A highly efficient and environmentally benign synthesis of N-fused heterocyclic compounds including pyrido[1,2-a]pyrimidine-6-one moiety is successfully achieved via a sequential four-component reaction. The process involves the formation of diversely substituted 9-nitro-1,2,3,4,7,8-hexahydro-6H-pyrido[1,2-a]pyrimidin-6-ones from the reaction of Meldrum’s acid, benzaldehydes, 1,1-bis(methylthio)-2-nitroethylene and various diamines in the presence of p-toluene sulfonic acid (PTSA) as an acidic catalyst in water as a green solvent. The salient features of the present methodology are easily available starting materials, the use of water as environmentally benign solvent, simple execution, applicable to a wide range of starting materials and good to excellent yields.

Background: Colorectal familial adenomatous polyposis (FAP) adenomas exhibit a uniform pathogenetic basis caused by a germline mutation in the adenomatous polyposis gene (APC), but the molecular changes leading to their development are incompletely understood. However, dysregulated apoptosis is known to substantially affect the development of colonic adenomas. One of the key regulatory proteins involved in apoptosis is apoptosis repressor with caspase recruitment domain (ARC). Methods: The expression of nuclear and cytoplasmic ARC in 212 adenomas from 80 patients was analyzed by immunohistochemistry. We also compared expression levels of ARC with the expression levels of p53, Bcl-2, COX-2, and MMR proteins. Statistical analyses were performed by Spearman’s rank correlation and linear regression test. Results: ARC was overexpressed in the nuclei and cytoplasm of most FAP adenomas investigated. Cytoplasmic ARC staining was moderately stronger (score 2) in 49.1% (n = 104/212) and substantially stronger (score 3) in 32.5% (n = 69/212) of adenomas compared to non-tumorous colorectal mucosa. In 18.4% (n = 39/212) of adenomas, cytoplasmic ARC staining was equivalent to that in non-tumorous mucosa. Nuclear expression of ARC in over 75% of cells was present in 30.7% (n = 65/212) of investigated adenomas, and nuclear expression in 10-75% of cells was detected in 62.7% (n = 133/212). ARC expression in under 10% of nuclei was found in 6.6% (n = 14/212) of adenomas. The correlation between nuclear ARC expression and cytoplasmic ARC expression was highly significant (p = 0.001). Moreover, nuclear ARC expression correlated positively with overexpression of Bcl-2, COX-2 p53 and beta-catenin. Cytoplasmic ARC also correlated with overexpression of Bcl-2. Sporadic MMR deficiency was detected in very few FAP adenomas and showed no correlation with nuclear or cytoplasmic ARC. Conclusions: Our results demonstrated that both cytoplasmic and nuclear ARC are overexpressed in FAP adenomas, thus in a homogenous collective. The highly significant correlation between nuclear ARC and nuclear beta-catenin suggested that ARC might be regulated by beta-catenin in FAP adenomas. Because of its further correlations with p53, Bcl-2, and COX-2, nuclear ARC might play a substantial role not only in carcinomas but also in precursor lesions.

