The NAD(+)-dependent deacetylase SIRT2 represents an attractive target for drug development. Here, we designed and synthesized drug-like SIRT2-selective inhibitors based on an analysis of the putative binding modes of recently reported SIRT2-selective inhibitors and evaluated their SIRT2-inhibitory activity. This led us to develop a more drug-like diketopiperazine structure as a hydrogen bond (H-bond) hunter to target the substrate-binding site of SIRT2. Thioamide 53, a conjugate of diketopiperazine and 2-anilinobenzamide which is expected to occupy the selectivity pocket of SIRT2, exhibited potent SIRT2-selective inhibition. Inhibition of SIRT2 by 53 was mediated by the formation of a 53-ADP-ribose conjugate, suggesting that 53 is a mechanism-based inhibitor targeting the selectivity pocket, substrate-binding site, and NAD(+)-binding site. Furthermore, 53 showed potent antiproliferative activity toward breast cancer cells and promoted neurite outgrowth of Neuro-2a cells. These findings should pave the way for the discovery of novel therapeutic agents for cancer and neurological disorders.

N-Formamide synthesis usingN-formyl imide with primary and secondary amines with catalytic amounts ofp-toluenesulfonic acid monohydrate (TsOH center dot H2O) is described. Thioamides could be conveniently synthesized in good to excellent yields via DMSO-promoted oxidative coupling of methylhetarenes with amines in the presence of a near stoichiometric amount of sulfur (1.25 equiv.). Both aliphatic and aromatic amines were found to be competent substrates. When anilines o-substituted by cyclizable groups such as OH, NH2, NHPh, SH and CONH2 were used as amine substrates, the corresponding hybrid bis-aza-heterocycles were formed in high yields even with a sulfur loading as low as 0.5 equiv.

A series of 4,5-diphenyl-7H-thieno[2 ‘,3 ‘:3,4]pyrido[2,1-b]quinazolin-7-ones was synthesized via the Rh(iii)-catalyzed annulation of 2-thienylquinazolin-4(3H)-ones with diphenylacetylene. 2-Phenylquinazolin-4(3H)-one amide alcoholysis and double C-H functionalization proceeded under the same reaction conditions with the formation of 2,3,7,8-tetraphenyl-1H-benzo[d,e][1,8]-naphthyridine derivatives. All compounds possess fluorescence properties in MeCN and toluene solutions as well as in the solid state. The aggregation induced emission (AIE) properties and the ability to detect Fe3+ cations were evaluated.

Polyesters are commonly used as food contact materials. During manufacture of polyesters different low molecular mass oligomers (<1000 Da) are formed in the polymer melt. These so-called non-intentionally added substances (NIAS) are potential migrants into foods. In this work, different polyester samples made of polyethylene terephthalate (PET), glycol-modified PET (PETG) and Tritan (TM) were investigated on their qualitative and quantitative oligomer composition. The analysis of acetonitrile extracts by HPLC-DAD/ESI-MS revealed the presence of about 100 linear (different combinations of hydroxyl-, carboxyl-, methyl ester end groups) and cyclic oligomers depending on the main and co-monomers. The identified oligomers were quantified in different extracts and after reprecipitation by HPLC-DAD using bis-hydroxyethylene terephthalate (BHET) as external standard. The amount of oligomers isolated by reprecipitation ranged between 0.80 and 3.4% in the respective polyester. Cyclic oligomers generally made up 90% or more of the isolated oligomers. Compared to the exhaustive extracts the leaching of oligomers into 20% ethanol (1 h, 60 degrees C) resulted in a considerable change of the oligomer distribution with a predominant detection of linear oligomers. This suggests linear oligomers to be relevant for migration into aqueous foods despite the dominant amount of extractable cyclic oligomers in polyesters. Analysis of the extractable oligomers of a PET preform and a PETG container and their corresponding raw material pellets revealed that the injection moulding process did not significantly change the amount of cyclic oligomers but did increase the amount of low molecular mass linear oligomers about twofold. A series of N-benzoylanthranilamide derivatives have been synthesized with the substitution of competitive HB acceptors and investigated by NMR spectroscopy and single crystal XRD. The interesting rivalry for HB acceptance between (sic)C= and X (F or OMe) is observed in the investigated molecules which leads to an unusual increase in the electron density at the site of one of the NH protons, reflecting in the high field resonance in the H-1 NMR spectrum. The NMR experimental findings and single crystal XRD are further reinforced by the DFT studies.

In an attempt to develop potential and selective antitumor agents, a series of novel sulphamide tethered quinazolinone hybrids were efficiently synthesized and evaluated for antitumor activity against four cancer cell lines such as HeLa (cervical), MDA-MB-231 (breast), PANC-1 (pancreatic), and A549 (lung) in vitro. All the compounds (5a-j, 6a-g) exhibited significant anti-proliferative activity with GI(50) values ranging from 0.045 to 6.94 mu M, while compound 10c showed potent activity against all the cell lines (He La, MDA-MB-231, PANC-1 and A549) with GI(50) values ranging from 0.09 to 0.21 mu M. Sialylation can influence the stability, half-life, and immunogenicity of glycoproteins, but sialylated N-glycans are known to be difficult to analyze. Human alpha1-acid glycoprotein (AGP) is reported to have glycans that consist of sialylated N-glycans. The N-glycan profiling of AGP is qualitatively and quantitatively investigated here by UPLC and LC-ESI-MS/MS. Three fluorescent tags (AB, AA, and ProA) and two separation columns (HILIC and AEX-HILIC) were adopted to confirm and compare each analytical characteristic. The results of AA were comparable to those of the well-established AB. The qualification of ProA was notable due to its superior fluorescence intensity and ionization efficiency, and ProA showed smaller quantitative or larger-sized fragments in LC-ESI-MS/MS compared to AB and AA. However, the MS quantification of ProA was distorted because the increased sialylation level decreased the LC-ESI-MS/MS ionization efficiency. HILIC had better peak separability, AEX-HILIC had an advantage in UPLC sialylation profiling, and each isomeric glycan could be Identified by both columns in LC-ESI-MS/MS. In conclusion, ProA is favored for UPLC and LC-ESI-MS/MS detection but not reliable for MS quantification. This study firstly demonstrates the qualification and quantification of sialylated N-glycans by comparing the conunonly used analytical conditions with different fluorescent tags, columns, and instruments.

A benign and straightforward method to access quinazolinones have been developed using easily preparable Cu-3(BTC)(2)MOF as a sustainable solid-Lewis acid catalyst under mild condition. Cu-3(BTC)(2)MOF was prepared and characterized using various analytical tools such as PXRD, FT-IR, SEM, TGA, and ICP-OES. Synthesis of the quinazolinone is catalyzed by the presence of coordinatively unsaturated open Cu(II)sites in Cu-3(BTC)(2)MOF using renewable ethanol as a solvent with minimum copper loading (0.07 mmol) without any harsh reaction condition. A series of substituted quinazolinones were synthesized with good to excellent yields. The efficiency of the present framework copper catalysts was rationalized by comparing with other MOFs and homogeneous catalytic systems. The stability of the catalyst was demonstrated by six consecutive runs and heterogeneity test, which was also evidenced from the technical supports such as PXRD, FT-IR and SEM analyses of the recovered catalyst.
