A new series of 1,2,4-triazole was designed, synthesized, and characterized as remarkable antimicrobial and antioxidant agents. These heterocycles have been prepared from the cyclization reactions of Schiff bases 3(a-k) with phenylhydrazine by refluxing under the alkaline medium. The Schiff bases in turn were realized in good yields from the condensation reactions of N-phenylurea with different aromatic aldehydes. The structures of the intermediates 3(a-k) and final heterocycles 4(a-k) have been fully characterized through their spectral parameters.

Spain is the first olive oil producer worldwide (representing around 45%), with more than 80% of its agricultural area dedicated to this crop. The goal of this study is to assess the environmental impact of the Spanish virgin olive oil production, attending to the farming and industrial phases. A Life Cycle Assessment (LCA) is conducted for 5 harvests in approximately 4.000 ha of olive grove in Jaen, the largest producer region of Spain. The type of farming is the most representative in Spain: conventional (non-organic), with a medium-low slope, extensive (100-150 trees per ha) and around 60% dryland orchards. The industrial phase is based on 2-phase extraction process and olive pomace valorization. The functional unit (FU) chosen for the comparative analysis is 1 kg of unpacked virgin olive oil under a perspective from cradle to gate. One of the most representative categories, climate change, places the average environmental impact in 2.43 kg CO2 eq/kg, while the range is between 1.93 and 3.00 kg CO2 eq/kg depending on the harvest. Huge differences between values are observed in the farming phase and they are mostly caused by the virgin olive oil yield of every harvest. The carbon sequestration analyses and the impact produced per FU ranges from 43.78% (for 15/16 harvest) to 46.36% (for 17/18). The impact hotspots detected in the farming phase, in terms of climate change potential, are the categories of plant protection products and herbicides (24.11% of the average value) and fertilizers (20.01%). However, the environmental impact of the industrial phase is relatively constant, with a value of 0.547-0.554 kg of CO2 equivalent. PRMT4 is a type I protein arginine methyltransferase and plays important roles in various cellular processes. Overexpression of PRMT4 has been found to be involved in several types of cancers. Selective and in vivo effective PRMT4 inhibitors are needed for demonstrating PRMT4 as a promising therapeutic target. On the basis of compound 6, a weak dual PRMT4/6 inhibitor, we constructed a tetrahydroisoquinoline scaffold through a cut-and-sew scaffold hopping strategy. The subsequent SAR optimization efforts employed structure-based approach led to the identification of a novel PRMT4 inhibitor 49. Compound 49 exhibited prominently high potency and selectivity, moderate pharmacokinetic profiles, and good antitumor efficacy in acute myeloid leukemia xenograft model via oral administration, thus demonstrating this compound as a useful pharmacological tool for further target validation and drug development in cancer therapy.

The abiotic photochemical reactions that take place naturally in sunlit surface waters can degrade many contaminants that pose concern to water bodies for their potentially toxic and long-term effects. This works aims at assessing the ability of European rivers to photoproduce reactive transient intermediates, such as HO center dot radicals and the excited triplet states of chromophoric dissolved organic matter ((CDOM)-C-3*), involved in pollutant degradation. A photochemical mapping of the steady-state concentrations of these transients was carried out by means of a suitable modeling tool, in the latitude belt between 40 and 50 degrees N. Such a map allowed for the prediction of the photochemical lifetimes of the phenylurea herbicide isoproturon (mostly undergoing photodegradation upon reaction with HO center dot and especially (CDOM)-C-3*) across different European countries. For some rivers, a more extensive dataset was available spanning the years 1990-2002, which allowed for the computation of the steady-state concentration of the carbonate radicals (CO3 center dot-). With these data, it was possible to assess the time trends of the photochemical half-lives of further contaminants (atrazine, ibuprofen, carbamazepine, and clofibric acid). The calculated lifetimes were in the range of days to weeks, which might or might not allow for efficient depollution depending on the river-water flow velocity.

