With the growing concern about safety and environmental issues, amino acid-based surfactants have great potential as sustainable and eco-friendly substances. In the present study, six N-acyl oligopeptide surfactants were synthesized using fatty acid methyl esters (i.e., methyl caprate, methyl laurate, and methyl myristate) and glycyl oligopeptides (i.e., glycylglycine and glycylglycylglycine). Surface active properties and aggregation behaviors of these surfactants were investigated using surface tension, fluorescence probe, dynamic light scattering, and microscopy measurements. The adsorption at the air-water interface and aggregation in the aqueous solution depend on the structure of both hydrophilic head-groups and hydrophobic tails. Spherical vesicles were observed in the aqueous solutions of glycylglycine derivatives, whereas short tubular aggregates were found in that of corresponding glycylglycylglycine products. Intermolecular hydrogen-bonding interaction between the amide head-groups might be responsible for the different self-assembly behavior and microstructure of N-acyl oligopeptide surfactants in the aqueous solution.

12-Deoxyphorbol-13-phenylacetate (DPP), found in Euphorbia resinifera, is a prototype of a new class of phorbol derivatives that function as protein kinase C activators with potent anti-tumor-promoting activity. To better understand the biosynthesis of DPP in E. resinifera and develop methods for its production, we used next-generation sequencing technologies to build a transcriptome dataset. We obtained a total of approximately 27,102,078 clean reads that were assembled into 90,448 unigenes. Of those, 129 and 59 unigenes were identified in the Terpenoid Backbone Biosynthesis (TBB) (KEGG map00900) and the Diterpenoid Biosynthesis (DB) (KEGG map00904) pathways, respectively. We investigated the expression patterns of 23 DPP biosynthesis-related genes and validated the results by quantitative real time PCR. The results showed that CMK (4-diphosphocytidyl-2-C-methyl-o-erythritol kinase), MECS (2-C-methyl-D-erythritol 2,4-cyclodiphosphate synthase), HDS (1-hydroxy-2-methyl-2-(E)-butenyl 4-diphosphate synthase), and IPI (Isopentenyl-diphosphate Delta-isomerase) of the methylerythritol 4-phosphate (MEP) pathway had a higher expression compared to other genes of TBB, whereas the expression levels of genes involved in the mevalonic acid (MVA) pathway were relatively lower and not significantly different from each other. Furthermore, the expression of the 23 genes in root, stem and young stem differed, and the statistical results indicated that HMGS (Hydroxymethylglutaryl-CoA synthase), MVK (mevalonic acid), MECT (2-C-methyl-D-erythritol 4-phosphate cytidylyltransferase), MECS, HDS and IPI in the TBB pathway as well as KS in the DB pathway were significantly and positively correlated with the CS (casbene synthase) gene. These results suggested that both cytosol-localized MVA and plastid-localized MEP pathways may have a significant contribution in DPP accumulation, with MEP pathway playing a dominant role. To the best of our knowledge, this study is the first transcriptome analysis of E. resinifera and the results reported here will aid the identification of functional genes and better understanding of DPP biosynthesis.

The rapid and effective removal of organic micropollutants (OMPs) from water remains a huge challenge for traditional water treatment techniques. Compared with powder adsorbents such as polymers and nanomaterials, the free-standing adsorptive membrane is possible for large-scale applications and shows promise in removing OMPs. Herein, inspired by aquatic plants, a novel free-standing adsorptive membrane (NPPM) with high water flux, strong adsorption affinity, and excellent reproducibility was prepared by one-step UV surface grafting. N-Vinylformamide (NVF) was employed to introduce multiple hydrophilic and hydrogen bonding sites on the surface of commercial polypropylene fiber membranes (PPM). The NPPM exhibits excellent water permeability and ultrahigh water flux (up to 40 000 L/(m(2) h)) and could continuously remove a broad spectrum of OMPs from water. Its adsorption performance is 5-100 times higher than that of PPM and commercial membranes. Even in natural water sources such as tap water and river water, the NPPM shows unchanged adsorption performance and high OMPs removal efficiency (>95%). Notably, the NPPM has excellent regeneration performance and can be regenerated by hot water elution, which provides an environmentally friendly regeneration method without involving any organic solvent. Moreover, the synergy between hydrogen bonding and hydrophobic interaction is revealed, and the hydrophobic interaction provided by the hydrophobic substrate is proved to play a fundamental role in OMPs adsorption. The strong hydrogen bonds between the grafts and the OMPs are demonstrated by variable-temperature FTIR spectroscopy (vt-FTIR), C-13 nuclear magnetic resonance spectroscopy (C-13 NMR), and simulation calculations. The strong hydrogen bonds could increase the enthalpy change and enhance the adsorption affinity, so the NPPM has a strong adsorption affinity, which is 100 times that of similar adsorption membranes. This study not only presents an adsorptive membrane with great commercial potential in the rapid remediation of a water source but also opens a pathway to develop an adsorptive membrane with high water flux and strong adsorption affinity.

