Exploring new material as adsorbent for the efficient enrichment of pollutants is always of great significance in analytical chemistry. In this work, a magnetic azobenzene framework (labeled as MAzo) was constructed as a magnetic solid phase extraction (MSPE) adsorbent by a simple and environmentally benign strategy. The MAzo exhibited the attractive features of strong magnetism, outstanding adsorption performance, as well as good reusability. Combining MAzo-based MSPE with high performance liquid phase chromatography, a simple and effective method was developed for simultaneous determination of four phenylurea herbicides in pear juice and apple juice samples. Under optimized experimental conditions, the method offered low limits of detection of 0.05-0.15 ng mL(-1), high recoveries of 86.7-109.2% with RSD less than 7%. Density functional theory calculation indicated that the good adsorption performance of MAzo for PUHs can be ascribed to the strong H-bonding forces and weak pi-pi interactions. The facile, green, low-cost synthesis method together with the excellent adsorption performance endows the MAzo great application prospect in sample preparation. (c) 2020 Elsevier B.V. The EPHA2 tyrosine kinase receptor is implicated in tumor progression and targeted therapies resistance. We evaluated EPHA2 as a potential resistance marker to the antiepidermal growth factor receptor (EGFR) monoclonal antibody cetuximab in colorectal cancer. We studied activation of EPHA2 in a panel of human colorectal cancer cell lines sensitive or resistant to anti-EGFR drugs. The in vitro and in vivo effects of ALW-II-41-27 (an EPHA2 inhibitor) and/or cetuximab treatment were tested. Fonnalin-fixed paraffin-embedded tumor specimens from 82 RAS wild-type (WV) metastatic colorectal cancer patients treated with FOLFIRI + cetuximab as first-line therapy in the CAPRI-COIM trial were assessed for EPHA2 expression by immunohistochemistry and correlated with treatment efficacy. EPHA2 was differentially activated in colorectal cancer cell lines. Combined treatment with ALW-II-41-27 plus cetuximab reverted primary and acquired resistance to cetuximab, causing cell growth inhibition, inducing apoptosis and cell-cycle G1-G2 arrest. In tumor xenograft models, upon progression to cetuximab, ALW-II-41-27 addition significantly inhibited tumor growth. EPHA2 protein expression was detected in 55 of 82 tumor samples, frequently expressed in less-differentiated and left-sided tumors. High levels of EPHA2 significantly correlated with worse progression-free survival [8.6 months; confidence interval (CI) 95%, 6.4-10.8; vs. 12.3 months; CI 95%, 10.4-14,2; P = 0.03] and with increased progression rate (29% vs. 9%, P = 0.02). A specific EPHA2 inhibitor reverts in vitro and in vivo primary and acquired resistance to anti-ECFR therapy. EPHA2 levels are significantly associated with worse outcome in patients treated with FOLFIRI + cetuximab. These results highlight EPHA2 as a potential therapeutic target in metastatic colorectal cancer.

In recent years, supercritical fluid chromatography (SFC) has become a powerful tool in modern analytical chemistry, and the diversity of stationary phases in SFC promotes phenyl-type phases to confront with a significant resurgence of interest. In this paper, a series of phenyl-type stationary phases with different substituted benzenes involving N-propylbenzamide (PB), 4-fluoro-N-propylbenzamide (PB-F), and 4-ethyl-N-propylbenzamide (PB-ET) were synthesized. Retention mechanism of these phases in SFC was investigated using a linear solvation energy relationship (LSER) model. The phenyl-type stationary phases with all positive parameters can provide all types of interaction, typically involving hydrogen bonding, dipole-dipole and dispersive interactions. The different benzene’s substituents of the stationary phases mainly affected their hydrogen bonding and dipole-dipole interactions, which could be reflected by the angle between the solvation vectors to some extent. The k-k plot showed that the selectivity difference of phenyl-type stationary phases was closely related to the type of solute. Thus, based on twenty-five natural phenolic compounds, two systems with high orthogonality (63.49%) were constructed using three columns, namely phenyl column (PHE) x PB-F and PB x PB-F. Finally, after investigating the influence of chromatographic conditions, ten flavonoids could be separated by using PB, PB-F and PHE columns in SFC. (C) 2019 Published by Elsevier B.V.

