Organic pollutants associated with diesel exhaust particles (DEP), such as polycyclic aromatic hydrocarbons (PAHs) and their derivatives, may negatively impact human health. However, a comprehensive overview of their effects on endocrine nuclear receptor activities is still missing. Here, we evaluated the effects of extracts and chromatographic fractions (fractionated according to increasing polarity) of two standard reference materials derived from distinct types of diesel engines (SRM 2975, SRM 1650b), on activation of androgen receptor (AR), estrogen receptor alpha (ER alpha), peroxisome proliferator-activated receptor gamma (PPAR gamma), glucocorticoid receptor (GR) and thyroid receptor alpha (TR alpha), using human cell-based reporter gene assays. Neither DEP standard modulated AR or GR activities. Crude extracts and fractions of SRM 1650b and SRM 2975 suppressed ER alpha-mediated activity in the ER-CALUX (TM) assay; however, this effect could be partly linked to their cytotoxicity in this cell line. We observed that only SRM 2975 extract and its fractions were partial PPAR gamma inducers, while SRM 1650b extract was not active towards this receptor. Importantly, we found that both extracts and polar fractions of SRM activated TR alpha and significantly potentiated the activity of endogenous TR alpha ligand, triiodothyronine. Based on a detailed chemical analysis of both extracts and their polar fractions, we identified several oxygenated PAH derivatives, that were present at relatively high levels in the analyzed DEP standards, including 3-nitrobenzanthrone (3-NBA), anthracene-9,10-dione, phenanthrene-9,10-dione. 9H-fluoren-9-one or benzo[a] anthracene-7,12-dione, to activate TR alpha activity. Nevertheless, these compounds provided only a minor contribution to the overall TR alpha activity identified in polar fractions. This suggests that yet unidentified polar polyaromatic compounds associated with DEP may, apart from their known impact on the aryl hydrocarbon receptor or steroid signaling, deregulate activities of additional nuclear receptors, in particular of TR alpha. The oxidation of a series of alkylaromatic compounds with H2O2 catalyzed by an imine-based non-heme iron complex prepared in situ by reaction of 2-picolylaldehyde, 2-picolylamine, and Fe(OTf)(2) in a 2 : 2 : 1 ratio leads to a marked chemoselectivity for aromatic ring hydroxylation over side-chain oxidation. This selectivity is herein investigated in detail. Side-chain/ring oxygenated product ratio was found to increase upon decreasing the bond dissociation energy (BDE) of the benzylic C-H bond in line with expectation. Evidence for competitive reactions leading either to aromatic hydroxylation via electrophilic aromatic substitution or side-chain oxidation via benzylic hydrogen atom abstraction, promoted by a metal-based oxidant, has been provided by kinetic isotope effect analysis.

The number of documented dengue cases has increased dramatically in recent years due to transmission through theAedes aegyptimosquito bite. Vector control remains the most effective measure to protect against this and other arboviral diseases including Zika, chikungunya and (urban) yellow fever, with an established vaccine only available for yellow fever. Although the quinone class shows potential as leading compounds for larvicide development, limited information restricts the development of optimized structures and/or formulations. Thus, in this contribution we investigated the larvicidal and pupicidal activity of three quinone compounds isolated from aConnarus suberosusroot wood ethyl acetate extract together with 28 quinones from other sources. Eight quinones demonstrated larvicidal activity, of which tectoquinone (4) proved to be the most active (LC(50)1.1 mu g/mL). The essential residual effect parameter of four of these quinones was evaluated in laboratory trials, with tectoquinone (4) and 2-ethylanthraquinone (7) presenting the most prolonged activity. In small-scale field residual tests, tectoquinone (4) caused 100% larvae mortality over 5 days, supporting its selection for formulation trials to develop a prototype larvicide to controlAe. aegypti.

We report herein an efficient method to synthesize triptycenes by the reaction of benzynes and anthranoxides, which are electron-rich and readily prepared from the corresponding anthrones. Using this method, 1,9-syn-substituted triptycenes were regioselectively obtained employing 3-methoxybenzynes. This method was also applied to synthesize pentiptycenes. A DFT study revealed that the cycloaddition of lithium anthranoxide and benzyne proceeds stepwise.

