A naphthalimide-based ratiometric fluorescent probe for determining glutathione (GSH) was constructed by installing two oxidized morpholine (e.g. thiomorpholine-S-dioxide and morpholine-N-oxide) components on the off-to-on GSH probe. The radical cations of piperazine, morpholine, thiomorpholine, and thioxane were investigated by electron paramagnetic resonance (EPR) and electron-nuclear double resonance (ENDOR) spectroscopy in a solid Freon matrix. Electrochemical stability of eight complexes of the general formula [Co(III)Rdtc(1-8)cyclam](ClO4)(2), where cyclam = 1,4,8,11-tetraazacyclotetradecane and Rdtc(-) (1-8) = 4-morpholine (Morphdtc), 4-thiomorpholine (Timdtc), 4-piperazine (Pzdtc), N-methyl piperazine (N-Mepzdtc), piperidine (Pipdtc), 2-, 3- or 4-methylpiperidine (2-, 3- or 4-Mepipdtc) dithiocarbamates, respectively, were studied. The substances were examined in aqueous NaClO4 solution and nonaqueous LiClO4 in CH3CN solution by cyclic voltammetry. In aqueous solution, macrocyclic ligand cyclam is characterized by the anodic peak at 0.95 V. The Rdtc(-) ligands have two anodic peaks, one in the region 0.25-0.30 V and the other in the 0.78-0.95 V region. Absence of these anodic peaks in the case of the complexes indicates that coordination to cobalt(III) stabilizes both cyclam and Rdtc(-) ligands, but reversible peaks in the range -0.68 to -0.78 V support the Co(III) redox reaction. In nonaqueous solutions cyclam has one anodic peak at 1.75 V. The ligands with heteroatom in the ring (Morphdtc, Timdtc, Pzdtc, N-Mepzdtc) have two anodic peaks, while the other four ligands (Pipdtc, 2-, 3- and 4-Mepipdtc) have only one anodic peak. In nonaqueous solution again, coordination to Co(III) ion stabilizes the Rdtc(-) ligands and contrary to aqueous solution no Co(III) redox reaction occurs, indicating a greater stability of the complexes in this media. Finally, the electrochemical results are compared with spectroscopic data obtained previously.

Hydroxypyr(id) ones constitute an emerging platform for the design of drug molecules, owing to their favorable biocompatibility and toxicity profiles. Herein, [Ru-II(eta(6)-p-cymene)] complexes with 3-hydroxy-2-pyridinone functionalized with morpholine and thiomorpholine, as a means often used in medicinal chemistry to alter the physicochemical properties of drug compounds, are reported. The compounds underwent hydrolysis of the Ru-Cl bond and the aqua species were stable for up to 48 h in aqueous solution, as observed by H-1 NMR spectroscopy and ESI-MS. The compounds formed adducts with amino acids and proteins through cleavage of the pyridinone ligand. Binding experiments to the nucleosome core particle by means of X-ray crystallography revealed similar reactivity and exclusive binding to histidine moieties of the histone proteins. Preliminary cyclin-dependent kinase 2 (CDK2)/cyclin A kinase inhibitory studies revealed promising activity similar to that of structurally related organometallic compounds.

We evaluated the antihypertensive properties of 4-tert-buthyl-2,6-bis(thiomorpholine-4-ilmethyl)phenol (TBTIF). TBTIF diminished BP and reduced angiotensin I- or angiotensin II-induced vasoconstriction. The presence of Ang-(1-7) induced a greater reduction in vasoconstriction, and this effect was reversed by l-N-G-monomethyl arginine. Moreover, TBTIF decreased the mRNA of ACE and increased the mRNA of ACE2. In conclusion, TBTIF diminished rat BP through nitric oxide-dependent and nitric oxide-independent mechanisms. In contrast to captopril, TBTIF exhibits better antihypertensive properties through mechanisms that involve ACE2.

Specifically, M-[N-co-D] nano-objects are synthesized via direct RAFT dispersion copolymerization of NAT and DAAM at 70 degrees C employing PDMA35 macro-CTA. To produce M-N-D and M-D-N triblock copolymers, PDMA-PNAT (M-N) and PDMA-PDAAM (M-D) nano-objects are prepared via RAFT dispersion PISA syntheses of NAT and DAAM respectively utilizing PDMA35 macro-CTA and then used for seeded dispersion polymerization of DAAM and NAT respectively without intermediate postpolymerization purification. The thioether moiety in NAT can be oxidized by reactive oxygen species (ROS) into a hydrophilic sulfoxide. Therefore, in the precense of hydrogen peroxide (H2O2), oxidation-responsive morphological degradation of these nano-objects occurs due to the increasing hydrophilicity of NAT units. Given the poor control over polymerization of NAT in pure water, 1,4-dioxane is used as a cosolvent to the PNAT block. So the PISA syntheses are conducted in water/1,4-dioxane (9/1, V/V) mixture to achieve a good control over the molecular weight and narrow distribution. H-1-NMR spectra indicate that quantitative monomer conversions (> 99%) are achieved within 5 h. Differential scanning calorimeter (DLS) and transmission electron microscopy (TEM) are used to characterize final morphologies of PISA-generated nano-objects and morphological evolution of nano-objects in the presence of H2O2 (10 mol/L). These aqueous sequence-controlled PISA formulations are expected to provide responsive nanoparticles with tunable kinetics due to the response-dependent morphological transitions, which may be potentially used as carriers for drug delivery and controlled release.