Magnetic porous microspheres composed of Fe3C, MnO and graphitic carbon (Fe3C/MnO/GC) were prepared by a one-pot method. A polycondensate obtained from urea and formaldehyde served as the carbon source and was calcined in the presence of metallic iron and manganese to yield Fe3C/MnO/GC. The resultant hybrid has highly mesoporous architecture, large specific surface, graphitic structure and adequate saturation magnetism. This study explores the synthesis of cyclic cis-vicinal phenyl ethylenes from oxotriphenylhexanoates. The reaction is a BBr3-promoted cyclization of 1,6-ketoesters (1) to five-membered diketo compounds (2). A simple, efficient, and mild method for the preparation of aryl-fused heterocycles has been developed using alpha,omega-alkynoic acids and arylethylamines. In the present method, one of Grubbs’ ruthenium carbenes is able to catalyze the formation of exocyclic enol lactones from alpha,omega-alkynoic acids, which subsequently undergo aminolysis and N-acyl iminium ion formation/cyclization with arylethylamines in the presence of TFA, affording one kind of drug-privileged scaffold. Hydrogen sulfide (H2S) is a biologically important gaseous molecule that exhibits promising protective effects against a variety of pathological processes. For example, it was recognized as a blood pressure lowering agent. Aligned with the need for easily modifiable platforms for the H2S supply, we report here the preparation and the H2S release kinetics from a series of structurally diversified thioamides, thiolactams and thioureas. Three different thionation methods based on the usage of a phosphorus pentasulfide and Lawesson reagent were applied to prepare the target thioamides and thiolactams. Furthermore, obtained H2S donors were evaluated both in in vivo and in vitro studies. The kinetic parameters of the liberating H2S was determined and compared with NaHS and GYY4137 using two different detection technics i.e.; fluorescence labeling 7-azido-4-methyl-2H-chromen-2-one and 5,5′-dithiobis (2-nitrobenzoic acid), sulfhydryl probe, also known as the Ellman’s reagent. We have proved that the amount of releasing H2S from these compounds is controllable through structural modifications. Finally, the present study shows a hypotensive response to an intravenous administration of the developed donors in the anesthetized rats.

During the past few decades, fabrication of functional rotaxane-branched dendrimers has become one of the most attractive yet challenging topics within supramolecular chemistry and materials science. Herein, we present the successful fabrication of a family of new rotaxane-branched dendrimers containing up to 21 platinum atoms and 42 photosensitizer moieties through an efficient and controllable divergent approach. Notably, the photosensitization efficiencies of these rotaxane-branched dendrimers gradually increased with the increase of dendrimer generation. For example, third-generation rotaxane-branched dendrimer PG3 revealed 13.3-fold higher O-1(2) generation efficiency than its corresponding monomer AN. The enhanced O-1(2) generation efficiency was attributed to the enhancement of intersystem crossing (ISC) through the simple and efficient incorporation of multiple heavy atoms and photosensitizer moieties on the axles and wheels of the rotaxane units, respectively, which has been validated by UV-visible and fluorescence techniques, time-dependent density functional theory calculations, photolysis model reactions, and apparent activation energy calculations. Therefore, we develop a new promising platform of rotaxane-branched dendrimers for the preparation of effective photosensitizers.
