Objective: To assess the antidiabetic effect of Opuntia dillenii seed oil on rats with diabetes mellitus. Methods: A rat diabetes model was established by intraperitoneal injection of rats with 50 mg/kg streptozotocin. Thirty albino Wistar rats were divided into five groups: the diabetic control group and normal control group were treated only with distilled water, two diabetic groups received 1 and 2 mL/kg of oil per day, respectively, for 30 days and one diabetic group received 2 mg/kg of glibenclamide. In addition, blood glucose was determined weekly. Body weight, average daily food, water intake and urinary volume of each animal were determined before and after the treatment period. After the treatment period, hepatic glycogen was determined using the anthrone reagent, and glycosuria, total cholesterol, triglycerides, alanine aminotransferase, aspartate aminotransferase, urea, creatinine and uric acid were estimated using common clinical diagnostic kits. Results: Oral intake of the oil at 1 and 2 mL/kg for the diabetic animals significantly diminished blood glucose, glycosuria, total cholesterol, triglycerides, alanine aminotransferase, aspartate aminotransferase, urea, creatinine and uric acid, accompanied by a noticeable elevation in the amount of hepatic glycogen in comparison with the diabetic control group. Similarly, Opuntia dillenii seed oil significantly increased the food intake and decreased the urinary volume per day in treated rats of the same groups in comparison with the period before the treatment intervention and attenuated body weight loss in the diabetic rats. Moreover, this effect of the oil was dose dependent. On the other hand, the oil did not affect their need for water. Conclusions: The results show that Opuntia dillenii seed oil has a very important antidiabetic effect on streptozotocin-induced diabetic rats. Hence, we suggest it as a preventive control of diabetes mellitus.

12-Deoxyphorbol-13-phenylacetate (DPP), found in Euphorbia resinifera, is a prototype of a new class of phorbol derivatives that function as protein kinase C activators with potent anti-tumor-promoting activity. To better understand the biosynthesis of DPP in E. resinifera and develop methods for its production, we used next-generation sequencing technologies to build a transcriptome dataset. We obtained a total of approximately 27,102,078 clean reads that were assembled into 90,448 unigenes. Of those, 129 and 59 unigenes were identified in the Terpenoid Backbone Biosynthesis (TBB) (KEGG map00900) and the Diterpenoid Biosynthesis (DB) (KEGG map00904) pathways, respectively. We investigated the expression patterns of 23 DPP biosynthesis-related genes and validated the results by quantitative real time PCR. The results showed that CMK (4-diphosphocytidyl-2-C-methyl-o-erythritol kinase), MECS (2-C-methyl-D-erythritol 2,4-cyclodiphosphate synthase), HDS (1-hydroxy-2-methyl-2-(E)-butenyl 4-diphosphate synthase), and IPI (Isopentenyl-diphosphate Delta-isomerase) of the methylerythritol 4-phosphate (MEP) pathway had a higher expression compared to other genes of TBB, whereas the expression levels of genes involved in the mevalonic acid (MVA) pathway were relatively lower and not significantly different from each other. Furthermore, the expression of the 23 genes in root, stem and young stem differed, and the statistical results indicated that HMGS (Hydroxymethylglutaryl-CoA synthase), MVK (mevalonic acid), MECT (2-C-methyl-D-erythritol 4-phosphate cytidylyltransferase), MECS, HDS and IPI in the TBB pathway as well as KS in the DB pathway were significantly and positively correlated with the CS (casbene synthase) gene. These results suggested that both cytosol-localized MVA and plastid-localized MEP pathways may have a significant contribution in DPP accumulation, with MEP pathway playing a dominant role. To the best of our knowledge, this study is the first transcriptome analysis of E. resinifera and the results reported here will aid the identification of functional genes and better understanding of DPP biosynthesis.

Cinchona-based chiral hyperbranched polymers (HBPs) were designed and successfully synthesized via the Mizoroki-Heck (MH) coupling reaction. AB(2) and A(2)B-type chiral monomers were prepared from cinchona squaramide derivatives, where A (vinyl) reacted only with B (iodophenyl) under MH reaction conditions. The chiral HBPs obtained by the MH polymerization contained cinchona squaramide moieties and demonstrated excellent diastereoselectivity and enantioselectivity in asymmetric Michael addition reactions of methyl 2-oxocyclopentanecarboxylate and trans-beta-nitrostyrene. These newly designed HBPs were structurally robust and could be reused for further reaction without losing their catalytic activity. (C) 2020 Elsevier Ltd. All rights reserved.

