In this study, novel quinazolinones were designed, synthesized, characterized by FT-IR, H-1-NMR, C-13-NMR spectral data, and LC-MS. New compounds inhibitory activities on urease were assessed. All of the compounds exhibited potent urease inhibitory activities. Especially in the synthesized compounds, 2-benzyl-3-({5-[(4-nitrophenyl)amino]-1,3,4-thiadiazol2-yl}methyl)quinazolin-4(3H)-one has the best inhibitory effect against Jack bean urease with IC50 = 3.30 +/- 0.09 mu g/mL. And also, N-(4-nitrophenyl)-2-[(4-oxoquinazolin-3(4H)-yl)acetyl] hydrazinecarbothioamide, N-(4-fluorophenyl)-2-[(4-oxoquinazolin-3(4H)-yl)acetyl] hydrazinecarbothioamide, and 2-benzyl-3-({5-[(4-fluorophenyl)amino]-1,3,4-thiadiazol-2yl} methyl)quinazolin-4(3H)-one have best activities among the synthesized compounds.

In this study, we describe the synthesis and molecular properties of anthranilamide-based short peptides which were synthesised via ring opening of isatoic anhydride in excellent yields. These short peptides were incorporated as low molecular weight gelators (LMWG), bola amphiphile, and C-3-symmetric molecules to form hydrogels in low concentrations (0.07-0.30% (w/v)). The critical gel concentration (CGC), viscoelastic properties, secondary structure, and fibre morphology of these short peptides were influenced by the aromaticity of the capping group or by the presence of electronegative substituent (namely fluoro) and hydrophobic substituent (such as methyl) in the short peptides. In addition, the hydrogels showed antibacterial activity against S. aureus 38 and moderate toxicity against HEK cells in vitro.

Two photon (TP) fluorescence bioimaging proves to be a powerful tool for biomarker detection and disease diagnosis. However, the probes possessing both good optical properties and biocompatibility are limited. Herein a novel scaffold based on the conjugate of coumarin and quinazolinone (CQ) exhibited brilliant TP fluorescence properties (action cross-section value Phi delta(max) = 299 GM and TP action cross section per molecular weight Phi delta(max)/MW = 0.83) in water. The fluorescence properties are mainly attributed to the enlarged conjugate structure by intramolecular hydrogen bond. The TP fluorescence can be easily switched via amide group modification-deprotection strategy to regulate the formation of hydrogen bond. As a proof of concept, CQ was further engineered with p-nitrobenzyl moiety to afford 2-(7-(diethylamino)-2-oxo-2H-chromen-3-yl)-3-(4-nitrobenzyl) quinazolin-4(3H)-one (NBCQ) for mitochondria nitroreductase (NTR) imaging in normoxia and hypoxia, which proved to be a sensitive and selective off-on NTR fluorescent probe.

With the aim of obtaining new antitumor agents, a series of bis-quinazolin-4(3H)-ones (3a-3 f) were designed and synthesized. These products contain 4-oxo-1,2,3,4-tetrahydro-quinazoline and 3H-quinazolin-4-one moieties linked together via a propyl chain. Cytotoxic activities of 3a-3 f were evaluated against lung adenocarcinoma (A549), breast carcinoma (MCF-7) and ovarian cancer (SKOV3) cell lines using MTT method. Cisplatin was used as a positive control. Among the tested compounds 3a, 3b, and 3e showed the best cytotoxic activities against all cancerous cell lines with IC50 values even less than cisplatin. Compounds 3d and 3f also showed desirable cytotoxic activities especially against A549 and MCF-7.

The NAD(+)-dependent deacetylase SIRT2 represents an attractive target for drug development. Here, we designed and synthesized drug-like SIRT2-selective inhibitors based on an analysis of the putative binding modes of recently reported SIRT2-selective inhibitors and evaluated their SIRT2-inhibitory activity. This led us to develop a more drug-like diketopiperazine structure as a hydrogen bond (H-bond) hunter to target the substrate-binding site of SIRT2. Thioamide 53, a conjugate of diketopiperazine and 2-anilinobenzamide which is expected to occupy the selectivity pocket of SIRT2, exhibited potent SIRT2-selective inhibition. Inhibition of SIRT2 by 53 was mediated by the formation of a 53-ADP-ribose conjugate, suggesting that 53 is a mechanism-based inhibitor targeting the selectivity pocket, substrate-binding site, and NAD(+)-binding site. Furthermore, 53 showed potent antiproliferative activity toward breast cancer cells and promoted neurite outgrowth of Neuro-2a cells. These findings should pave the way for the discovery of novel therapeutic agents for cancer and neurological disorders.

An easy green synthetic approach for fused isoindoloquinazolinones has been developed under neat reaction (yields up to 91%) conditions. This new one-pot tandem methodology involves condensation of readily available anthranilamide with 3-(2-formylcycloalkenyl)-acrylic ester under solvent- and catalyst-free conditions. This strategy avoids the use of oxidant, and heavy metal catalysts and also is free from work-up and generation of toxic by-products. A dramatic change of photophysical properties of dihydroisoindoloquinazolinones in basic and aqueous media has also been documented in our study. Moreover, our model synthetic compound shows cytotoxic activity towards metastatic HepG2 and PC3 cancer cell lines.