Background: Nivolumab was the first immune checkpoint inhibitor approved for previously treated advanced nonesmall cell lung cancer (NSCLC). Before its introduction in the market, nivolumab was made available to NSCLC patients through an expanded access program (EAP). Here we present the Italian cohort of patients with non-squamous NSCLC enrolled in a worldwide nivolumab EAP, with subgroup analyses involving elderly patients, patients with central nervous system (CNS) metastases and patients receiving nivolumab beyond progression. Methods: Pretreated patients with advanced non-squamous NSCLC received nivolumab at 3 mg/kg every 2 weeks up to 24 months. Efficacy data (investigator-assessed tumour response, progression date and survival) and safety data were collected. Findings: 1588 patients were treated across 153 Italian centres. Overall response rate and disease control rate were 18% and 44%, respectively; median overall survival (OS) was 11.3 months (95% CI: 10.2-12.4). Elderly patients (>= 70 n = 522; >= 75 n = 232) achieved outcomes similar to the global study population; patients with CNS metastases (n = 409) had an OS of 8.6 months (95% CI: 6.4-10.8), and a 1-year OS rate of 43%. Nivolumab was administered beyond progression to 276 patients (26%), 57 of whom achieved subsequent disease control; the median OS of patients receiving nivolumab beyond progression was 16.2 months (95% CI: 14.0-18.4), while 1-year OS rate was 62%. Interpretation: To date, this is the largest clinical experience with nivolumab in a real-world setting. Cinchona-based chiral hyperbranched polymers (HBPs) were designed and successfully synthesized via the Mizoroki-Heck (MH) coupling reaction. AB(2) and A(2)B-type chiral monomers were prepared from cinchona squaramide derivatives, where A (vinyl) reacted only with B (iodophenyl) under MH reaction conditions. The chiral HBPs obtained by the MH polymerization contained cinchona squaramide moieties and demonstrated excellent diastereoselectivity and enantioselectivity in asymmetric Michael addition reactions of methyl 2-oxocyclopentanecarboxylate and trans-beta-nitrostyrene. This work presents a detailed study of the reactivity of three mononuclear Cu-II complexes supported by derivatives of the tetradentate ligand N,N’-bis(2-pyridylmethyl)-1,2-ethylenediamine (bispicen). The Cu-II complexes are capable of performing C – H bond activation in the presence of NEt3 and H2O2 through what has been proposed computationally to be a [CuO](+) intermediate. A wavefunction-based quantum chemical investigation into the electronic structure of the proposed [CuO](+) intermediate reveals a triplet ground state predominantly consistent with an S = 1/2 Cu-II center ferromagnetically coupled to an oxyl radical, though contributions from the corresponding biradicaloid CM-oxen resonance structure may be nontrivial. Furthermore, correlation of the electronic structure of the proposed intermediate with analogous high-valent metal-oxo species capable of olefin epoxidation suggests that the Cu-II complexes might be also capable of olefin epoxidation in the presence of NEt3 and H2O2. To test this hypothesis experimentally, the Cu-II complexes are treated with NEt 3 and H2O2 in the presence of alkene substrates, resulting in the formation of epoxides.

A new ionic liquid was synthesised, and its chemical structure was elucidated by FT-IR, 1D NMR, 2D NMR, and mass analyses. Some physical properties, thermal behaviour, and thermal stability of this ionic liquid were investigated. The formation of a two-protonic acid salt namely 4,4′-trimethylene-N,N’-dipiperidinium sulfate instead of 4,4′-trimethylene-N,N’-dipiperidinium hydrogensulfate was evidenced by NMR analyses. The catalytic activity of this ionic liquid was demonstrated in the esterification reaction of n-butanol and glacial acetic acid under different conditions. The desired acetate was obtained in 62-88% yield without using a Dean-Stark apparatus under optimal conditions of 10 mol-% of the ionic liquid, an alcohol to glacial acetic acid mole ratio of 1.3 : 1.0, a temperature of 75-100 degrees C, and a reaction time of 4 h. alpha-Tocopherol (alpha-TCP), a highly efficient form of vitamin E, was also treated with glacial acetic acid in the presence of the ionic liquid, and O-acetyl-alpha-tocopherol (Ac-TCP) was obtained in 88.4% yield. The separation of esters was conducted during workup without the utilisation of high-cost column chromatography. The residue and ionic liquid were used in subsequent runs after the extraction of desired products. The ionic liquid exhibited high catalytic activity even after five runs with no significant change in its chemical structure and catalytic efficiency.

