BACKGROUND Ara h 1 is a major food allergen in peanuts. Recently, many studies have revealed that the Maillard reaction (MR) affects the allergenicity of food proteins. RESULTS To investigate the influence of the MR on the allergenicity of Ara h 1, R-Ara h 1 was processed with glucose in dry heating conditions for different periods. The extent of the MR was assessed by four methods. The changes in secondary and tertiary structures were characterized through spectroscopy assays. Advanced glycation end products (AGE) structures were identified by protein sample dry heating for 60 min, indicating the formation of AGE-Ara h 1. Simulated gastric fluid (SGF) digestion analysis showed that AGE-Ara h 1 has higher resistance to peptic digestion than R-Ara h 1. The BALB/c mouse model was also utilized to explore the effect of the MR on the allergenicity of Ara h 1, and the results showed that the Th2-type cytokines, antibodies, and histamine content increased, and there was a greater degree of degranulation of rat basophilic leukemia (RBL) cells in the AGE-Ara h 1 group compared with the R-Ara h 1 group. CONCLUSION During the process of dry heating, proteins participated in the MR with changes in secondary and tertiary structures. The condition applying a temperature of 100 degrees C for 60 min caused the formation of AGE-Ara h 1. Simulated gastric fluid digestion analysis showed that AGE-Ara h 1 had a greater resistance to peptic digestion than R-Ara h 1. The BALB/c mouse model showed that AGE-Ara h 1 had more allergenicity, indicating that the MR could enhance the allergenicity of Ara h 1. (c) 2020 Society of Chemical Industry

Laccases produced by Leucoagaricus gongylophorus act in lignocellulose degradation and detoxification processes. Therefore, the use of L. gongylophorus laccase (Lac1Lg) was proposed in this work for degradation of anthracene and others polycyclic aromatic hydrocarbons without the use of mediators. Degradation reactions were performed in buffer aqueous solution with 10 ppm of anthracene and other PAHs, Tween-20 in 0.25% v/v and a laccase preparation of 50 U. The optimum condition (pH 6.0 and 30 degrees C) was determined by response surface methodology with an excellent coefficient of determination (R-2) of 0.97 and an adjusted coefficient of determination (R-adj(2)) of 0.93. In addition, the employment of the mediator ABTS decreased the anthracene biodegradation from 44 +/- 1% to 30 +/- 1%. This optimum pH of 6.0 suggests that the reaction occurs by a hydrogen atom transfer mechanism. Additionally, in 24 h Lac1Lg biodegraded 72 +/- 1% anthracene, 40 +/- 3% fluorene and 25 +/- 3% phenanthrene. The yellow laccase from L. gongylophorus biodegraded anthracene and produced anthrone and anthraquinone, which are interesting compounds for industrial applications. Moreover, this enzyme also biodegraded the PAHs phenanthrene and fluorene justifying the study of Lac1Lg for bioremediation of these compounds in the environment.

Severe acute respiratory syndrome coronavirus (SARS-CoV-2) disease is a global rapidly spreading virus showing very high rates of complications and mortality. Till now, there is no effective specific treatment for the disease. Aloe is a rich source of isolated phytoconstituents that have an enormous range of biological activities. Since there are no available experimental techniques to examine these compounds for antiviral activity against SARS-CoV-2, we employed an in silico approach involving molecular docking, dynamics simulation, and binding free energy calculation using SARS-CoV-2 essential proteins as main protease and spike protein to identify lead compounds from Aloe that may help in novel drug discovery. Results retrieved from docking and molecular dynamics simulation suggested a number of promising inhibitors from Aloe. Root mean square deviation (RMSD) and root mean square fluctuation (RMSF) calculations indicated that compounds 132, 134, and 159 were the best scoring compounds against main protease, while compounds 115, 120, and 131 were the best scoring ones against spike glycoprotein. Compounds 120 and 131 were able to achieve significant stability and binding free energies during molecular dynamics simulation. In addition, the highest scoring compounds were investigated for their pharmacokinetic properties and drug-likeness. The Aloe compounds are promising active phytoconstituents for drug development for SARS-CoV-2.

Voltammetric sensor using a symmetrical derivative of anthrone3 (1,7-diamino-3,9-dibutyl benzo[1,2,3-de:4,5,6-d’e’]diquinoline-2,8(3H,9H)-dione) (SPE-A) has been developed as a probe for Hg(II) ions. Performance of the probe as screen-printed electrode modified with the receptor (SPE-A) has been compared with anthrone3 in solution phase, using 1:1 water-acetonitrile solvent system. Anthrone3 displayed an electrochemically quasi-reversible nature in voltammograms with both the systems and is presented as a novel disposable voltammetric sensor for mercury ions. Upon interaction with cations, both the electrode systems showed sensitivity towards Hg2+ ions with a lower detection limit of 0.61 mu M. The magnitude of the voltammetric current with the SPE-A exhibited three times the current obtained with a bare glassy carbon electrode (GC). Kinetic performance of the SPE-A electrode is better than the GC electrode. The morphological studies indicate reusability of the electrodes.

Both Cr-0 and W-0 alkynyl carbene complexes require tertiary amines to promote the formation of 1,4-C-addition products, but a marked dependence of the reactivity on the metal was observed. Although W-0 complexes form exclusively the 1,4-C-adducts in the presence of a catalytic amount of base, Cr-0 complexes lead to new anthrone-derived bis-carbene complexes, obtained by a sequential 1,4-C- and 1,4-O-addition process with high diastereoselectivity and only in the presence of an excess of base. DFT computational studies show that the gap between the LUMO of the complex and the HOMO of the anthrone determines the differences in reactivity and that the stability of the intermediate metal allenyls formed after the initial 1,4-addition is decisive for the outcome of the reaction.

Some antagonistic bacteria contribute to the management of plant diseases by stimulating the host natural defense and/or by providing direct biocontrol of pathogens. The objective of this study was to evaluate the biocontrol performances and the ability to induce a systemic response to crown gall of four strains belonging to Bacillus and Pseudomonas genera. Evaluation of in vitro antagonistic ability showed that the four bacteria had a different spectrum of activity against the three tested strains of Agrobacterium tumefaciens. A delay in time of appearance and a significant reduction of the tumor size were observed in tomato plants obtained by bio-primed seeds and stem inoculated with A. tumefaciens. Seed priming with antagonistic strains stimulated some systemic defense mechanisms in tomato plants that can be related to the reduction of disease symptoms induced by A. tumefaciens. Both antagonists and A. tumefaciens strains induced variations in phenol content and peroxidase activity while polyphenol oxidase activity was mainly affected by the single A. tumefaciens strain. The response was different in relation to the various combinations of antagonist/pathogen, however Pseudomonas brassicacearum EPR3 strain was the most effective strain.
