A set of novel Kv7.2/7.3 (KCNQ2/3) channel blockers was synthesized to address several liabilities of the known compounds XE991 (metabolic instability and CYP inhibition) and the clinical compound DMP 543 (acid instability, insolubility, and lipophilicity). Using the anthrone scaffold of the prior channel blockers, alternative heteroarylmethyl substituents were installed via enolate alkylation reactions. Incorporation of a pyridazine and a fluorinated pyridine gave an analog (compound 18, JDP-107) with a promising combination of potency (IC50=0.16 mu M in a Kv7.2 thallium flux assay), efficacy in a Kv7.2/7.3 patch clamp assay, and drug-like properties.

Ecological wastewater treatment plant (EWWTP), a kind of emerging wastewater treatment plant (WWTP) in recent years, combined microbiology with botany which is efficient for the removal of nitrogen and organic matter, as well as deodorization. The occurrence and removal of micro-organic pollutants in EWWTPs were still not well known. Polycyclic aromatic hydrocarbons (PAHs) and their typical derivatives (SPAHs) including the oxygenated PAHs (OPAHs), chlorinated PAHs (ClPAHs), and methyl PAHs (MPAHs) were investigated in an EWWTP in Guangdong Province, China. The concentrations of the sigma 6 OPAHs (114-384ng/L) were higher than the sigma 16 PAHs (92-250ng/L), and much higher than the sigma 4 MPAHs (13-64ng/L) and sigma 9 ClPAHs (2-3ng/L) in the EWWTP and the effluent receiving river. The total removal efficiencies of the PAHs, OPAHs, MPAHs, and ClPAHs in the EWWTP (43 +/- 14%, 41 +/- 7%, 55 +/- 16%, and 18 +/- 4%) were lower than the traditional WWTPs, probably due to the lower concentration of the sludge in the ecological treatment. The advanced treatment process (microfiltration and UV disinfection treatment) contributed much less (0-20%) to the whole removal efficiency than the ecological treatment (80-100%). The effluent from the EWWTP slightly reduced the PAHs and SPAHs concentrations in the receiving river. The high concentrations of the PAHs and SPAHs in the receiving river were similar to the influent of the EWWTP, indicating that some untreated wastewater was directly discharged to the river, especially in the upstream.

Background: Aloe vera is commonly used in the primary health care of human beings since time immemorial. It is an herb widely used in various traditional systems of medicine worldwide. Systematic and scientific investigation on A. vera as a medicinal plant has drawn considerable attention, and many laboratories are involved in isolation, characterization and evaluation of phytoconstituents for their nutraceutical and pharmaceutical applications. Purpose: The aim of this study was to provide an overview of the phytochemical, biological and medicinal attributes of A. vera against various diseases with special emphasis on underlying mechanisms of action. Methods: PubMed, EBOSCO host, Science Direct, Scopus, and Cochrane library databases were utilized to search literature published between1977 and 2019 (fill March). Major keywords used in various combinations included: Aloe vera, phytochemistry, metabolism, pharmacological activity, prevention, treatment, health, disease, in vivo, in vitro, and clinical studies. Results: Various biological and pharmacological activities of A. vera, such as antioxidant, anti-inflammatory, immuno-modulatory, antimicrobial, antiviral, antidiabetic, hepatoprotective, anticancer, and skin-protective and wound-healing responses, have been attributed to the presence of many active compounds, including anthraquinones, anthrones, chromones, flavonoids, amino acids, lipids, carbohydrates, vitamins and minerals. Conclusion: Based on various preclinical studies, A. vera constituents have enormous potential to prevent and treat various diseases. Randomized clinical trials are needed to understand the full therapeutic potential of this unique medicinal plant.

Biological fouling, also called biofouling, is the undesirable formation of organisms on a surface immersed in water. When the exterior surface of an object such as a vessel is submerged in seawater for a prolonged period, micro and macro organisms usually accumulate on the surface. Biofilms are a set of microorganisms attached to a surface through exopolymers they produce, also known as extracellular polymeric substances (EPS). EPS are a mixture of polymers produced by microorganisms such as bacteria in the presence of biomass. EPS are mainly made up of carbohydrates and proteins, but in smaller quantities, they also contain lipids, nucleic acids, and humic substances. Proteins and carbohydrates found in eps can be quantified by using biochemical methods. The Modified Lowry method, BCA method, and Bradford methods are the common biochemical methods used to quantify protein. Anthrone-sulphuric acid and phenol-sulphuric acid are most common methods used to quantify carbohydrate. The Lowry method was determined to be the best method to quantify protein because it has high precision, low error and low limit of detection. Also, it is highly sensitive and reproducible. Coal fluidity is an important parameter in coal blending techniques for coke making because it strongly influences coke qualities. On the other hand, recently, the amount of high fluidity coal has been limited. To cope with this problem, caking additive method which improves fluidity of coal has been developed and commercialized. However, since tight supply of high fluidity coal is anticipated in the future, it is of great importance to develop more effective caking additive. Therefore, in this study, we investigated effect of 11 kinds of polyaromatic hydrocarbons which include oxygen, sulfur and nitrogen containing compounds on coal fluidity in order to search for more effective chemical substances. The additives were added to low fluidity coal, and fluidity analyses were carried out according to the Gieseler plastometer method. Addition of sulfur and oxygen containing compounds lowered fluidity of coal, whereas addition of aromatic amines enhanced fluidity of coal. Coal fluidity ameliorated with increasing the molecular weight of aromatic amine, and N,N’-di-2-naphthyl-1,4-phenylenediamine (DNPD) was the most effective aromatic amine in this study. Carbonization tests in an electric furnace were conducted to investigate an effect of DNPD on coke strength. As a result of adding only 1 wt% DNPD, fluidity of blended coal and coke strength (Drum Index) were highly improved.

