The effective control of biointerfacial interactions is of outstanding interest in a broad range of biomedical applications, ranging from cell culture tools to biosensors and implantable medical devices. For many of these applications, highly specific interactions between cells and material surfaces are desired. Sophisticated control over these interactions requires reducing or preventing non-specific interactions on the one hand and displaying highly specific signals that can be recognized by extracellular receptors on the other. We have recently developed ultra-low fouling coatings that can be applied in a single step using photoreactive copolymers of 2-hydroxypropyl acrylamide and N-benzophenone acrylamide. Here, we have expanded this approach by incorporating polymerizable peptide monomers into these copolymers. The monomers QQGWFGAGK(acrylamide) and acrylamide-GAGQQGWF were synthesized after identifying the QQGWF sequence as a binding motif for CD44 by phage display for the first time. Our results demonstrate that UV-crosslinked coatings fabricated using the QQGWFGAGK(acrylamide) monomer are effective at selectively binding hMSC in the presence of HepG2 and HEK293 cells due to the difference in CD44 expression. Our results also demonstrate that the peptide modified coatings retain their low biofouling character using a BCA protein binding assay as well as an E. coli bacterial attachment assay over a 24 h period. Our approach provides an alternative to traditional integrin-mediated selective cell binding on surfaces and opens the door to new diagnostic applications, exploiting the fact that the transmembrane protein CD44 is highly expressed in multiple diseases.

We investigated a base-promoted protocol for the intermolecular anti-Markovnikov hydroamidation of vinyl arenes with arylamides to furnish the arylethylbenzamides with excellent chemo- and regioselectivity. The reaction tolerates an extensive variety of functional groups and has been successfully extended with electronically varied handles, aminobenzamides, electron-rich/electron-deficient heterocyclic amides, and vinyl arenes to afford the hydroamidated products. Excellent chemoselectivity was observed for the amide group over amine. The proposed mechanism and vital role of the solvent was well supported by deuterium labeling studies and control experiments.

Examined here are the structures of complexes of benzophenone microsolvated with up to three water molecules by using broadband rotational spectroscopy and the cold conditions of a molecular jet. The analysis shows that the water molecules dock sideways on benzophenone for the water monomer and dimer moieties, and they move above one of the aromatic rings when the water cluster grows to the trimer. The rotational spectra shows that the water trimer moiety in the complex adopts an open-loop arrangement. Ab initio calculations face a dilemma of identifying the global minimum between the open loop and the closed loop, which is only solved when zero-point vibrational energy correction is applied. An OH…pi bond and a Burgi-Dunitz interaction between benzophenone and the water trimer are present in the cluster. This work shows the subtle balance between water-water and water-solute interactions when the solute molecule offers several different anchor sites for water molecules.

A convenient and eco-friendly, one-pot synthetic protocol for pharmaceutically important pyran-based heterocyclic compounds is repeated herein. The reactions were carried out at room temperature in a water-ethanol solvent mixture under direct irradiation of UV365 light source in the absence of photocatalyst. The merits of the present protocol include mild and catalyst-free reactions, green procedures, and high yields. The added advantage of this protocol is that it can be applied for large-scale synthesis of various pyran-based heterocyclic compounds without employing any expensive catalyst. Also, in vitro antibacterial screening of pyran derivative was investigated against both Gram-positive Bacillus cereus (B. cereus) and Gram-negative bacteria Escherichia coli (E. coli).

