Background Non-neoplastic anal sac disorders (ASD) are frequent presentations for dogs in primary-care practice but evidence-based information on disease occurrence and risk is sparse. This study estimates prevalence, breed associations and other risk factors as well as reporting on clinical management. Methods A cohort study of dogs attending VetCompass practices between 1 January 2013 and 31 December 2013. Risk factor analysis used multivariable logistic regression methods. Results Of 104,212 dogs attending 110 UK practices, the 1-year period prevalence of ASD was 4.40% (95% CI: 4.22-4.57). Compared to crossbreds, six breeds showed increased odds of ASD (Cavalier King Charles spaniel, King Charles spaniel, Cockapoo, Shih-tzu, Bichon Frise and Cocker spaniel), and six breeds showed reduced odds (Labrador Retriever, Border collie, Staffordshire Bull Terrier, Lurcher, German Shepherd Dog and Boxer). Brachycephalic types had 2.6 times the odds for ASD compared to dolichocephalic types. Medication prescribed for ASD included antimicrobials (n = 480, 20.24%) and analgesics (n = 284, 11.97%). Anal sacculectomy was performed in under 1% of cases. Conclusions High prevalence, strong breed predispositions and evidence of severity suggested from the antimicrobial and analgesic therapies combined with current substantial knowledge gaps identify ASD as a key research-neglected syndrome in dogs.

Development of a low-cost, environmentally benign and robust catalyst for multipurpose industrially relevant organic transformations is highly desirable for the sustainable future of chemical industries. A hybrid cobalt doped-cerium oxide nanocatalyst (Co@CeO2NC) was prepared via simple coprecipitation method using water as the solvent. The characterization of Co@CeO2NC was performed using different techniques such as XRD, TGA, FE-SEM, HR-TEM, EDAX-mapping, BET, and XPS analysis. The structural characterization of the prepared sample by XRD and XPS analysis revealed the presence of the mixed phase of cobalt oxide and cobalt doped-cerium oxide as a hybrid (Co@CeO2NC). Industrially relevant organic transformations such as (i) nitrile formation using aldehyde with hydroxyl amine hydrochloride, (ii) reductive amination of aldehydes to form tertiary N,N-dimethyl amines, and (iii) direct acetylation of alcohols/amines with acetic acid were achieved in an excellent manner using Co@CeO2NC hybrid as the multifunctional catalyst. Excellent catalytic activity of Co@CeO2NC was noticed for the conversion of 4-chlorobenzaldehyde to 4-chlorobenzonitrile with 99% conversion and 99% selectivity and 100% conversion of benzaldehyde to N,N-dimethylbenzylamine using DMF as NMe2 source, reductant, and solvent. Moreover, acetylation of 4-methoxybenzyl alcohol and 2-methyl aniline gave excellent conversion and selectivity toward the formation of -O and -N acetyl. The scope of the Co@CeO2NC was further evaluated for other aldehydes, alcohols, and amines with an excellent conversion and high selectivity.

Supercritical methanol depolymerization and hydrodeoxygenation (SCM-DHDO) of maple wood and lignin extracted with GVL from maple wood, was studied using a copper porous metal oxide catalyst. Phenols (P or H), guaiacols (G), and syringols (S) with deoxygenated 1 to 3-carbon (C-1-C-3) alkyl tails were the early products (15 min reaction time) from both the maple wood and the GVL extracted lignin. Furthermore, at 15 min reaction time, the maple wood products showed increased levels of demethoxylation with an S/G/P ratio of 1.0/4.2/1.2 versus 1.0/1.0/0.1 from the GVL extracted lignin products. After 4 h of reacting lignin monomers, dimers and trimers were produced with both the maple wood and extracted lignin. However, the 4 h maple wood products yielded twice the amount of lignin monomers (43.1%) as compared to the 4 h GVL extracted lignin products (20.1%). The GVL extracted lignin products were in the dimer to trimer molecular weight (MW, similar to 180-750 Da) ranges whereas the maple wood products were in the monomer to dimer MW ranges (similar to 94-500 Da) according to the GPC results. NMR and FT-ICR MS analyses show that both the 4 h maple wood products and 4 h GVL extracted lignin products undergo a high degree of hydrodeoxygenation, saturation, and repolymerization by C-C bond formation. The higher yield of lignin monomers from the maple wood is likely due to the lower lignin concentration that occurs during the reaction whereas higher lignin concentration during the reaction leads to more oligomerization reactions.

Nucleophilic aromatic substitution (SNAr) is routinely used to install (19)F(-)and (18)F(-)in aromatic molecules, but is typically limited to electron-deficient arenes due to kinetic barriers associated with C-F bond formation. Here we demonstrate that a polarity-reversed photoredox-catalysed arene deoxyfluorination that operates via cation-radical-accelerated SNAr enables the fluorination of electron-rich arenes with (19)F(-)and (18)F(-)under mild conditions, and thus complements the traditional arene polarity requirements necessary for SNAr-based fluorination. The utility of our radiofluorination strategy is highlighted by short reaction times, compatibility with multiple nucleofuges and high radiofluorination yields, especially that of an important cancer positron emission tomography agent [F-18]5-fluorouracil. Taken together, our fluorination approach enables the development of fluorinated and radiofluorinated compounds that can be difficult to access by classical SNAr strategies, with the potential for use in the synthesis and discovery of positron emission tomography radiopharmaceuticals.

