Background: Due to the presence of several serious side effects such as erosion and bleeding in the gastric mucosal layer which are linked with Non-steroidal anti-inflammatory agents and a lot of these adverse effects may be dangerous for life and may cause numerous mortalities, demand for new antiinflammatory and analgesic agents has become an important task in medicinal chemistry to improve therapeutic efficacy as well as safety. Method: In the search for new anti-inflammatory and analgesic molecules with greater potency, some novel indole derivatives (SK-1 to SK-9) were attained by chloroacetylation process of indole after that reaction through some aromatic aldehydes with the presence of acetic acid in absolute ethyl alcohol. IR, H-1 NMR, and C-13 NMR spectra were used for the structure elucidation of obtained chemical compounds. In this context, the analgesic and anti-inflammatory activity were evaluated for synthetically obtained new chemical compounds. Furthermore, the molecular docking studies were performed to elucidate the binding modes of synthesized ligands in the active pockets of Cox-1 / 2 enzymes. Result: Among the compounds assessed, SK-2 and SK-8 demonstrated potent analgesic & antiinflammatory activities when compared to ibuprofen and diclofenac sodium as reference drugs. Compound SK-4 has exhibited significant anti-inflammatory activity while compounds SK-1 and SK-5 have shown significant analgesic action when compared to the control. Molecular docking studies indicated that SK2 compound exhibited appreciable binding affinity (-9.16 kcal/mol) towards the COX-2 enzyme and mandatory interactions for COX-2 enzyme inhibition. Conclusion: It might be established from the present study that prepared compounds are unique in terms of their structure and noticeable biological action. Purpose Wound healing after Ahmed glaucoma valve (AGV) implantation often entails fibrosis as a foreign body reaction to the silicone plate. Poly(2-methacryloyloxyethyl phosphorylcholine) (PMPC) forms an antifouling surface that inhibits fibrosis during wound healing. In this study, we aimed to compare the effects of the implantation of AGV coated with PMPC (wPMPC) versus AGV without PMPC (woPMPC) in rabbits. Methods Six New Zealand White rabbit does underwent AGV implantation in both eyes. For each rabbit, one eye was randomly selected for implantation of AGV wPMPC and a conventional AGV (woPMPC) was implanted in the contralateral eye. Gross conjunctival vascularity was compared between the two groups at the first, second, and fourth weeks after surgery. The eyes were enucleated in four weeks and subjected to staining with hematoxylin and eosin and Masson’s trichrome stain. The fibrosis and inflammation status among the eye samples were compared by measuring the thickness of the fibrotic walls and counting the number of chronic inflammatory cells around the AGV. Counting of inflammatory cells and measuring fibrotic wall thickness were done in a blinded method to eliminate observer bias. Statistical analysis was performed using the Mann-Whitney U test. Results Gross and histological examinations revealed no toxic effects of PMPC. There were no apparent differences in overall conjunctival vascularity between the two groups at weeks 1, 2, and 4 after surgery. The average inflammatory cell counts were 14.3 5.8 per slide and 27.3 +/- 8.6 per slide in the wPMPC and woPMPC groups, respectively (p = 0.037). The average thicknesses of the fibrotic wall were 57.9 +/- 11.3 mu m and 81.5 +/- 21.3 mu m in the wPMPC and woPMPC groups, respectively (p = 0.025). Conclusion Compared to the woPMPC group, the number of inflammatory cells and fibrosis were significantly decreased in the wPMPC group.

The analysis of complex oligosaccharides is traditionally based on multidimensional workflows where liquid chromatography is coupled to tandem mass spectrometry (LC-MS/MS). Due to the presence of multiple isomers, which cannot be distinguished easily using tandem MS, a detailed structural elucidation is still challenging in many cases. Recently, ion mobility spectrometry (IMS) showed great potential as an additional structural parameter in glycan analysis. While the time-scale of the IMS separation is fully compatible to that of LC-MS-based workflows, there are very few reports in which both techniques have been directly coupled for glycan analysis. As a result, there is little knowledge on how the derivatization with fluorescent labels as common in glycan LC-MS affects the mobility and, as a result, the selectivity of IMS separations. Here, we address this problem by systematically analyzing six isomeric glycans derivatized with the most common fluorescent tags using ion mobility spectrometry. We report >150 collision cross-sections (CCS) acquired in positive and negative ion mode and compare the quality of the separation for each derivatization strategy. Our results show that isomer separation strongly depends on the chosen label, as well as on the type of adduct ion. In some cases, fluorescent labels significantly enhance peak-to-peak resolution which can help to distinguish isomeric species.

A convenient protocol for the one-pot synthesis of quinazolin-4(3H)-ones using palladium(II) complexes via dehydrogenative coupling of readily available benzyl alcohols and 2-aminobenzamide has been described. New structurally related Pd(II) N boolean AND O chelating complexes of general configuration [Pd(L)Cl(PPh3)] (where L = dimethylamino benzoylhydrazone ligands) have been designed and synthesized. The formation of the complexes has been recognized by analytical and spectral methods (FT-IR, NMR, HR-MS). The presence of a square-planar geometry around the palladium(II) ion was confirmed by single crystal X-ray diffraction study. A wide range of substituted quinazolinones have been successfully achieved from a diverse range of benzyl alcohols in good to excellent yields using 1.0 mol % of catalyst loading under aerobic conditions. Furthermore, control experiments reveal that the dehydrogenative coupling reaction involves initially the formation of an aldehyde intermediate and subsequent formation of a cyclic aminal intermediate.

