N-Tosylhydrazones have proven to be versatile synthons over the past several decades. However, to our knowledge, the construction of isoxazolines based on N-tosylhydrazones has not been examined. Herein, we report the first demonstrations of [2 + 2 + 1] cycloaddition reactions that allow the facile synthesis of isoxazolines, employing N-tosylhydrazones, tert-butyl nitrite (TBN) and alkenes as reactants. This process represents a new type of cycloaddition reaction with a distinct mechanism that does not involve the participation of nitrile oxides. This approach is both general and practical and exhibits a wide substrate scope, nearly universal functional group compatibility, tolerance of moisture and air, the potential for functionalization of complex bioactive molecules and is readily scaled up. Both control experiments and theoretical calculations indicate that this transformation proceeds via the in situ generation of a nitronate from the coupling of N-tosylhydrazone and TBN, followed by cycloaddition with an alkene and subsequent elimination of a tert-butyloxy group to give the desired isoxazoline.

In this study, an efficient hybrid nanocatalyst made of guar gum (guarana, as a natural basis), magnetic iron oxide nanoparticles, and copper(I) oxide nanoparticles (Cu2O NPs) is fabricated and suitably applied for catalyzing the multicomponent (three- and four-component) synthesis reactions of imidazole derivatives. Here, an easy preparation strategy for this novel catalytic system (Cu2O/Fe3O4@guarana) is presented. Then, the application of this catalytic system for the synthesis of imidazole derivatives is precisely investigated. For this purpose, ultrasonication is introduced as an efficient and fast method. In summary, the high catalytic efficiency of Cu2O/Fe3O4@guarana nanocomposite is well demonstrated by high reaction yields obtained in the presence of a small amount of this nanocomposite, under mild conditions. Wide active surface area, substantial magnetic behavior, excellent heterogeneity, suitable stability, well reusability, and etc. have distinguished this catalytic system as an instrumental tool for facilitating the complex synthetic reactions.

In this work, 6-tert-butyl 3-ethyl 2-amino-4,5-dihydrothieno[2,3-c]pyridine-3,6(7H)-dicarboxylate is synthesized from starting tert-butyl 4-oxopiperidine-1-carboxylate, ethyl cyanomalonate, and sulfur, and then, coupled with same aromatic aldehyde affords the corresponding Schiff base compounds. These compounds (2a-d) are characterized using FTIR, H-1 and C-13 NMR spectroscopic methods. The crystal and molecular structure of (E)-6-tert-butyl 3-ethyl 2-((2-hydroxy-3-methoxybenzylidene)amino)-4,5-dihydrothieno[2,3-c]pyridine-3,6(7H)-dicarboxylate (2a) is characterized by the X-ray crystallographic analysis. Compound 2a crystallizes in the monoclinic space group P2(1)/c. The molecular and crystal structure is stabilized by two O-HMIDLINE HORIZONTAL ELLIPSISN and O-HMIDLINE HORIZONTAL ELLIPSISO intramolecular hydrogen bonds (OMIDLINE HORIZONTAL ELLIPSISN and OMIDLINE HORIZONTAL ELLIPSISO are 2.598(5) angstrom and 2.990(5) angstrom, respectively; O-HMIDLINE HORIZONTAL ELLIPSISN = 147 degrees and O-HMIDLINE HORIZONTAL ELLIPSISO = 134 degrees). According to DFT, compound 2d also shows the intramolecular hydrogen bonding, while there is no this type of interaction in compound 2b and 2c.

A one-pot quick and efficient multicomponent reaction has been developed for the synthesis of a new series of functionalized 8-hydroxy-4-phenyl-1,2-dihydroquinoline derivatives using 30 mol% ammonium acetate in ethanol as solvent. This economical protocol run smoothly to give variety of quinoline derivatives in 55% to 98% yield from inexpensive reagents and catalyst in mild reaction conditions. Various spectroscopic techniques like FTIR, H-1 NMR and C-13 NMR, MALDI-TOF-MS, and EI-MS were used to study and confirm their structure.

