Novel series of some triazolo [4,3-b]pyridazine derivatives were designed and synthesized. All the newly synthesized compounds were evaluated for their cytotoxic activity at 10(-5) M concentration towards 60 cancer cell lines according to USA NCI protocol. Most of the synthesized compounds showed good activity against SR (leukemia) cell panel. The most active compounds, 2f and 4a were subjected for further evaluation at a five dose level screening and their efficacy for c-Met kinase inhibition was determined in vitro. Binding mode of these derivatives was explored via molecular docking.

In this study, a new fluorescence probe HMAQ based on quinazoline and diaminomaleonitrile was constructed for sensing ClO- and Al3+. A fluorescence blue-shift with 102 nm together with a color change from golden-yellow to colorless was found by hypochlorite-induced hydrolysis of -CH = N- group to release the initial fluorophore. Besides, Al3+ could cause a 72-nm blue-shifted emission spectra and a color change from golden-yellow to brown. As expected, HMAQ exhibited a satisfactory selectivity and sensitivity to ClO-/Al3+ with a quick response. Most notably, the reversibility of the [HMAQ+Al3+] complex could be used to detect ClO- and Al3+ simultaneously without mutual interferences. The detection limits of HMAQ for ClO- and Al3+ were turned out to be 102 nM and 1.56 nM, respectively. This paper presents a comparative experimental study on the frosting characteristics of an air source heat pump system that contains two operation modes, i.e., the vapor-bypassed evaporation (VB) mode and the conventional operation (CO) mode. For VB mode, a phase separator is installed inside the evaporator re-frigerant circuit to bypass vapor refrigerant, which can increase the average evaporation temperature for retarding frosting by reducing refrigerant pressure drop and decreasing the area of refrigerant inferior heat transfer coefficient region (mist flow region). According to the experimental results, under H2 con-dition (indoor: 20/15 degrees C; outdoor: 2/1 degrees C), as the initial refrigerant mass flow rate ranges 35-45 g / s , the VB mode yields 4.30-7.06% lower frost mass, 3.9-7.3% higher average heating capacity and 6.4-10.8% higher coefficient of performance (COP) over CO mode with the frosting duration of 60 min. Moreover, under H3 condition (indoor: 20/15 degrees C; outdoor:-7/-8 degrees C), the frosting duration is set as 100 min, and the VB mode obtains 9.67% lower frost mass, 7.0% higher average heating capacity and 13.3% higher COP over CO mode. Generally, the vapor-bypassed evaporation technique has a great application potential in retarding frosting and improving performance of an air source heat pump system especially under high heating capacity or low ambient temperature conditions. The NAD(+)-dependent deacetylase SIRT2 represents an attractive target for drug development. Here, we designed and synthesized drug-like SIRT2-selective inhibitors based on an analysis of the putative binding modes of recently reported SIRT2-selective inhibitors and evaluated their SIRT2-inhibitory activity. This led us to develop a more drug-like diketopiperazine structure as a hydrogen bond (H-bond) hunter to target the substrate-binding site of SIRT2. Thioamide 53, a conjugate of diketopiperazine and 2-anilinobenzamide which is expected to occupy the selectivity pocket of SIRT2, exhibited potent SIRT2-selective inhibition. Inhibition of SIRT2 by 53 was mediated by the formation of a 53-ADP-ribose conjugate, suggesting that 53 is a mechanism-based inhibitor targeting the selectivity pocket, substrate-binding site, and NAD(+)-binding site. Furthermore, 53 showed potent antiproliferative activity toward breast cancer cells and promoted neurite outgrowth of Neuro-2a cells. These findings should pave the way for the discovery of novel therapeutic agents for cancer and neurological disorders.

Thioamides could be conveniently synthesized in good to excellent yields via DMSO-promoted oxidative coupling of methylhetarenes with amines in the presence of a near stoichiometric amount of sulfur (1.25 equiv.). Production of nanocomposites from macro-scale materials in situ in a melting compounder is highly desirable yet challenging. In this study, we develop a highly efficient approach for the preparation of cellulose nanofiber (CNF)-reinforced high-density polyethylene (HDPE) composite materials from as-received wood pulp fibers, in which nanofibrillation-assisting plasticizers (urea and urea derivatives) and a diblock copolymer dispersant are used. The most effective plasticizer is urea, which plays a crucial role in producing CNF with less fragmentation during the kneading step, owing to the plasticization/nanofibrillation and the reaction with hydroxyl groups of cellulose fibers into a carbamate. The diblock copolymer as a dispersant enables the stabilization of the appropriate dispersion of the produced CNF in nonpolar HDPE. The resulting composites exhibit significantly improved mechanical properties, including a 6.9-fold increase in the Youngs modulus with 10 wt % loading of wood pulp fibers over that of neat HDPE.

The present experiment was conducted to determine whether, during periods of negative energy balance, the increase in glucose availability, despite similar DMI and greater milk production, induced by a combined supplement of folic acid and vitamin B-12 was related to effects of insulin on metabolism. Sixteen multiparous Holstein cows averaging 45 days in milk (standard deviation: 3) were assigned to 8 blocks of 2 animals each according to their milk production (45 kg/d; standard deviation: 6) during the week preceding the beginning of the experiment. Within each block, they received weekly intramuscular injections of either saline (CON) or folic acid and vitamin B-12 (VIT) during 5 consecutive weeks. During the last week, the cows were fed 75% of their ad libitum intake during 4 d. Blood samples were taken the morning before starting the feed restriction and on the third day of feed restriction. On the fourth day of feed restriction, the daily meal was not served and an intravenous glucose tolerance test was performed. During the 4 wk preceding the feed restriction, milk production and DMI were not affected by treatments. During the feed restriction, the vitamin supplement tended to decrease milk fat concentration and increase milk concentration of lactose. Plasma concentrations of homocysteine, Ile, Leu, Val, and branched-chain AA increased in VIT cows during the restriction but not in CON cows. During the glucose tolerance test, insulin peak height was lower and insulin incremental positive area under the curve tended to be lower for VIT than for CON [83 (95% confidence interval, CI: 64-108) vs. 123 (95% CI: 84-180) mu g.180 min/L, respectively]. Free fatty acid nadir was reached earlier for VIT than for CON [34 (95% CI: 26-43) vs. 46 (95% CI: 31-57) min, respectively]. Glucose area under the curve, clearance rate and peak height, insulin time to reach the peak and clearance rate, and free fatty acid nadir did not differ between VIT and CON. The reduction in insulin release during a glucose tolerance test without changes in glucose clearance rate or area under the curve suggests that the vitamin supplement improved insulin sensitivity in feed-restricted lactating dairy cows.

A series of 4,5-diphenyl-7H-thieno[2 ‘,3 ‘:3,4]pyrido[2,1-b]quinazolin-7-ones was synthesized via the Rh(iii)-catalyzed annulation of 2-thienylquinazolin-4(3H)-ones with diphenylacetylene. 2-Phenylquinazolin-4(3H)-one amide alcoholysis and double C-H functionalization proceeded under the same reaction conditions with the formation of 2,3,7,8-tetraphenyl-1H-benzo[d,e][1,8]-naphthyridine derivatives. All compounds possess fluorescence properties in MeCN and toluene solutions as well as in the solid state.
