BACKGROUND Ara h 1 is a major food allergen in peanuts. Recently, many studies have revealed that the Maillard reaction (MR) affects the allergenicity of food proteins. RESULTS To investigate the influence of the MR on the allergenicity of Ara h 1, R-Ara h 1 was processed with glucose in dry heating conditions for different periods. The extent of the MR was assessed by four methods. The changes in secondary and tertiary structures were characterized through spectroscopy assays. Advanced glycation end products (AGE) structures were identified by protein sample dry heating for 60 min, indicating the formation of AGE-Ara h 1. Simulated gastric fluid (SGF) digestion analysis showed that AGE-Ara h 1 has higher resistance to peptic digestion than R-Ara h 1. The BALB/c mouse model was also utilized to explore the effect of the MR on the allergenicity of Ara h 1, and the results showed that the Th2-type cytokines, antibodies, and histamine content increased, and there was a greater degree of degranulation of rat basophilic leukemia (RBL) cells in the AGE-Ara h 1 group compared with the R-Ara h 1 group. CONCLUSION During the process of dry heating, proteins participated in the MR with changes in secondary and tertiary structures. The condition applying a temperature of 100 degrees C for 60 min caused the formation of AGE-Ara h 1. Simulated gastric fluid digestion analysis showed that AGE-Ara h 1 had a greater resistance to peptic digestion than R-Ara h 1. The BALB/c mouse model showed that AGE-Ara h 1 had more allergenicity, indicating that the MR could enhance the allergenicity of Ara h 1. (c) 2020 Society of Chemical Industry

As a member of the Janus OAK) family of nonreceptor tyrosine kinases, TYK2 plays an important role in mediating the signaling of pro-inflammatory cytokines including IL-12, IL-23, and type 1 interferons. The nicotinamide 4, identified by a SPA-based high-throughput screen targeting the TYK2 pseudokinase domain, potently inhibits IL-23 and IFN alpha signaling in cellular assays. The described work details the optimization of this poorly selective hit (4) to potent and selective molecules such as 47 and 48. The discoveries described herein were critical to the eventual identification of the clinical TYK2 JH2 inhibitor (see following report in this issue). Compound 48 provided robust inhibition in a mouse IL-12-induced IFN gamma pharmacodynamic model as well as efficacy in an IL-23 and IL-12-dependent mouse colitis model. These results demonstrate the ability of TYK2 JH2 domain binders to provide a highly selective alternative to conventional TYK2 orthosteric inhibitors.

Production of nanocomposites from macro-scale materials in situ in a melting compounder is highly desirable yet challenging. In this study, we develop a highly efficient approach for the preparation of cellulose nanofiber (CNF)-reinforced high-density polyethylene (HDPE) composite materials from as-received wood pulp fibers, in which nanofibrillation-assisting plasticizers (urea and urea derivatives) and a diblock copolymer dispersant are used. The most effective plasticizer is urea, which plays a crucial role in producing CNF with less fragmentation during the kneading step, owing to the plasticization/nanofibrillation and the reaction with hydroxyl groups of cellulose fibers into a carbamate. The diblock copolymer as a dispersant enables the stabilization of the appropriate dispersion of the produced CNF in nonpolar HDPE. The resulting composites exhibit significantly improved mechanical properties, including a 6.9-fold increase in the Youngs modulus with 10 wt % loading of wood pulp fibers over that of neat HDPE.