A series of chalcone ligands and their corresponding vanadyl complexes of composition [VO (LI-IV)(2)(H2O)(2)]SO4(where L-I= 1,3-Diphenylprop-2-en-1-one, L-II= 3-(2-Hydroxy-phenyl)-1-phenyl-propenone, L-III= 3-(3-Nitro-phenyl)-1-phenyl-propenone, L-IV= 3-(4-Methoxy-phenyl)-1-phenyl-propenone) have been synthesized and characterized using various spectroscopic (Fourier-transform infrared, electrospray ionization mass, nuclear magnetic resonance, electron paramagnetic resonance, thermogravimetric analysis, vibrating sample magnetometer) and physico-analytic techniques. Antidiabetic activities of synthesized complexes along with chalcones were evaluated by performing in vitro and in silico alpha-amylase and alpha-glucosidase inhibition studies. The obtained results displayed moderate to significant inhibition activity against both the enzymes by vanadyl chalcone complexes. The most potent complexes were further investigated for the enzyme kinetic studies and displayed the mixed inhibition for both the enzymes. Further, antioxidant activity of vanadyl chalcone complexes was evaluated for their efficiency to release oxidative stress using 2,2-diphenyl-1-picryl-hydrazyl-hydrate assay, and two complexes (Complexes2and4) have demonstrated remarkable antioxidant activity. All the complexes were found to possess promising antidiabetic and antioxidant potential.

Uracil compounds participate in a wide range of biological activities; however, reports on their synthesis using heterogeneous catalysts are scarce. In this work, the multicomponent synthesis of uracil derivatives assisted by layered double hydroxides (LDH) was studied under green chemistry conditions. The incorporation of Ni2+ or Co2+ was successfully performed by co-precipitation method. The yields to uracil derivatives are associated with the presence of weak basic sites and a better interaction of the reagents when the reaction is carried out in solvent-free conditions. The reaction pathway involves the formation of an enone between benzaldehyde and ethyl cyanoacetate, and subsequent reaction with urea, which is assisted by the presence of a basic catalyst. The scope of this synthesis is illustrated with nine examples.

An efficient four-component strategy has been developed for the synthesis of a novel series of (E)-N-benzylidene-3-(benzylthio)-5-(3,5-dimethyl-1H-pyrazol-1-yl)-4H-1,2,4-triazol-4-amines in good yields. The method is effective for the synthesis of benzylidene aralkyl/allyl/propargyl thio pyrazolyl-1,2,4-triazol-4-amines in a one-pot operation. This method is operationally simple, metal-free, and works under reflux conditions with diverse substrates. Among all the synthesized compounds 5a, b, c, h, j , and n were shown good results in silico studies. 5n is showing good results in both DNA binding and molecular docking studies. A novel structure of sinomenine isoxazole derivatives is synthesised from sinomenine hydrochloride and aromatic aldehydes and requires six steps. 19 target compounds have been obtained in good yields. The sinomenine hydrochloride transforms to 4-alkynyl sinomenine, which is a key intermediate product to synthesise the target sinomenine isoxazole compounds, after a neutralisation reaction with ammonia and substitution reaction with 3-chloropropyne. Another key intermediate product is 1,3-dipole, which can be obtained from aromatic aldehyde. After treatment with hydroxylamine hydrochloride and then sodium carbonate solution, aromatic aldehyde is converted to aldehyde oxime, which reacts with N-chlorosuccinimide (NCS) to afford aryl hydroximino chloride. 1,3-Dipole is eventually formed in situ while triethylamine (TEA) in DMF is added dropwise. Then 4-alkynyl sinomenine is added to provide the sinomenine isoxazole derivative via 1,3-dipolar cycloaddition reaction as the key step. All the target compounds are characterised by melting point, H-1 NMR, C-13 NMR, HRMS and FT-IR spectroscopy.

Nitrogen-doped carbon catalysts have attracted increasing research attention due to several advantages for catalytic application. Herein, cost-effective, renewable biomass chitosan was used to prepare a N-doped carbon modified with iron oxide catalyst (Fe2O3@NC) for nitrile synthesis. The iron oxide nanoparticles were uniformly wrapped in the N-doped carbon matrix to prevent their aggregation and leaching. Fe2O3@NC-800, which was subjected to carbonization at 800 degrees C, exhibited excellent activity, selectivity, and stability in the catalytic ammoxidation of aromatic aldehydes to aromatic nitriles. This study may provide a new method for the fabrication of an efficient and cost-effective catalyst system for synthesizing nitriles.
