Antihypertensive Properties of a Novel Morphologic Derivative (4-tert-buthyl-2,6-bis(thiomorpholine-4-ilmethyl)phenol)

We evaluated the antihypertensive properties of 4-tert-buthyl-2,6-bis(thiomorpholine-4-ilmethyl)phenol (TBTIF). Spontaneously hypertensive rats were treated with TBTIF or captopril (both at 1 mg center dot kg(-1)center dot d(-1) intramuscularly for 4 days), and their blood pressure (BP) was assessed. In some experiments, concentration response curves to angiotensin I or angiotensin II were generated in rat aortic rings and in the absence or presence of Ang-(1-7), N-G-monomethyl l-arginine, or both; additionally, the angiotensin-converting enzyme (ACE) and ACE2 mRNA levels were quantified in the aortic rings using reverse transcription-polymerase chain reaction. TBTIF diminished BP and reduced angiotensin I- or angiotensin II-induced vasoconstriction. The presence of Ang-(1-7) induced a greater reduction in vasoconstriction, and this effect was reversed by l-N-G-monomethyl arginine. Moreover, TBTIF decreased the mRNA of ACE and increased the mRNA of ACE2. In conclusion, TBTIF diminished rat BP through nitric oxide-dependent and nitric oxide-independent mechanisms. In contrast to captopril, TBTIF exhibits better antihypertensive properties through mechanisms that involve ACE2.

Common degradative pathways of morpholine, thiomorpholine, and piperidine by Mycobacterium aurum MO1: Evidence from H-1-nuclear magnetic resonance and ionspray mass spectrometry performed directly on the incubation medium

In order to see if the biodegradative pathways for morpholine and thiomorpholine during degradation by Mycobacterium aurum MO1 could be generalized to other heterocyclic compounds, the degradation of piperidine by this strain was investigated by performing H-1-nuclear magnetic rc:resonance directly with the incubation medium. Ionspray mass spectrometry, performed without purification of the samples, was also used to confirm the structure of some metabolites during morpholine and thiomorpholine degradation. The results obtained with these tyro techniques suggested a general pathway for degradation of nitrogen heterocyclic compounds by M. aurum MO1. The first step of the degradative pathway is cleavage of the C-N bond; this leads formation of an intermediary amino acid, which is followed by deamination and oxidation of this amino acid into a diacid. Except in the case of thiodiglycolate obtained from thiomorpholine degradation, the dicarboxylates are completely mineralized by the bacterial cells. A comparison with previously published data showed that this pathway could be a general pathway for degradation by other strains of members of the genus Mycobacterium.

Investigations of primary and secondary amine carbamate stability by H-1 NMR spectroscopy for post combustion capture of carbon dioxide

Carbamate formation is one of the major chemical reactions that can occur in solution in the capture of CO2 by amine-based solvents, and carbamate formation makes a significant enthalpy contribution to the absorption-desorption of CO2 that occurs in the absorber/stripper columns of the PCC process. Consequently, the formation of carbamates of selected series of primary and secondary amines over the temperature range (288 to 318) K has been investigated by equilibrium H-1 NMR studies, and the stability constants (K-9) for the equilibrium: RNH2 + HCO3- reversible arrow(K9) RNHCOO- + H2O are reported. van’t Hoff analyses have resulted in standard molar enthalpies, Delta H-m(o), and entropies, Delta S-m(o), of carbamate formation. A Delta H-m(o) – Delta S-m(o) plot generates a linear correlation for carbamate formation (providing a mean standard molar free energy, Delta G(m)(o), for carbamate formation of about -7 kJ . mol (1)), and this relationship helps provide a guide to the selection of an amine(s) solvent for CO2 capture, in terms of enthalpy considerations. A linear Delta H-m(o) – Delta S-m(o) plot also occurs for carbamate protonation. The formation of the carbamates has been correlated with systematic changes in composition and structure, and steric effects have been identified by comparing molecular geometries obtained using density functional B3LYP/6-311++G(d,p) calculations. Trends in steric effects have been identified in the series of compounds monoethanolamine (MEA), 1-amino-2-propanol, 2-amino-1-propanol (AP) and 2-amino-2-methyl-1-propanol (AMP). In the case of 2-piperidinemethanol, 2-piperidineethanol and 3-piperidinemethanol, strong intramolecular hydrogen bonding is shown to be the likely cause for lack of carbamate formation, and in the ring systems of pyrrolidine, morpholine, piperidine and thiomorpholine trends in carbamate formation (as given by K-9) have been correlated with the internal ring angle at the amine nitrogen, as well as the planarity of the environment around the nitrogen atom. Thiomorpholine-functionalized nanoporous mesopore as a sensing material for Cd2+ carbon paste electrode

