This study describes synthesis and evaluation of novel 5-Chloro-2-thiophenyl-1,2,3-triazolylmethyldihydroquinolines 7a-o as dual inhibitors of Mycobacterium tuberculosis and influenza virus. Huisgen’s [ 3+ 2] dipolar cycloaddition of 6-(azidomethyl)-5-chloro-2-(thiophen-2-yl)-7,8-dihydroquinoline 5 with various alkynes 6a-o using sodium ascorbate and copper sulphate gave new dihydroquinoline-1,2,3-triazoles 7a-o in good to excellent yields. The new compounds were evaluated for in vitro antimycobacterial against M. tuberculosis H37Rv (Mtb) and antiviral activity against influenza virus A/Puerto Rico/8/34 (H1N1). Among the fifteen new analogs, compounds 7a (MIC: 3.12 mu g/mL), 7j and 7k (MIC: 6.25 mu g/mL) were identified as potent antitubercular agents. The virus-inhibiting activity of all the fifteen compounds was found to be moderate, and among them the compound 7l, bearing thiophene moiety appeared the most active with good selectivity index (IC50 = 19.5 mu g/mL; SI = 15). The results presented here will help developing newer dual inhibitors of tuberculosis and influenza virus.

We report a universal straightforward strategy for the chemical synthesis of modified oligoribonucleotides containing functional groups of different structures at the 2 ‘ position of ribose. Methyl glycolate (MG) was synthesized as a precursor to ethylene glycol from the catalytic carbonylation of formaldehyde followed by esterification with methanol by using metal-free, efficient and recyclable SO3H-functionalized ionic liquids (BAILs) as catalysts. At 160 degrees C, 5.0 MPa, and reactant mole ratio of BAIL: H2CO:H2O = 1:40: 80, 98 % conversion of formaldehyde was achieved with 94 % selectivities of MG. Catalysts did not show any significant deterioration in performance in repeated use up to eight batches.

The new compounds were evaluated for their in vitro antifungal and antibacterial activities. The screening revealed compounds with specific activity against pathogenic fungi, including Candida albicans, Candida auris, and Cryptococcus. Compound 3d, which incorporated N-phenylpiperazine moiety, exhibited the highest growth inhibition against C. albicans with a minimum inhibitory concentration of 16 mu g/mL. The compounds were superior to fluconazole in inhibiting Candida biofilm mass at sub-inhibitory concentration. Furthermore, the MTS assay confirmed that compounds 1 and 3d exhibited an excellent toxicity profile (not toxic, up to 256 mu g/mL, for mammalian cells). Collectively, the presented results demonstrate that the synthesized pyrazolo-triazines warrant further exploration for potential use as antifungal agents.

Polymerization-induced self-assembly (PISA) represents a powerful technique for the preparation of nanostructures comprising various morphologies. Herein, we demonstrate that the recently introduced monomerN-acryloylthiomorpholine (NAT) features a unique self-assembly behaviour during an aqueous PISA. The one-pot, aqueous RAFT dispersion polymerization starting from short poly(N-acryloylmorpholine) (PNAM) enables access to all common solution morphologies including spheres, worms, vesicles and lamellae, at very low molar masses (< 8 kDa). Moreover, all these structures can be obtained for the same polymer composition and size by the variation of the polymerization temperature and concentration of the monomer. This exceptional self-assembly behavior is associated with the combination of a high glass transition temperature, excellent water solubility of the monomer, and the early onset of aggregation during the polymerization, which stabilizes the morphology at different stages. This PISA system opens up new opportunities to reproducibly create versatile, functional nanostructures and enables an independent evaluation of morphology-property relationships, as it is exemplarily shown for the oxidative degradation of spherical and wormlike micelles, as well as vesicles.