Polycyclic aromatic hydrocarbons are difficult substrates for hydrogenation because of the thermodynamic stability caused by aromaticity. We report here the first chromium- and cobalt-catalyzed, regiocontrolled hydrogenation of polycyclic aromatic hydrocarbons at ambient temperature. These reactions were promoted by low-cost chromium or cobalt salts combined with diimino/carbene ligand and methylmagnesium bromide and are characterized by high regioselectivity and expanded substrate scope that includes tetracene, tetraphene, pentacene, and perylene, which have rarely been reduced. The approach provides a cost-effective catalytic protocol for hydrogenation, is scalable, and can be utilized in the synthesis of tetrabromo- and carboxyl-substituted motifs through functionalization of the hydrogenation product. The systematic theoretical mechanistic modelings suggest that low-valent Cr and Co monohydride species, most likely from zerovalent transition metals, are capable of mediating these hydrogenations of fused PAHs.

Several ethylenedioxy-bridged bisarenes with a variety of type and number of aryl groups were synthesized to study non-covalent dispersion-driven inter- and intramolecular aryl-aryl interactions in the solid state and gas phase. Intramolecular interactions are preferably found in the gas phase. DFT calculations with and without dispersion correction show larger interacting aromatic groups increase the stabilization energy of folded conformers and decrease the intermolecular centroid-centroid distance. Single-molecule structures generally adopt folded conformations with short intramolecular aryl-aryl contacts. Gas electron diffraction experiments were performed exemplarily for 1-(pentafluorophenoxy)-2-(phenoxy)ethane. A new procedure for structure refinement was developed to deal with the conformational complexity of such molecules. The results are an experimental confirmation of the existence of folded conformations of this molecule with short intramolecular aryl-aryl distances in the gas phase. Solid-state structures are dominated by stretched structures without intramolecular aryl-aryl interactions but interactions with neighboring molecules.

Organic pollutants associated with diesel exhaust particles (DEP), such as polycyclic aromatic hydrocarbons (PAHs) and their derivatives, may negatively impact human health. However, a comprehensive overview of their effects on endocrine nuclear receptor activities is still missing. Here, we evaluated the effects of extracts and chromatographic fractions (fractionated according to increasing polarity) of two standard reference materials derived from distinct types of diesel engines (SRM 2975, SRM 1650b), on activation of androgen receptor (AR), estrogen receptor alpha (ER alpha), peroxisome proliferator-activated receptor gamma (PPAR gamma), glucocorticoid receptor (GR) and thyroid receptor alpha (TR alpha), using human cell-based reporter gene assays. Neither DEP standard modulated AR or GR activities. Crude extracts and fractions of SRM 1650b and SRM 2975 suppressed ER alpha-mediated activity in the ER-CALUX (TM) assay; however, this effect could be partly linked to their cytotoxicity in this cell line. We observed that only SRM 2975 extract and its fractions were partial PPAR gamma inducers, while SRM 1650b extract was not active towards this receptor. Importantly, we found that both extracts and polar fractions of SRM activated TR alpha and significantly potentiated the activity of endogenous TR alpha ligand, triiodothyronine. Based on a detailed chemical analysis of both extracts and their polar fractions, we identified several oxygenated PAH derivatives, that were present at relatively high levels in the analyzed DEP standards, including 3-nitrobenzanthrone (3-NBA), anthracene-9,10-dione, phenanthrene-9,10-dione. 9H-fluoren-9-one or benzo[a] anthracene-7,12-dione, to activate TR alpha activity. Nevertheless, these compounds provided only a minor contribution to the overall TR alpha activity identified in polar fractions. This suggests that yet unidentified polar polyaromatic compounds associated with DEP may, apart from their known impact on the aryl hydrocarbon receptor or steroid signaling, deregulate activities of additional nuclear receptors, in particular of TR alpha. Cinchona-based chiral hyperbranched polymers (HBPs) were designed and successfully synthesized via the Mizoroki-Heck (MH) coupling reaction. AB(2) and A(2)B-type chiral monomers were prepared from cinchona squaramide derivatives, where A (vinyl) reacted only with B (iodophenyl) under MH reaction conditions. The chiral HBPs obtained by the MH polymerization contained cinchona squaramide moieties and demonstrated excellent diastereoselectivity and enantioselectivity in asymmetric Michael addition reactions of methyl 2-oxocyclopentanecarboxylate and trans-beta-nitrostyrene.