Using green chemical method a series of fifteen systematically substituted 3-(9-substituted anthracen-10-yl)-1-(4-phenyl substituted)prop-2-en-1-one derivatives having high nonlinear absorption coefficient have been synthesized. The nonlinear absorption properties of these compounds were studied using nanosecond pulses at 532 nm wavelength. All 15 derivatives are found to show high nonlinear absorption in the range of 35-640 cm/GW. Among these the derivatives with-NO2 as one of the substitution group show higher nonlinear absorption. The origin of high nonlinear absorption with low linear absorption in the compounds has been attributed to two-step two-photon absorption process. We have shown that the energy bands of these compounds are distributed such that for excitation at 532 nm they have strong nonlinear absorption. These properties of the reported compounds make them a potential candidate for biological imaging and other applications which are based on nonlinear absorption. (C) 2020 Elsevier B.V. Novel thiadiazole derivatives of Schiff bases namely (E)-N-(anthracen-9-ylmethylene)5-(4-nitrophenyl)-1,3,4-thiadiazol-2-amine (A9CNPTDA) and N-(anthracen-9(10H)-ylidene)5-(4-nitrophenyl)-1,3,4-thiadiazol-2-amine (ANNPTDA) were synthesized, characterized and corrosion inhibition behaviour, as well as the mechanism of inhibition were investigated by different monitoring techniques like gravimetric measurements, electrochemical impedance spectroscopy, potentiodynamic polarization, quantum chemical and SEM studies. Both of the thiadiazole derivatives showed excellent corrosion inhibitor action on carbon Steel in acid medium. A9CNPTDA exhibited highest inhibition efficiency of 98.04% at 1mM concentration while ANNPTDA showed a maximum of 95.32%. In HCl medium, both derivatives obeyed Langmuir adsorption isotherm and thermodynamic parameters (K-ads , Delta G(ads)(0)) were calculated. An acceptable relationship was observed between the results of quantum chemical calculations and other corrosion monitoring analysis.

A bioinspired iron(III)porphyrinic Zr-MOF, PCN-222(Fe), was modified by post-synthetic cluster metalation with iron(III) chloride, as a cheap, earth-abundant, and environmentally friendly metal precursor, towards formation a new multifunctional MOF, namely Fe@PCN-222(Fe). The MOF consists of bimetallic (Zr-oxo-Fe) nodes linked by Fe(III)porphyrin struts. The cluster metalation and pre-activation treatment of PCN-222(Fe) were performed cooperatively using the FeCl3. The respective MOF was characterized through various techniques, such as FT-IR, PXRD, ICP-AES, BET surface area, SEM, UV-Vis DRS, TGA/DSC, PL, and XPS analyses. The solid showed catalytic activity for one-pot tandem synthesis of quinazolin-4(3H)-ones from alcohols and 2-aminobenzamide through a three-consecutive-step reaction (oxidation-cyclization-oxidation) under visible light irradiation using air or oxygen without adding any additive. In addition, its catalytic performance was superior to that of the bare PCN-222(Fe) and the corresponding homogeneous catalysts. The experiments indicate that the solid MOF acts as both a photoredox and Lewis acid catalyst. Hot-filtration and Fe-leaching tests as well as reusability experiments confirm that the nominal MOF acts as an efficient reusable heterogeneous catalyst for at least three runs without significant decrease in its activity. This work demonstrates the potential of using MOFs as supports for single-site metal species towards preparation of multifunctional MOFs for modern organic transformations combining photocatalysis and catalysis. Inhibition of cyclin dependent kinase 4 (Cdk4) prevents cancer cells from entering the early G(0)/G(1) phase of the cell division cycle whereas inhibiting tubulin polymerization blocks cancer cells’ ability to undergo mitosis (M) late in the cell cycle. We had reported earlier that two non-planar and relatively non-toxic fascaplysin derivatives, an indole and a tryptoline, inhibit Cdk4 with IC50 values of 6.2 and 10 mu M, respectively. Serendipitously, we had also found that they inhibited tubulin polymerization. The molecules were efficacious in mouse tumor models. We have now identified Cink4T in a 59-compound quinazolinone library, designed on the basis of ligand-based virtual screening, as a compound that inhibits Cdk4 and tubulin. Its IC50 value for Cdk4 inhibition is 0.47 mu M and >50 mu M for inhibition of Cdk1, Cdk2, Cdk6, Cdk9. Cink4T inhibits tubulin polymerization with an IC50 of 0.6 mu M. Molecular modelling studies on Cink4T with Cdk4 and tubulin crystal structures lend support to these observations. Cancer cell cycle analyses confirm that Cink4T blocks cells at both G(0)/G(1) and M phases as it should if it were to inhibit both Cdk4 and tubulin polymerization. Our results show, for the very first time, that virtual screening can be used to design novel inhibitors that can potently block two crucial phases of the cell division cycle. Biodegradation of the phenylurea herbicide linuron appears a specialization within a specific Glade of the Variovorax genus. The linuron catabolic ability is likely acquired by horizontal gene transfer but the mechanisms involved are not known. The full-genome sequences of six linuron-degrading Variovorax strains isolated from geographically distant locations were analyzed to acquire insight into the mechanisms of genetic adaptation toward linuron metabolism. Whole-genome sequence analysis confirmed the phylogenetic position of the linuron degraders in a separate Glade within Variovorax and indicated that they unlikely originate from a common ancestral linuron degrader. The linuron degraders differentiated from Variovorax strains that do not degrade linuron by the presence of multiple plasmids of 20-839 kb, including plasmids of unknown plasmid groups. The linuron catabolic gene clusters showed 1) high conservation and synteny and 2) strain-dependent distribution among the different plasmids. Most of them were bordered by IS1071 elements forming composite transposon structures, often in a multimeric array configuration, appointing IS1071 as a key element in the recruitment of linuron catabolic genes in Variovorax. Most of the strains carried at least one (catabolic) broad host range plasmid that might have been a second instrument for catabolic gene acquisition. We conclude that Glade 1 Variovorax strains, despite their different geographical origin, made use of a limited genetic repertoire regarding both catabolic functions and vehicles to acquire linuron biodegradation.