Polycyclic aromatic hydrocarbons are difficult substrates for hydrogenation because of the thermodynamic stability caused by aromaticity. We report here the first chromium- and cobalt-catalyzed, regiocontrolled hydrogenation of polycyclic aromatic hydrocarbons at ambient temperature. These reactions were promoted by low-cost chromium or cobalt salts combined with diimino/carbene ligand and methylmagnesium bromide and are characterized by high regioselectivity and expanded substrate scope that includes tetracene, tetraphene, pentacene, and perylene, which have rarely been reduced. The approach provides a cost-effective catalytic protocol for hydrogenation, is scalable, and can be utilized in the synthesis of tetrabromo- and carboxyl-substituted motifs through functionalization of the hydrogenation product. The systematic theoretical mechanistic modelings suggest that low-valent Cr and Co monohydride species, most likely from zerovalent transition metals, are capable of mediating these hydrogenations of fused PAHs.

In soluble methane monooxygenase enzymes (sMMO), dioxygen (O-2) is activated at a diiron(II) center to form an oxodiiron(IV) intermediate Q that performs the challenging oxidation of methane to methanol. An analogous mechanism of O-2 activation at mono- or dinuclear iron centers is rare in the synthetic chemistry. Herein, we report a mononuclear non-heme iron(II)cyclam complex, 1-trans, that activates O-2 to form the corresponding iron(IV)-oxo complex, 2-trans, via a mechanism reminiscent of the O-2 activation process in sMMO. The conversion of 1-trans to 2-trans proceeds via the intermediate formation of an iron(III)-superoxide species 3, which could be trapped and spectroscopically characterized at -50 degrees C. Surprisingly, 3 is a stronger oxygen atom transfer (OAT) agent than 2-trans; 3 performs OAT to 1-trans or PPh 3 to yield 2-trans quantitatively. Furthermore, 2-trans oxidizes the aromatic C-H bonds of 2,6-di-tert-butylphenol, which, together with the strong OAT ability of 3, represents new domains of oxoiron(IV) and superoxoiron(III) reactivities.

Polycyclic aromatic hydrocarbons (PAHs) are widespread organic contaminants in the environment, much being accumulated in soils. Although their concentrations in large cities have been studied, their levels in small cities were less studied. This study determined the concentrations, distributions, and sources of 16 USEPA priority PAHs (legacy PAHs) and 6 emerging PAHs in urban soils of four small cities. A total of 100 soil samples were collected in Florida, USA. The average Sigma 16-PAHs in urban soils of Clay county, Ocala, Pensacola, and West Palm Beach were 1821, 2748, 3115 and 4055 mu g kg(-1), respectively. Based on benzo[a]pyrene-equivalent (BaP-EQ), the 7 USEPA carcinogenic PAHs (7cPAHs) and 3 emerging carcinogenic PAHs (3cPAHs) in urban soils in Clay County averaged 223 and 3703, Ocala 319 and 4521, Pensacola 302 and 5423, and West Palm Beach 449 and 5916 mu g kg(-1), respectively. Although Sigma 7cPAHs in 87-89% of samples were lower than the Florida Soil Cleanup Target Levels (FSCTLs) for industrial sites at 700 mu g kg(-1), Sigma 3cPAHs were 13-18 times greater than Sigma 7cPAHs. Based on the PMF model and molecular diagnostic ratios, soil PAHs were dominated by similar sources in small cities, mainly from pyrogenic sources including biomass, coal and coke combustion and vehicle emissions. It is important to evaluate both legacy and emerging PAHs concentrations in urban soils when considering soil remediation and human health risk assessment. (C) 2019 Published by Elsevier B.V.

Anthrone tautomerized predominantly, under the reaction conditions, into anthrol and involved in the azo coupling reaction with aryl diazonium chloride to give the corresponding azo dye. UV-Vis experiments have supported the tautomerization of anthrone and the presence of the azo dye products in the hydrazone form. Aryl amines with broad biological applications have been involved to increase the utility of the obtained products. Herein, we disclose a unique directing effect of 9-substituted triptycenes in electrophilic substitution to achieve the regioselective functionalization of the triptycene core. The Hirshfeld population analysis was adopted to predict the selectivity in electrophilic substitution. TMS and t-Bu groups were found to considerably accelerate the reaction at C2 positions to produce C3symmetric isomers. Correlation between distortion and charge distribution within benzene rings was systematically examined.

The approach provides a cost-effective catalytic protocol for hydrogenation, is scalable, and can be utilized in the synthesis of tetrabromo- and carboxyl-substituted motifs through functionalization of the hydrogenation product. The systematic theoretical mechanistic modelings suggest that low-valent Cr and Co monohydride species, most likely from zerovalent transition metals, are capable of mediating these hydrogenations of fused PAHs.
