A palladium-catalyzed domino approach to the synthesis of 10,11-dihydro-5H-dibenzo[b,e][1,4]diazepinones from 2-aminobenzamides and 1,2-dihaloarenes has been developed. Our strategy integrating double N-arylations (inter- and intra-molecular) of 2-aminobenzamides with 1,2-dihaloarenes under palladium-catalyzed conditions is clearly distinct from the current literature available for the synthesis of dibenzodiazepinones. Unlike a previous report described for regioselective N-arylation of 2-aminobenzamide at the amine group, our mechanistic studies support the regioselective N-arylation of 2-aminobenzamide occurring first primarily at the amide group. The translational application of our protocol may be demonstrated in the synthesis of a marketed drug, clozapine.

Multidrug resistance (MDR) is a major obstacle for effective chemotherapeutic treatment of cancer frequently leading to failure of the therapy. MDR is often associated with the overexpression of ABC transport proteins like ABCB1 or ABCG2 which efflux harmful substances out of cells at the cost of ATP hydrolysis. One way to overcome MDR is to apply potent inhibitors of ABC transporters to restore the sensitivity of the cells toward cytostatic agents. This study focusses on the synthesis and evaluation of novel 2,4-disubstituted quinazoline derivatives regarding the structure-activity-relationship (SAR), their ability to reverse MDR and their mode of interaction with ABCG2. Hence, the inhibitory potency and selectivity toward ABCG2 was determined. Moreover, the intrinsic cytotoxicity and the reversal of MDR were investigated. Interaction type studies with the substrate Hoechst 33342 and conformational analyses of ABCG2 with 5D3 monoclonal antibody were performed for a better understanding of the underlying mechanisms. In our study we could further enhance the inhibitory effect against ABCG2 (compound 31, IC50: 55 nM) and identify the structural features that are crucial for inhibitory potency, the impact on transport activity and binding to the protein. The effluent from conventional treatment process (including anaerobic digestion and anoxic-oxic treatment) for pig farm wastewater was difficult to treat due to its low ratio of biochemical oxygen demand to chemical oxygen demand (BOD5/CODCr) (<0.1). In the present study, electro-Fenton (EF) was used to improve the biodegradability of the mentioned effluent and the properties of self-prepared CeO2 doped multi-wall carbon nanotubes (MWCNTs) electrodes were also studied. An excellent H2O2 production (165 mg L-1) was recorded, after an 80-min electrolysis, when the mass ratio of MWCNTs, CeO2 and pore-forming agent (NH4HCO3) was 6:1:1. Results of scanning electron microscopy (SEM), transmission electron microscope (TEM) and x-ray photoelectron spectroscopy (XPS) showed that addition of NH4HCO3 and the doping of CeO2 could increase the superficial area of the electrode as well as the oxygen reduction reaction (ORR) electro-catalytic performance. The BOD5/CODCr of the wastewater from the first stage AO process increased from 0.08 to 0.45 and CODCr reduced 71.5% after an 80-min electrolysis, with 0.3 mM Fe2+ solution. The non-biodegradable chemical pollutants from the first stage AO process were degraded by EF. The non-biodegradable pollutants identified by LC-MS/MS in the effluent from AO process including aminopyrine, oxadixyl and 3-methyl-2-quinoxalinecarboxylic acid could be degraded by EF process, with the removal rates of 81.86%, 34.39% and 7.13% in 80 min, and oxytetracycline with the removal rate of 100% in 20 min. Therefore, electro-Fenton with the new CeO2-doped MWCNTs cathode electrode will be a promising supplement for advanced treatment of pig farm wastewater. (C) 2020 Elsevier Ltd. All rights reserved. An HMPA catalyzed reductive alkylation of ureas and thioureas with trichlorosilane under mild reaction conditions has been developed. Both aldehydes and ketones could be used as efficient alkylation reagents in this method to obtain the desired products in moderate to high yields. A variety of di- and trisubsituted ureas and thioureas were easily prepared by this new reductive alkylation method. This protocol exhibits excellent functional group tolerance, chemoselectivity and practicality.

Imitating the structures and behaviors of natural creatures is of great significance to scientists to explore novel materials for practical applications. However, the design and fabrication of biomimetic devices with complex and outstanding performances is still on the way. A bilayer film composed of liquid crystalline networks (LCN) film and hydrochromic aggregation-induced-emission molecule-doped hydrophilic layer is prepared. Under different relative humidity, the composite film can deform and change fluorescence color simultaneously. The influence of the content of the hydrophilic matrix on the fluorescent property and humidity-responsive behaviors of the bilayer film is investigated. Thanks to the mechanical anisotropy provided by uniform-aligned LCN film, different modes of deformation of the bilayer film are achievable, like bending, curling, and twisting. More importantly, due to the independence of the LCN film and the non-mesogenic molecules brought by this bimorph strategy, complex alignment of the LCN film and modification by immiscible molecules are realized in a single LCN actuator. Based on the functional composite film, artificial flowers showing synergistic blooming and shape-changing is prepared. By regulating the molecular alignment of the LC mesogens of the LCN film, the artificial flowers can imitate various blooming behaviors of natural flowers like confederate jasmine and jade lotus.