BACKGROUND Ara h 1 is a major food allergen in peanuts. Recently, many studies have revealed that the Maillard reaction (MR) affects the allergenicity of food proteins. RESULTS To investigate the influence of the MR on the allergenicity of Ara h 1, R-Ara h 1 was processed with glucose in dry heating conditions for different periods. The extent of the MR was assessed by four methods. The changes in secondary and tertiary structures were characterized through spectroscopy assays. Advanced glycation end products (AGE) structures were identified by protein sample dry heating for 60 min, indicating the formation of AGE-Ara h 1. Simulated gastric fluid (SGF) digestion analysis showed that AGE-Ara h 1 has higher resistance to peptic digestion than R-Ara h 1. The BALB/c mouse model was also utilized to explore the effect of the MR on the allergenicity of Ara h 1, and the results showed that the Th2-type cytokines, antibodies, and histamine content increased, and there was a greater degree of degranulation of rat basophilic leukemia (RBL) cells in the AGE-Ara h 1 group compared with the R-Ara h 1 group. CONCLUSION During the process of dry heating, proteins participated in the MR with changes in secondary and tertiary structures. The condition applying a temperature of 100 degrees C for 60 min caused the formation of AGE-Ara h 1. Simulated gastric fluid digestion analysis showed that AGE-Ara h 1 had a greater resistance to peptic digestion than R-Ara h 1. The BALB/c mouse model showed that AGE-Ara h 1 had more allergenicity, indicating that the MR could enhance the allergenicity of Ara h 1. (c) 2020 Society of Chemical Industry

Using green chemical method a series of fifteen systematically substituted 3-(9-substituted anthracen-10-yl)-1-(4-phenyl substituted)prop-2-en-1-one derivatives having high nonlinear absorption coefficient have been synthesized. The nonlinear absorption properties of these compounds were studied using nanosecond pulses at 532 nm wavelength. All 15 derivatives are found to show high nonlinear absorption in the range of 35-640 cm/GW. Among these the derivatives with-NO2 as one of the substitution group show higher nonlinear absorption. The origin of high nonlinear absorption with low linear absorption in the compounds has been attributed to two-step two-photon absorption process. We have shown that the energy bands of these compounds are distributed such that for excitation at 532 nm they have strong nonlinear absorption. These properties of the reported compounds make them a potential candidate for biological imaging and other applications which are based on nonlinear absorption. (C) 2020 Elsevier B.V. A series of pendant anthracene-functionalized ethylene/propylene copolymer rubbers (EPRs) with UV-light response and self-healing properties are prepared in this work. The reversible crosslinked networks are achieved based on the highly reversible nature of [4 + 4] cycloaddition of anthracene derivatives. The reversible crosslinking process is characterized by UV-vis spectrophotometer, which gives a high conversion yield of approximate to 90% as well as the reversibility of approximate to 70%. This reversible reaction is successfully applied to the self-healing of EPR material. The healing efficiency is evaluated quantitatively by tensile test measurement. In terms of stress at break, the healing efficiency of crosslinked EPR with anthracene content of 5.7 mol% can reach 82.9%. The anthracene-functionalized EPR might have a potential prospect of application in self-healing field.

A new protocol for ring-closing metathesis/isomerization sequence was developed. The reactions of selected dienes were performed in overheated 2-methyltetrahydrofuran at 120 degrees C and provided a wide range of cyclic vinyl ethers and amides with good yields and selectivities. Computational analysis suggests that the relative yield of products depends on a thermodynamically driven process on the basis of relative stabilities of isomers.

This work presents a detailed study of the reactivity of three mononuclear Cu-II complexes supported by derivatives of the tetradentate ligand N,N’-bis(2-pyridylmethyl)-1,2-ethylenediamine (bispicen). The Cu-II complexes are capable of performing C – H bond activation in the presence of NEt3 and H2O2 through what has been proposed computationally to be a [CuO](+) intermediate. A wavefunction-based quantum chemical investigation into the electronic structure of the proposed [CuO](+) intermediate reveals a triplet ground state predominantly consistent with an S = 1/2 Cu-II center ferromagnetically coupled to an oxyl radical, though contributions from the corresponding biradicaloid CM-oxen resonance structure may be nontrivial. Furthermore, correlation of the electronic structure of the proposed intermediate with analogous high-valent metal-oxo species capable of olefin epoxidation suggests that the Cu-II complexes might be also capable of olefin epoxidation in the presence of NEt3 and H2O2. To test this hypothesis experimentally, the Cu-II complexes are treated with NEt 3 and H2O2 in the presence of alkene substrates, resulting in the formation of epoxides.

Chiral polyureas of cinchona alkaloids were synthesized via repetitive Mizoroki-Heck (MH) coupling reaction. The Pd-catalyzed polycondensation of cinchona urea dimers6and aromatic diiodide7afforded the chiral polyureas (P1-P4). The catalytic activity of the chiral polymers was subsequently investigated. The asymmetric Michael addition of ketoesters to nitroolefins was successfully catalyzed by the polymeric organocatalysts (P1-P4) to give the corresponding Michael adducts with high catalytic activities and excellent enantionselectivities (up to >99% ee). The polymeric catalysts were insoluble in commonly used organic solvents. They were easily recovered and reused several times without any loss of the catalytic activity.