In this study, new tri-amides 5a-1 have been synthesized via a one-pot four-component Ugi reaction between repaglinide, aniline derivatives, aldehyde derivatives, and cyclohexyl isocyanide in good yields. These compounds were evaluated against yeast alpha-glucosidase. Obtained in vitro alpha-glucosidase results demonstrated that all the synthesized compounds 5a-1 were more potent than standard inhibitor acarbose. Among them, the most potent compounds were compounds 5j, 5k, and 5h. The kinetic analysis of the most potent compound 5j revealed that this compound is a competitive inhibitor for alpha-glucosidase (K-i = 24 mu M). Furthermore, docking study of the most potent compounds was also performed in the alpha-glucosidase active site to find interaction modes and binding energies of these compounds. Amyotrophic lateral sclerosis (ALS) is a fatal motor neuron disease for which the existing candidate biomarkers (neurofilaments) have low specificity. Changes in blood IgG N-glycosylation have been observed in several diseases, including ALS, whereas cerebrospinal fluid (CSF) IgG has been less studied. Here, we characterized N-glycans of CSF IgG from ALS patients in comparison with a control group of other neurological diseases. Cerebrospinal fluid was collected from patients with ALS (n=26) and other neurological diseases (n=10). N-Glycans were released from CSF purified IgG with peptide N-glycosidase F, labeled with 2-aminobenzamide and analyzed by NP-HPLC chromatography in combination with exoglycosidase digestion and MALDI-TOF mass spectrometry. The N-glycosylation profile of ALS CSF IgG consisted of diantennary N-glycans predominantly with proximal fucose and some bisecting GlcNAc; agalacto-, mono-, and digalactosylated as well as alpha 2,6-sialylated structures were detected. Differences between ALS and control patients were observed; most relevant was the increase in ALS CSF IgG of the level of galactosylated structures defined here as Gal-index (median 46.87 and 40.50% for ALS and controls, respectively; p=0.006). The predictive value of the Gal-index (AUC=0.792, p=0.007) considering ROC analysis had potential utility as a diagnostic test for ALS and was comparable to that of phosphoneurofilament heavy chain (AUC=0.777, p=0.011), which was used as benchmark marker for our group of patients. The results provide the basis to further explore the potential of IgG N-glycan galactosylation as biomarker for ALS by using larger cohorts of patients and controls.