This study’s aim was to determine the pesticide residues in 10 different vegetable commodities from the Asir region, Saudi Arabia. We evaluated 211 vegetable samples, collected from supermarkets between March 2018 and September 2018, for a total of 80 different pesticides using ultrahigh-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) and gas chromatography-tandem mass spectrometry (GC-MS/MS) after extraction with a multi-residue method (the QuEChERS method). The results were assessed according to the maximum residue limit (MRL) provided by European regulations for each pesticide in each commodity. All lettuce, cauliflower, and carrot samples were found to be free from pesticide residues. A total of 145 samples (68.7%) contained detectable pesticide residues at or lower than MRLs, and 44 samples (20.9%) contained detectable pesticide residues above MRLs. MRL values were exceeded most often in chili pepper (14 samples) and cucumber (10 samples). Methomyl, imidacloprid, metalaxyl, and cyproconazole were the most frequently detected pesticides. Based on the results of this study, we recommend that a government-supported program for the monitoring of pesticide residues in vegetables be established to promote consumers’ health and achieve sustainable farming systems.

Despite certain limitations, bioaugmentation enhances the efficiency of bioremediation systems. In this study, three aqueous extracts (APE, ACE and APE) from aged residual biomixtures in three biopurification systems (BPSs) exposed to pesticides at a pilot scale were found to improve pesticide removal. The addition of ACEs and AVEs to solutions containing the model compound diuron increased removal rates 6- and 17-fold, respectively, as compared to APEs. These extracts also increased the removal of the metabolite 3,4-dichloroaniline, while AVEs, in particular, were found to remove all pesticides within 9 days. Three metabolites less hazardous than 3,4-dichloroaniline were identified by SPME/GC/MS. AVEs, which also enhance linuron removal in liquid media, were found to increase diuron removal 6-fold in BPSs. We observed an increase in the relative abundance of taxa, such as Chloroflexi, Acidobacteria, Gemmatimonadetes, Firmicutes, Deinococcus-Thermus and especially Proteobacteria (10%), in AV biomixtures, as well as an enrichment of gamma-proteobacteria and the actinobacterial genus Dokdonella in AVEs with respect to initial noncontaminated IV biomixture. We demonstrate that extracts containing a pollutant-acclimatized microbiome could be used as part of a bioaugmentation strategy to improve the functioning of on-farm BPSs and contaminated systems. According to Article 12 of Regulation (EC) No396/2005, EFSA has reviewed the maximum residue levels (MRLs) currently established at European level for the pesticide active substance fluometuron. OBJECTIVE In patients with type 2 diabetes uncontrolled with metformin, exenatide once weekly (QW) plus dapagliflozin produced greater reductions in glycemic parameters (glycated hemoglobin [HbA(1c)], fasting plasma glucose [FPG], and 2-h postprandial glucose [2-h PPG]), weight, and systolic blood pressure (SBP) than exenatide QW or dapagliflozin alone after 28 weeks of treatment in DURATION-8. Following a 24-week extension period, improvements were sustained at 52 weeks. In this study, we investigated efficacy and safety at 104 weeks after randomization. RESEARCH DESIGN AND METHODS DURATION-8 was a 104-week, multicenter, double-blind, randomized, active-controlled, phase 3 trial. In total, 695 adults (aged >= 18 years) with type 2 diabetes and inadequate glycemic control (HbA(1c)8.0-12.0% [64-108 mmol/mol]) despite stable metformin monotherapy (>= 1,500 mg/day) were randomly assigned (1:1:1) to receive exenatide 2 mg QW plus once-daily dapagliflozin 10 mg, exenatide QW plus placebo, or dapagliflozin plus placebo. All 104-week evaluations were exploratory. RESULTS At week 104, 431 (62.0%) patients completed treatment. The adjusted least squares mean change (SE) from baseline to week 104 in HbA(1c)was greater with exenatide QW plus dapagliflozin (-1.70% [0.11]) versus exenatide QW plus placebo (-1.29% [0.12];P= 0.007) and dapagliflozin plus placebo (-1.06% [0.12];P< 0.001). Clinically relevant changes in FPG, 2-h PPG, weight, and SBP were also observed with exenatide QW plus dapagliflozin. There were no unexpected safety findings, and exenatide QW plus dapagliflozin was well tolerated, with no