In the current research, two mesoporous structures of sodium zincate [Na2Zn(OH)(4)], spheres and aggregated particles, has been synthesized from sulfonation of melamine-formaldehyde (MF) and then bounding [Na2Zn(OH)(4)] particles onto the surface of sulfonated melamine-formaldehyde (SMF). Furthermore, in the present synthetic methodology the zinc oxide (ZnO) thin films on sulfonated melamine-formaldehyde was formed via the chemical deposition, and thermal treatment techniques of the sodium zincate. Several spectroscopic and analytical methods proved the structural and morphological properties of sodium zincate mesoporous structures based on sulfonated melamine formaldehyde [ SMF/Na2Zn(OH)(4)]. The experimental studies showed that these catalytic systems showed high activity in the Biginelli reaction for the synthesis of 3,4-dihydropyrimidin-2(1H)-one derivatives and will find recoverable potential application. The obtained results indicated that the synthesized mesoporous structures had a thermal stability near 300 degrees C, particle-size distribution around of 10-120 nm for spheres and the surface area of 6 and 2 m(2)/g for spheres and aggregated particles, respectively. The serine hydrolase monoacylglycerol lipase (MAGL) is involved in a plethora of pathological conditions, in particular pain and inflammation, various types of cancer, metabolic, neurological and cardiovascular disorders, and is therefore a promising target for drug development. Although a large number of irreversible-acting MAGL inhibitors have been discovered over the past years, there are only few compounds known so far which inhibit the enzyme in a reversible manner. Therefore, much effort is put into the development of novel chemical entities showing reversible inhibitory behavior, which is thought to cause less undesired side effects. To explore a wide range of chemical structures as MAGL binders, we have applied a virtual screening approach by docking small molecules into the crystal structure of human MAGL (hMAGL) and envisaged a library of 45 selected compounds which were then synthesized. Biochemical investigations included the determination of the inhibitory potency on hMAGL and two related hydrolases, i.e. human fatty acid amide hydrolase (hFAAH) and murine cholesterol esterase (mCEase). The most promising candidates from theses analyses, i.e. three w-quinazolinonylalkyl aryl ureas bearing alkyl spacers of three to five methylene groups, exhibited IC50 values of 20-41 mu M and reversible, detergent-insensitive behavior towards hMAGL. Among these compounds, the inhibitor 1-(3,5-bis(tri-fluoromethyl)phenyl)-3-(4-(4-oxo-3,4 dihydroquinazolin-2-yl)butyl)urea (96) was selected for further kinetic characterization, yielding a dissociation constant K-i = 15.4 mu M and a mixed-type inhibition with a pronounced competitive component (alpha = 8.94). This mode of inhibition was further supported by a docking experiment, which suggested that the inhibitor occupies the substrate binding pocket of hMAGL.

Pure organic room-temperature phosphorescence (RTP) and luminescence from nonconventional luminophores have gained increasing attention. However, it remains challenging to achieve efficient RTP from unorthodox luminophores, on account of the unsophisticated understanding of the emission mechanism. Herein, we propose a strategy to realize efficient RTP in nonconventional luminophores through incorporation of lone pairs together with clustering and effective electronic interactions. The former promotes spin-orbit coupling and boosts the consequent intersystem crossing, whereas the latter narrows energy gaps and stabilizes the triplets, thus synergistically affording remarkable RTP. Experimental and theoretical results of urea and its derivatives verify the design rationale. Remarkably, RTP from thiourea solids with unprecedentedly high efficiency of up to 24.5 % is obtained. Further control experiments testify the crucial role of through-space delocalization on the emission. These results will spur the future fabrication of nonconventional phosphors and advance the understanding of the underlying emission mechanism.

Production of nanocomposites from macro-scale materials in situ in a melting compounder is highly desirable yet challenging. In this study, we develop a highly efficient approach for the preparation of cellulose nanofiber (CNF)-reinforced high-density polyethylene (HDPE) composite materials from as-received wood pulp fibers, in which nanofibrillation-assisting plasticizers (urea and urea derivatives) and a diblock copolymer dispersant are used. The most effective plasticizer is urea, which plays a crucial role in producing CNF with less fragmentation during the kneading step, owing to the plasticization/nanofibrillation and the reaction with hydroxyl groups of cellulose fibers into a carbamate. The diblock copolymer as a dispersant enables the stabilization of the appropriate dispersion of the produced CNF in nonpolar HDPE. The resulting composites exhibit significantly improved mechanical properties, including a 6.9-fold increase in the Youngs modulus with 10 wt % loading of wood pulp fibers over that of neat HDPE.