This study evaluated the remobilization, mineralization, and metabolism of herbicide-bound residues in soils amended with various types of sugarcane waste. Soil with bound residues of three herbicides and fresh soil samples were added to the biometric flasks, followed by the addition of vinasse, filter cake, or sugarcane straw in order to reactivate the microbial activity. In sandy loam soil, higher mineralization was observed where filter cake was added, and the maximum mineralized percentage was 7.7, 46.7, and 8.1% of diuron-, hexazinone-, and metribuzin-bound residues, respectively. Conversely, this soil presented a greater percentage of re-extractable hexazinone and metribuzin residues when vinasse was added. Among the examined herbicides, a higher percentage of bound metribuzin residues remained in the soil (57.5-75.6%). It was possible to identify both metabolites and parent compounds in the re-extracted residues, implying either species could bind to the soil. Therefore, this study has shown that bound residues of three herbicides and their metabolites can become bioavailable, and mineralized or returned to the soil solution, which could adversely affect subsequent crops or non-target organisms. Hence, the remobilization of bound residues must be taken into account when assessing the environmental risk of herbicides in soils in registration processes.

Despite certain limitations, bioaugmentation enhances the efficiency of bioremediation systems. In this study, three aqueous extracts (APE, ACE and APE) from aged residual biomixtures in three biopurification systems (BPSs) exposed to pesticides at a pilot scale were found to improve pesticide removal. The addition of ACEs and AVEs to solutions containing the model compound diuron increased removal rates 6- and 17-fold, respectively, as compared to APEs. These extracts also increased the removal of the metabolite 3,4-dichloroaniline, while AVEs, in particular, were found to remove all pesticides within 9 days. Three metabolites less hazardous than 3,4-dichloroaniline were identified by SPME/GC/MS. AVEs, which also enhance linuron removal in liquid media, were found to increase diuron removal 6-fold in BPSs. We observed an increase in the relative abundance of taxa, such as Chloroflexi, Acidobacteria, Gemmatimonadetes, Firmicutes, Deinococcus-Thermus and especially Proteobacteria (10%), in AV biomixtures, as well as an enrichment of gamma-proteobacteria and the actinobacterial genus Dokdonella in AVEs with respect to initial noncontaminated IV biomixture. We demonstrate that extracts containing a pollutant-acclimatized microbiome could be used as part of a bioaugmentation strategy to improve the functioning of on-farm BPSs and contaminated systems. Background: Nivolumab was the first immune checkpoint inhibitor approved for previously treated advanced nonesmall cell lung cancer (NSCLC). Before its introduction in the market, nivolumab was made available to NSCLC patients through an expanded access program (EAP). Here we present the Italian cohort of patients with non-squamous NSCLC enrolled in a worldwide nivolumab EAP, with subgroup analyses involving elderly patients, patients with central nervous system (CNS) metastases and patients receiving nivolumab beyond progression. Methods: Pretreated patients with advanced non-squamous NSCLC received nivolumab at 3 mg/kg every 2 weeks up to 24 months. Efficacy data (investigator-assessed tumour response, progression date and survival) and safety data were collected. Findings: 1588 patients were treated across 153 Italian centres. Overall response rate and disease control rate were 18% and 44%, respectively; median overall survival (OS) was 11.3 months (95% CI: 10.2-12.4). Elderly patients (>= 70 n = 522; >= 75 n = 232) achieved outcomes similar to the global study population; patients with CNS metastases (n = 409) had an OS of 8.6 months (95% CI: 6.4-10.8), and a 1-year OS rate of 43%. Nivolumab was administered beyond progression to 276 patients (26%), 57 of whom achieved subsequent disease control; the median OS of patients receiving nivolumab beyond progression was 16.2 months (95% CI: 14.0-18.4), while 1-year OS rate was 62%. Interpretation: To date, this is the largest clinical experience with nivolumab in a real-world setting.