episodes of major hypoglycemia. CONCLUSIONS In this exploratory analysis, among those individuals who completed the trial without rescue therapy, there was clinically relevant efficacy over 2 years with exenatide QW plus dapagliflozin, with no unexpected safety findings. A novel and rapid voltammetric assay for the simultaneous determination of monolinuron (MLN) and linuron (LIN) using an electrochemically pretreated pencil graphite electrode (PGE*) is reported for the first time. Enhanced oxidation peak currents of MLN and LIN were observed at PGE* when compared with electrochemically non-pretreated PGE. Differential pulse voltammetric (DPV) results showed distinct anodic peaks at 1.064 V and 1.170 V for MLN and LIN, respectively. Under optimum conditions, the detection limits were 3.7 x 10(-7) mol L-1 and 5.8 x 10(-7) mol L-1 for MLN and LIN respectively. The DPV method was applied for the quantitative determination of the mentioned compounds in spiked tap water samples enriched by solid phase extraction. The voltammetric results were in good agreement with high-performance liquid chromatography data.

Spain is the first olive oil producer worldwide (representing around 45%), with more than 80% of its agricultural area dedicated to this crop. The goal of this study is to assess the environmental impact of the Spanish virgin olive oil production, attending to the farming and industrial phases. A Life Cycle Assessment (LCA) is conducted for 5 harvests in approximately 4.000 ha of olive grove in Jaen, the largest producer region of Spain. The type of farming is the most representative in Spain: conventional (non-organic), with a medium-low slope, extensive (100-150 trees per ha) and around 60% dryland orchards. The industrial phase is based on 2-phase extraction process and olive pomace valorization. The functional unit (FU) chosen for the comparative analysis is 1 kg of unpacked virgin olive oil under a perspective from cradle to gate. One of the most representative categories, climate change, places the average environmental impact in 2.43 kg CO2 eq/kg, while the range is between 1.93 and 3.00 kg CO2 eq/kg depending on the harvest. Huge differences between values are observed in the farming phase and they are mostly caused by the virgin olive oil yield of every harvest. The carbon sequestration analyses and the impact produced per FU ranges from 43.78% (for 15/16 harvest) to 46.36% (for 17/18). The impact hotspots detected in the farming phase, in terms of climate change potential, are the categories of plant protection products and herbicides (24.11% of the average value) and fertilizers (20.01%). However, the environmental impact of the industrial phase is relatively constant, with a value of 0.547-0.554 kg of CO2 equivalent. Study Objective: To investigate whether the rate of increase in the performance of abdominal myomectomy over a laparoscopic approach after the US Food and Drug Administration (FDA) safety communication regarding morcellator use for myomectomy differs among races. Design: Retrospective cohort study. Setting: The American College of Surgeons National Surgical Quality Improvement data. Patients: Patients aged 18 to 55 years who underwent either laparoscopic or abdominal myomectomy, excluding malignant cases, emergency cases, operations performed by nongynecologic specialists, and cases in which myomectomy was performed during cesarean section. Interventions: None. Measurements and Main Results: The odds ratios of abdominal myomectomy over laparoscopic myomectomy before and after the release of the FDA communication were calculated in 3 race categories: white, African American, and other races. In a logistic regression analysis adjusted for possible confounders, including all races, the odds ratio of abdominal myomectomy before and after the FDA communication was 1.30 (95% confidence interval [CI], 1.20-1.41; p <. 001). In a logistic regression analysis with a product term of FDA communication exposure and race as a possible effect modifier, the African American population showed a significantly greater change in the odds of abdominal myomectomy over laparoscopic myomectomy in comparison with the white population (1.22; 95% CI, 1.02-1.47; p =.03). In contrast, other races showed no significant change (.83; 95% CI,.64-1.08; p =.17). Conclusion: After the FDA communication, the odds ratio of abdominal myomectomy was disproportionately increased in the African American population.
