In this study, we describe a novel class of dual c-Kit/Aur inhibitors, characterized by a 1,2,4-triazole core and developed by a structure-based optimization of a previously developed hit, and report the evidence of their significance as drug candidates for the treatment of melanoma. Compound 6a, merging the best inhibitory profile against the target kinases, showed anti-proliferative efficacy against the human melanoma cell lines A2058, expressing the BRAFV600D mutation, and WM266-4, expressing BRAF V600E. Significantly, it displayed also a highly synergistic profile when tested in combination with vemurafenib, thus proving its efficacy not only per se but even in a combination therapy, which is nowadays acknowledged as the cornerstone approach of the forthcoming tumour management.

Exploring new material as adsorbent for the efficient enrichment of pollutants is always of great significance in analytical chemistry. In this work, a magnetic azobenzene framework (labeled as MAzo) was constructed as a magnetic solid phase extraction (MSPE) adsorbent by a simple and environmentally benign strategy. The MAzo exhibited the attractive features of strong magnetism, outstanding adsorption performance, as well as good reusability. Combining MAzo-based MSPE with high performance liquid phase chromatography, a simple and effective method was developed for simultaneous determination of four phenylurea herbicides in pear juice and apple juice samples. Under optimized experimental conditions, the method offered low limits of detection of 0.05-0.15 ng mL(-1), high recoveries of 86.7-109.2% with RSD less than 7%. Density functional theory calculation indicated that the good adsorption performance of MAzo for PUHs can be ascribed to the strong H-bonding forces and weak pi-pi interactions. The facile, green, low-cost synthesis method together with the excellent adsorption performance endows the MAzo great application prospect in sample preparation. (c) 2020 Elsevier B.V. A facile cascade reaction is reported via aza-Michael addition, ring opening, and cyclization between 3-bromoflavone and aniline derivatives or N -phenylurea in the presence of KO t -Bu and CuI in DMF under mild reaction conditions. Products were obtained as stereospecific trans -aminated aurones in good to excellent yields (61-83%). Our protocol is operationally successful with ease, avoids the requirement of additives and ligands, and offers broad substrate scope.

A rhodium-catalyzed synthesis of isoquinolino[1,2-b]quinazolines via C-H annulation using vinylene carbonate as an oxidizing acetylene surrogate in biomass-derived gamma-valerolactone (GVL) has been developed. Study Objective: To investigate whether the rate of increase in the performance of abdominal myomectomy over a laparoscopic approach after the US Food and Drug Administration (FDA) safety communication regarding morcellator use for myomectomy differs among races. Design: Retrospective cohort study. Setting: The American College of Surgeons National Surgical Quality Improvement data. Patients: Patients aged 18 to 55 years who underwent either laparoscopic or abdominal myomectomy, excluding malignant cases, emergency cases, operations performed by nongynecologic specialists, and cases in which myomectomy was performed during cesarean section. Interventions: None. Measurements and Main Results: The odds ratios of abdominal myomectomy over laparoscopic myomectomy before and after the release of the FDA communication were calculated in 3 race categories: white, African American, and other races. In a logistic regression analysis adjusted for possible confounders, including all races, the odds ratio of abdominal myomectomy before and after the FDA communication was 1.30 (95% confidence interval [CI], 1.20-1.41; p <. 001). In a logistic regression analysis with a product term of FDA communication exposure and race as a possible effect modifier, the African American population showed a significantly greater change in the odds of abdominal myomectomy over laparoscopic myomectomy in comparison with the white population (1.22; 95% CI, 1.02-1.47; p =.03). In contrast, other races showed no significant change (.83; 95% CI,.64-1.08; p =.17). Conclusion: After the FDA communication, the odds ratio of abdominal myomectomy was disproportionately increased in the African American population. Inhibition of the epidermal growth factor receptor (EGFR) has been proved to be one of the most promising strategies for the treatment of non-small cell lung cancers. A series of 2-aryl-4-amino substituted quinazoline derivatives were designed and synthesized with the purpose to overcome L858R/T790M/C797S (CTL) triple mutant drug resistance and the biological activity for inhibition of CTL kinases and EGFR wild type (WT) were evaluated. Three compounds (20, 24 and 27) showed excellent inhibitory activities against EGFR kinases triple mutant CTL (IC50 < 1 mu M) and high selectivity (IC50: WT/CTL >10000). Cell line evaluation showed that the most potent compound 27 was significantly potent against H1975-EGFR L858R/T790M (IC50 = 3.3 mu M) and H1975-EGFR L858R/T790M/C797S (IC50 = 1.2 mu M). Compound 27 also exhibited good microsomes stabilities in human, rat and mouse liver species, but low bioavailability. This work would be very useful for discovering new quinazoline derivatives as tyrosine kinase inhibitors targeting triple mutant L858R/T790M/C797S.