A new thiomorpholine-functionalized nanoporous mesopore Mobil Composition of Matter No. 41 (MCM-41), abbreviated as TMMCM-41, was synthesized and applied as a sensing material in construction of a cadmium carbon paste electrode. The design and synthesis of a series of C28 amine-based betulinic acid derivatives as HIV-1 maturation inhibitors is described. This series represents a continuation of efforts following on from previous studies of C-3 benzoic acid-substituted betulinic acid derivatives as HIV-1 maturation inhibitors (MIs) that were explored in the context of C-28 amide substituents. Compared to the C-28 amide series, the C-28 amine derivatives exhibited further improvements in HIV-1 inhibitory activity toward polymorphisms in the Gag polyprotein as well as improved activity in the presence of human serum. However, plasma exposure of basic amines following oral administration to rats was generally low, leading to a focus on moderating the basicity of the amine moiety distal from the triterpene core. The thiomorpholine dioxide (TMD) 20 emerged from this study as a compound with the optimal antiviral activity and an acceptable pharmacokinetic profile in the C-28 amine series. Compared to the C-28 amide 3, 20 offers a 2-to 4-fold improvement in potency towards the screening viruses, exhibits low shifts in the EC50 values toward the V370A and Delta V370 viruses in the presence of human serum or human serum albumin, and demonstrates improved potency towards the polymorphic T371A and V362I virus variants. Synthesis of Oxidation Responsive Vesicles with Different Block Sequences via RAFT Polymerization-induced Self-assembly

Soluble N-acryloyloxy thiomorpholine (NAT) and diacetone acrylamide (DAAM) are used as monomers to form insoluble core blocks in water, while PDMA(35) bearing a trithiocarbonate is utilized as stabilizer and macromolecular chain transfer agent (macro-CTA) to render a RAFT control. Specifically, M-[N-co-D] nano-objects are synthesized via direct RAFT dispersion copolymerization of NAT and DAAM at 70 degrees C employing PDMA35 macro-CTA. To produce M-N-D and M-D-N triblock copolymers, PDMA-PNAT (M-N) and PDMA-PDAAM (M-D) nano-objects are prepared via RAFT dispersion PISA syntheses of NAT and DAAM respectively utilizing PDMA35 macro-CTA and then used for seeded dispersion polymerization of DAAM and NAT respectively without intermediate postpolymerization purification. The thioether moiety in NAT can be oxidized by reactive oxygen species (ROS) into a hydrophilic sulfoxide. Therefore, in the precense of hydrogen peroxide (H2O2), oxidation-responsive morphological degradation of these nano-objects occurs due to the increasing hydrophilicity of NAT units. Given the poor control over polymerization of NAT in pure water, 1,4-dioxane is used as a cosolvent to the PNAT block. So the PISA syntheses are conducted in water/1,4-dioxane (9/1, V/V) mixture to achieve a good control over the molecular weight and narrow distribution. H-1-NMR spectra indicate that quantitative monomer conversions (> 99%) are achieved within 5 h. Differential scanning calorimeter (DLS) and transmission electron microscopy (TEM) are used to characterize final morphologies of PISA-generated nano-objects and morphological evolution of nano-objects in the presence of H2O2 (10 mol/L). These aqueous sequence-controlled PISA formulations are expected to provide responsive nanoparticles with tunable kinetics due to the response-dependent morphological transitions, which may be potentially used as carriers for drug delivery and controlled release.

Thiomorpholine Derivatives with Hypolipidemic and Antioxidant Activity

A number of thiomorpholine derivatives that are structurally similar to some substituted morpholines possessing antioxidant and hypocholesterolemic activity were synthesized. The new compounds incorporate an antioxidant moiety as the thiomorpholine N-substituent. The derivatives were found to inhibit the ferrous/ascorbate-induced lipid peroxidation of microsomal membrane lipids, with IC50 values as low as 7.5 mu M.