Three 2D coordination polymers, [Cu-2(mu(4)-dpa)(bipy)(2)(H2O)](n)center dot 6nH(2)O (1), [Mn-2(mu(6)-dpa)(bipy)(2)](n) (2), and [Zn-2(mu(4)-dpa)(bipy)(2)(H2O)(2)](n)center dot 2nH(2)O (3), were prepared by a hydrothermal method using metal(II) chloride salts, 3-(2 ‘,4 ‘-dicarboxylphenoxy)phthalic acid (H(4)dpa) as a linker, as well as 2,2 ‘-bipyridine (bipy) as a crystallization mediator. Thermal stability and catalytic behavior of 1-3 were also studied. In particular, zinc(II) coordination polymer 3 functions as a highly active and recoverable heterogeneous catalyst in the mild cyanosilylation of benzaldehydes with trimethylsilyl cyanide to give cyanohydrin derivatives. The influence of various parameters was investigated, including a time of reaction, a loading of catalyst and its recycling, an effect of solvent type, and a substrate scope. As a result, up to 93% product yields were attained in a catalyst recoverable and reusable system when exploring 4-nitrobenzaldehyde as a model substrate. This study contributes to widening the types of multifunctional polycarboxylic acid linkers for the design of novel coordination polymers with notable applications in heterogeneous catalysis.

Chiral hyperbranched polymers (HBP) containing cinchona alkaloids were synthesized using a Mizoroki-Heck (MH) coupling polymerization reaction between a cinchona squaramide dimer and tri- or tetrasubstituted aromatic iodides. This is a new type of polymeric chiral organocatalyst. We found that the as-obtained chiral HBPs show excellent catalytic activity in the asymmetric Michael reaction. Almost perfect enantioselectivity (>99% ee) was achieved in the reaction of beta-ketoester and trans-beta-nitrostyrene. The (R)- and (S)-enantiomers of the chiral herbicide napropamide (NAP) show different biological activities and ecotoxicities. These two enantiomers behave differently in the environment due to enantioselective catabolism by microorganisms. However, the molecular mechanisms underlying this enantioselective catabolism remain largely unknown. In this study, the genes (snaH and snpd) involved in the catabolism of NAP were cloned from Sphingobium sp. B2, which was capable of catabolizing both NAP enantiomers. Compared with (R)-NAP, (S)-NAP was much more rapidly transformed by the amidase SnaH, which initially cleaved the amide bonds of (S)/(R)-NAP to form (S)/(R)-2-(1-naphthalenyloxy)-propanoic acid [(S)/(R)-NP] and diethylamine. The alpha-ketoglutarate-dependent dioxygenase Snpd, showing strict stereoselectivity for (S)-NP, further transformed (S)-NP to 1-naphthol and pyruvate. Molecular docking and site-directed mutagenesis analyses revealed that when the (S)-enantiomers of NAP and NP occupied the active sites, the distance between the ligand molecule and the coordination atom was shorter than that when the (R)-enantiomers occupied the active sites, which facilitated formation of the transition state complex. This study enhances our understanding of the preferential catabolism of the (S)-enantiomer of NAP on the molecular level.

An efficient four-component strategy has been developed for the synthesis of a novel series of (E)-N-benzylidene-3-(benzylthio)-5-(3,5-dimethyl-1H-pyrazol-1-yl)-4H-1,2,4-triazol-4-amines in good yields. The method is effective for the synthesis of benzylidene aralkyl/allyl/propargyl thio pyrazolyl-1,2,4-triazol-4-amines in a one-pot operation. This method is operationally simple, metal-free, and works under reflux conditions with diverse substrates. Among all the synthesized compounds 5a, b, c, h, j , and n were shown good results in silico studies. 5n is showing good results in both DNA binding and molecular docking studies. Organic molecules bearing acetoxy moieties are important functionalities in natural products, drugs, and agricultural chemicals. Synthesis of such molecules via transition metal-catalyzed C-O bond formation can be achieved in the presence of a carefully chosen directing group to alleviate the challenges associated with regioselectivity. An alternative approach is to use ubiquitous carboxylic acids as starting materials and perform a decarboxylative coupling. Herein, we report conditions for a photocatalytic decarboxylative C-O bond formation reaction that provides rapid and facile access to the corresponding acetoxylated products. Mechanistic investigations suggest that the reaction operates via oxidation of the carboxylate followed by rapid decarboxylation and oxidation by Cu(OAc)(2).
