal packing forces, leading to a general conclusion that the stability of the mononuclear complexes results in the formation of more stable hydrogen-bonded cyclic dimers as a crystal packing pattern.

Copper(I) (Pseudo)Halide Complexes with Neocuproine and Aminomethylphosphines Derived from Morpholine and Thiomorpholine – In Vitro Cytotoxic and Antimicrobial Activity and the Interactions with DNA and Serum Albumins

Herein, a series of CuI or CuNCS complexes with neocuproine (2,9-dimethyl-1,10-phenanthroline: dmp) and two tris(aminomethyl)phosphines derived from morpholine (P(CH2N(CH2CH2)(2)O)(3)) or thiomorpholine (P(CH2N(CH2CH2)(2)S)(3)) were tested as cytotoxic agents in vitro towards mouse colon carcinoma (CT26) and human lung adenocarcinoma (A549). The studies showed that the complexes exhibit potential antitumor properties, displayed by IC50 values below 10m towards the tested cell lines, in the case of 4-h incubation time with the examined compounds. Moreover, a high antimicrobial activity of all the complexes was observed against Staphylococcus aureus and Candida albicans with minimal inhibitory concentrations equal to 1-2g/mL. To gain insight into the molecular mechanism of biological activity of the complexes, we investigated also their interactions with plasmid DNA (pUC18) and the human and bovine serum albumins. Gel electrophoresis experiments demonstrated that all the compounds were comparably efficient in DNA degradation process; however, luminescence quenching showed surprising dependence on the interactions strength of the used compounds with the albumins. Apart from exceptionally effective [CuI(dmp)P(CH2N(CH2CH2)(2)O)(3)], the complexes with P(CH2N(CH2CH2)(2)O)(3) quenched more strongly luminescence of bovine serum albumin, while the complexes with P(CH2N(CH2CH2)(2)S)(3) were more active in the quenching of human serum albumin luminescence.

Amides of non-steroidal anti-inflammatory drugs with thiomorpholine can yield hypolipidemic agents with improved anti-inflammatory activity

Novel amides of non steroidal anti-inflammatory drugs (NSAIDs), alpha-lipoic acid and indole-3-acetic acid with thiomorpholine were synthesised by a simple method and at high yields (60-92%). All the NSAID derivatives highly decreased lipidemic indices in the plasma of Triton treated hyperlipidemic rats. The most potent compound was the indomethacin derivative, which decreased total cholesterol, triglycerides and LDL cholesterol by 73%, 80% and 83%, respectively. They reduced acute inflammation equally or more than most parent acids. Hence, it could be concluded that amides of common NSAIDs with thiomorpholine acquire considerable hypolipidemic potency, while they preserve or augment their anti-inflammatory activity, thus addressing significant risk factors for atherogenesis.