Catalyst-free photoinduced processes in aqueous medium represent significant advancement toward development of green and sustainable pathways in organic synthesis. tert-Butyl hydroperoxide (TBHP) is a widely used oxidant in organic reactions, where the decomposition of TBHP into its radicals by metal catalysts or other reagents is a key factor for efficient catalytic outcome. Herein, we report a simple and environmentally friendly visible light-promoted synthetic pathway for the synthesis of N-heterocyclic moieties, such as quinazolinones and quinoxalines, in the presence of TBHP as an oxidizing agent in aqueous medium that requires no catalysts/photocatalysts. The enhanced rate of decomposition to generate free radicals from TBHP upon visible light irradiation is the driving force for the domino reaction.

We have developed a new method for the synthesis of 2,3-dihydroquinazolin-4(1H)-ones in good to excellent yields via phosphomolybdic acid (PMA)-catalyzed cyclocondensation of 2-aminobenzamides with aldehydes or ketones in N,N-dimethylformamide (DMF) at room temperature. The present method proves to be efficient in terms of short reaction time, high yield, simple workup and easy purification.

A total of 22 quinazoline thioether derivatives incorporating a 1,2,4-triazolo[4,3-a]pyridine moiety were designed, synthesized, and evaluated as antimicrobial agents in agriculture. Among these compounds, the chemical structure of compound 61 was further confirmed via single-crystal X-ray diffraction analysis. The bioassay results revealed that some of the compounds possessed noticeable in vitro antibacterial activities against the tested phytopathogenic bacteria. For example, compounds 6b and 6g had EC50 values as low as 10.0 and 24.7 mu g/mL against Xanthomonas axonopodis pv. citri (Xac), respectively, which were significantly better than that of the commercial agrobactericide bismerthiazol (56.9 mu g/mL). Particularly, compound 6b was also found to be capable of suppressing the pathogenic bacterium Xanthomonas oryzae pv. oryzae (Xoo) approximately 12-fold more potent than control bismerthiazol, in terms of their EC 50 values (7.2 versus 89.8 mu g/mL). Importantly, the most active compound 6b turned out to be one with the highest hydrophilicity and the lowest molecular weight within the series. In vivo bioassays further showed the application prospect of 6b as a promising plant bactericide for controlling Xoo. Additionally, in vitro antifungal activities of these compounds were also evaluated at the concentration of 50 mu g/mL. Overall, the present study demonstrated the potential of 1,2,4-triazolo[4,3-a]pyridine-bearing quinazoline thioether derivatives as efficient agricultural antibacterial agents for crop protection.

A novel aggregation induced emission (AIE) active anthracene based dihydroquinazolinone derivative (probe 1) has been synthesized and characterized by means of spectroscopic methods. The photophysical properties of this probe have been investigated in solvents of different polarity display that fluorescence states are of intramolecular charge transfer (ICT) character. Probe 1 show clear AIE behavior in water/THF mixture on reaching water fraction 95%. The AIE behavior of probe 1 have been exploited for the detection of metal ions in aqueous solution which reveals high selectivity and sensitivity towards Cu2+ ions by colorimetrically and function as a chemosensor in a remarkable turn-off fluorescence manner. Further, the experimental results were investigated by computational means by optimizing the ground state geometries of probe 1 and probe 1-Cu complex using density functional theory (DFT) at B3LYP/6-31G** and B3LYP/6-31G**(LANL2DZ) levels of theory. Intra-molecular charge transfer was observed in probe 1 while ligand to metal charge transfer (LMCT) for probe 1-Cu complex.