An efficient microwave assisted synthesis of 2-amino-3-carboxamide-1,1′-biaryls derivatives 4a-p and terphenyl derivative 5a from 2-amino-5-iodobenzamide 2a, 2-amino-3,5-diiodobenzamide 2b and (het)aryl boronic acids via Suzuki coupling has been achieved. Synthetic utility of the product 4-amino-4′-cyano-[1,1′-biphenyl]-3-carboxamide 4j has been demonstrated for the synthesis of 4-(aryl amino)-[1,1′-biphenyl]-3-carboxamide derivatiyes 10a-c via Chan-Evans-Lam coupling reaction. Furthermore, 4-(4′-oxo-3′, 4′-dihydro-1’H-spirof[fluorene-9,2′-quinazolin]-6′-yl)benzonitrile 12a was obtained from biaryl derivative 4j and fluorenone 11a and 6(4-methoxyphenyl)2-(ferrocenyl) quinazolin-4(3H)-one 12b from biaryl derivative 4k and ferrocenealdehyde lib using phosphotungstic acid as green catalyst and solvent free microwave irradiation condition. Remarkably, all the synthesized 2-amino-3-carboxamide-1,1′-biaryls and 4-(aryl amino)-[1,1′-biphenyl]-3-carboxamide derivatives 4a-p showed luminescence in the blue region with large Stokes shift. Significantly, 2-amino-3-carboxamide-1,1′-biaryls 4d and 4j showed high fluorescence quantum yields (Phi(f)) 0.54 and 0.84, respectively.

In this work, a novel and high-throughput parallel-dispersive droplet extraction (Pa-DDE) based on in situ formation of the hydrophobic MILs ([Co(C4IM)(4)(+2)]2[NTf2-], [Ni(C4IM)(4)(+2)]2[NTf2-] and [Ni(BeIM)(4)(+2)]2[NTf2]) is demonstrated, for the first time, for the determination of benzophenone, metolachlor, triclocarban, pendimethalin, 4-methylbenzylidene camphor, and 2-ethylhexyl-4-methoxycinnamate from aqueous environmental samples. This experimental setup is comprised of a 96-well plate system containing a set of magnetic pins which were used to collect the MIL droplet after in situ formation. This consolidated system enabled simultaneous extraction of up to 96 samples and MIL production in one step. Using this apparatus, sample preparation times of 0.78 min per sample was achieved. The experimental conditions were carefully optimized using uni and multivariate approaches. The optimal conditions were comprised of sample volume of 1.25 mL, 4 mg of [Co (C4IM)(4)(+2)]2[Cl] and 40 mu L of LiNTf2 for the in situ formation, and dilution in 20 mu L of acetonitrile. The analytical parameters of merit were successfully determined with LODs ranging from 7.5 to 25 mu g L-1 and coefficients of determination higher than 0.989. Intraday and interday precision ranged from 6.4 to 20.6% (n = 3) and 11.6-22.9% (n = 9), respectively, with analyte relative recovery ranging between 53.9 and 129.1%.

Quantitative structure-property relationship (QSPR) modelling has been used in many scientific fields. This approach has been extensively applied in environmental research to predict physicochemical properties of compounds with potential environmental impact. The soil sorption coefficient is an important parameter for the evaluation of environmental risks, and it helps to determine the final fate of substances in the environment. In the last few years, different QSPR models have been developed for the determination of the sorption coefficient. In this study, several QSPR models were generated and evaluated for the prediction of log K-oc from the relationship with log P. These models were obtained from an extensive and diverse training set (n = 639) and from subsets of this initial set (i.e. halves, fourths and eighths). The aim of this study was to investigate whether the size of the training set affects the statistical quality of the obtained models. Furthermore, statistical equivalence was verified between the models obtained from smaller sets and the model obtained from the total training set. The results confirmed the equivalence between the models, thus indicating the possibility of using smaller training sets without compromising the statistical quality and predictive capability, as long as most chemical classes in the test set are represented in the training set.

Current work describes the environmentally benign approach for the synthesis of o-ureidobenzonitriles, from o-aminobenzamides and isothiocyanates using T3P. Here, the conversion of thiourea to urea and amide to nitrile take place simultaneously via unprecedented intramolecular rearrangement. This protocol is operationally facile and offers wide variety of o-ureidobenzonitriles at room temperature in good to excellent yields.