Background: Colorectal familial adenomatous polyposis (FAP) adenomas exhibit a uniform pathogenetic basis caused by a germline mutation in the adenomatous polyposis gene (APC), but the molecular changes leading to their development are incompletely understood. However, dysregulated apoptosis is known to substantially affect the development of colonic adenomas. One of the key regulatory proteins involved in apoptosis is apoptosis repressor with caspase recruitment domain (ARC). Methods: The expression of nuclear and cytoplasmic ARC in 212 adenomas from 80 patients was analyzed by immunohistochemistry. We also compared expression levels of ARC with the expression levels of p53, Bcl-2, COX-2, and MMR proteins. Statistical analyses were performed by Spearman’s rank correlation and linear regression test. Results: ARC was overexpressed in the nuclei and cytoplasm of most FAP adenomas investigated. Cytoplasmic ARC staining was moderately stronger (score 2) in 49.1% (n = 104/212) and substantially stronger (score 3) in 32.5% (n = 69/212) of adenomas compared to non-tumorous colorectal mucosa. In 18.4% (n = 39/212) of adenomas, cytoplasmic ARC staining was equivalent to that in non-tumorous mucosa. Nuclear expression of ARC in over 75% of cells was present in 30.7% (n = 65/212) of investigated adenomas, and nuclear expression in 10-75% of cells was detected in 62.7% (n = 133/212). ARC expression in under 10% of nuclei was found in 6.6% (n = 14/212) of adenomas. The correlation between nuclear ARC expression and cytoplasmic ARC expression was highly significant (p = 0.001). Moreover, nuclear ARC expression correlated positively with overexpression of Bcl-2, COX-2 p53 and beta-catenin. Cytoplasmic ARC also correlated with overexpression of Bcl-2. Sporadic MMR deficiency was detected in very few FAP adenomas and showed no correlation with nuclear or cytoplasmic ARC. Conclusions: Our results demonstrated that both cytoplasmic and nuclear ARC are overexpressed in FAP adenomas, thus in a homogenous collective. The highly significant correlation between nuclear ARC and nuclear beta-catenin suggested that ARC might be regulated by beta-catenin in FAP adenomas. Because of its further correlations with p53, Bcl-2, and COX-2, nuclear ARC might play a substantial role not only in carcinomas but also in precursor lesions.

Recent studies have raised concerns about e-cigarette liquid inhalation toxicity by reporting the presence of chemicals with European Union CLP toxicity classification. In this scenario, the regulatory context is still developing and is not yet up to date with vaping current reality. Due to the paucity of toxicological studies, robust data regarding which components in e-liquids exhibit potential toxicities, are still inconsistent. In this study we applied computational methods for estimating the toxicity of poorly studied chemicals as a useful tool for predicting the acute toxicity of chemicals contained in e-liquids. The purpose of this study was 3-fold: (a) to provide a lower tier assessment of the potential health concerns associated with e-liquid ingredients, (b) to prioritize e-liquid ingredients by calculating the e-tox index, and (c) to estimate acute toxicity of e-liquid mixtures. QSAR models were generated using QSARINS software to fill the acute toxicity data gap of 264 e-liquid ingredients. As a second step, the potential acute toxicity of e-liquids mixtures was evaluated. Our preliminary data suggest that a computational approach may serve as a roadmap to enable regulatory bodies to better regulate e-liquid composition and to contribute to consumer health protection.

Recent studies have raised concerns about e-cigarette liquid inhalation toxicity by reporting the presence of chemicals with European Union CLP toxicity classification. In this scenario, the regulatory context is still developing and is not yet up to date with vaping current reality. Due to the paucity of toxicological studies, robust data regarding which components in e-liquids exhibit potential toxicities, are still inconsistent. In this study we applied computational methods for estimating the toxicity of poorly studied chemicals as a useful tool for predicting the acute toxicity of chemicals contained in e-liquids. The purpose of this study was 3-fold: (a) to provide a lower tier assessment of the potential health concerns associated with e-liquid ingredients, (b) to prioritize e-liquid ingredients by calculating the e-tox index, and (c) to estimate acute toxicity of e-liquid mixtures. QSAR models were generated using QSARINS software to fill the acute toxicity data gap of 264 e-liquid ingredients. As a second step, the potential acute toxicity of e-liquids mixtures was evaluated. Our preliminary data suggest that a computational approach may serve as a roadmap to enable regulatory bodies to better regulate e-liquid composition and to contribute to consumer health protection.

Polydopamine-coated polyurethane open cell foams are used as structured supports for molecular catalysts through the covalent anchoring of alkoxysilyl arms by the catechol groups of the mussel-inspired layer. This strong bonding prevents their leaching. No alteration of the mechanical properties of the flexible support is observed after repeated uses of the catalytic materials.