BACKGROUND: The objective of this study was to assess the relationship between inflammation-based scores and prognosis of patients who had undergone mechanical thrombectomy (MT) for large artery occlusion. METHODS: A total of 411 patients were enrolled and inflammation-based scores, such as neutrophil/lymphocyte ratio (NLR), lymphocyte/monocyte ratio (LMR), and monocyte/high-density lipoprotein cholesterol ratio (MHR) were calculated based on laboratory data. Prognoses were evaluated with unfavorable outcome (modified Rankin Scale score of 3-6), symptomatic intracranial hemorrhage, hemorrhagic transformation of infarct, and mortality. Multivariate analyses were performed to explore the relationships of inflammation-based scores with various clinical outcomes. RESULTS: Patients with unfavorable outcome showed higher mean NLR and MHR but lower mean LMR than those with favorable outcome (NLR, 7.32 vs. 3.78, P <= 0.001; MHR, 1.42 vs. 1.15, P = 0.012; LMR, 2.76 vs. 3.70, P = 0.003). In multivariate analysis, higher NLR (>= 5.1) (odds ratio [OR], 1.58; 95% confidence interval [CI], 1.04-2.12; P = 0.014) and higher MHR (>= 1.4) (OR, 1.32; 95% CI, 1.10-1.74; P = 0.028), lower LMR (<2.5) (OR, 1.28; 95% CI, 1.08-1.58; P = 0.032) were independently associated with unfavorable outcome. CONCLUSIONS: After MT, higher NLR, higher MHR, and lower LMR were found in patients with unfavorable outcome. A simple metal-ligand cooperative approach for the dehydrogenative functionalization of alcohols to various substituted quinolines and quinazolin-4(3H)-ones under relatively mild reaction conditions (<= 90 degrees C) is reported. Simple and easy-to-prepare air-stable Cu(II) complexes featuring redox-active azo-aromatic scaffolds, 2-arylazo-(1,10-phenanthroline) (L-1,L-2), are used as catalyst. A wide variety of substituted quinolines and quinazolin-4(3H)-ones were synthesized in moderate to good isolated yields via dehydrogenative coupling reactions of various inexpensive and easily available starting materials under aerobic conditions. A few control experiments and deuterium labeling studies were carried out to understand the mechanism of the dehydrogenative coupling reactions, which indicate that both copper and the coordinated azo-aromatic ligand participate in a cooperative manner during the catalytic cycle. In this study, in-situ synthesis of zinc oxide (ZnO) nanoparticles was utilized as an effective approach to prepare poly(vinyl alcohol) (PVA) and poly(vinyl alcohol)-graft-acrylonitrile/2-hydroxyethyl methacrylate (PVA-g-AN/HEMA) membranes with ZnO nanoparticles. The membranes were characterized by using Frourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), differential scanning calorimetry (DSC), X-Ray diffraction (XRD) and contact angle measurements. Both of the membranes were utilized in separation of acetone/water mixtures via pervaporation technique and the optimum conditions regarding ZnO content within the membranes, acetone content in the feed solution and operating temperature were determined. PVA-g-AN/HEMA-ZnO nanocomposite membranes showed high permeation rate but low separation factor compared to PVA-ZnO nanocomposite membranes. Permeation rate and separation factor values were found to be in the range of 0.028-0.137 and 0.035-0.174 kg m(-2) h(-1), 17.61-325 and 9.4-84.5 depending on the pervaporation conditions for PVA-ZnO and PVA-g-AN/HEMA-ZnO nanocomposite membranes, respectively. Background: Aloe vera is commonly used in the primary health care of human beings since time immemorial. It is an herb widely used in various traditional systems of medicine worldwide. Systematic and scientific investigation on A. vera as a medicinal plant has drawn considerable attention, and many laboratories are involved in isolation, characterization and evaluation of phytoconstituents for their nutraceutical and pharmaceutical applications. Purpose: The aim of this study was to provide an overview of the phytochemical, biological and medicinal attributes of A. vera against various diseases with special emphasis on underlying mechanisms of action. Methods: PubMed, EBOSCO host, Science Direct, Scopus, and Cochrane library databases were utilized to search literature published between1977 and 2019 (fill March). Major keywords used in various combinations included: Aloe vera, phytochemistry, metabolism, pharmacological activity, prevention, treatment, health, disease, in vivo, in vitro, and clinical studies. Results: Various biological and pharmacological activities of A. vera, such as antioxidant, anti-inflammatory, immuno-modulatory, antimicrobial, antiviral, antidiabetic, hepatoprotective, anticancer, and skin-protective and wound-healing responses, have been attributed to the presence of many active compounds, including anthraquinones, anthrones, chromones, flavonoids, amino acids, lipids, carbohydrates, vitamins and minerals. Conclusion: Based on various preclinical studies, A. vera constituents have enormous potential to prevent and treat various diseases. Randomized clinical trials are needed to understand the full therapeutic potential of this unique medicinal plant.

Facile green synthesis of BiOCl-TiO2 was done using combustion technique by Ixora coccinea leaf extract as fuel source. The said material was characterized using XRD, SEM, EDX, HRTEM, SAED, FTIR, and UV-DRS. The particle size was found to be approximately 60 nm and a crystallite size of 0.3 nm from TEM. The photocatalytic activity of the material was found out using photoluminescence studies, dye degradation and photocatalytic organic conversion. The material showed excellent dye degradation capacity for methylene blue with 80% of the dye degraded under 3 hrs. The stabilisation of electron-hole pair by the heterostructure gave it the ability to perform easy degradation. The degradation kinetics have also been studied. It also showed an excellent organic conversion property with formylation yield reaching up to 96% and total conversion of the reactant molecule. The material is a potent photocatalyst due to its great efficiency and can have a remarkable role in the synthesis of important organic molecules and detoxification of environment.Graphical AbstractThe heterostructure catalyses the conversion of amine to amides and mineralizes methylene blue under visible light condition.
