Background: Colorectal familial adenomatous polyposis (FAP) adenomas exhibit a uniform pathogenetic basis caused by a germline mutation in the adenomatous polyposis gene (APC), but the molecular changes leading to their development are incompletely understood. However, dysregulated apoptosis is known to substantially affect the development of colonic adenomas. One of the key regulatory proteins involved in apoptosis is apoptosis repressor with caspase recruitment domain (ARC). Methods: The expression of nuclear and cytoplasmic ARC in 212 adenomas from 80 patients was analyzed by immunohistochemistry. We also compared expression levels of ARC with the expression levels of p53, Bcl-2, COX-2, and MMR proteins. Statistical analyses were performed by Spearman’s rank correlation and linear regression test. Results: ARC was overexpressed in the nuclei and cytoplasm of most FAP adenomas investigated. Cytoplasmic ARC staining was moderately stronger (score 2) in 49.1% (n = 104/212) and substantially stronger (score 3) in 32.5% (n = 69/212) of adenomas compared to non-tumorous colorectal mucosa. In 18.4% (n = 39/212) of adenomas, cytoplasmic ARC staining was equivalent to that in non-tumorous mucosa. Nuclear expression of ARC in over 75% of cells was present in 30.7% (n = 65/212) of investigated adenomas, and nuclear expression in 10-75% of cells was detected in 62.7% (n = 133/212). ARC expression in under 10% of nuclei was found in 6.6% (n = 14/212) of adenomas. The correlation between nuclear ARC expression and cytoplasmic ARC expression was highly significant (p = 0.001). Moreover, nuclear ARC expression correlated positively with overexpression of Bcl-2, COX-2 p53 and beta-catenin. Cytoplasmic ARC also correlated with overexpression of Bcl-2. Sporadic MMR deficiency was detected in very few FAP adenomas and showed no correlation with nuclear or cytoplasmic ARC. Conclusions: Our results demonstrated that both cytoplasmic and nuclear ARC are overexpressed in FAP adenomas, thus in a homogenous collective. The highly significant correlation between nuclear ARC and nuclear beta-catenin suggested that ARC might be regulated by beta-catenin in FAP adenomas. Because of its further correlations with p53, Bcl-2, and COX-2, nuclear ARC might play a substantial role not only in carcinomas but also in precursor lesions.

Dialkylaluminium amides iBu(2)Al(TMP) and iBu(2)Al(HMDS) can perform catalytic hydroboration of ketones with pinacolborane to form the expected boronic esters. However, repeating the same reactions stoichiometrically without a ketone leads unexpectedly to ring-opening of pinacolborane and insertion of its open chain into the Al-N(amido) bond. To date there has been limited knowledge on decomposition pathways of HBpin despite its prominent role in hydroboration chemistry. X-ray crystallography shows these mixed Al-B products [iBu(2)Al{OC(Me)(2)C(Me)(2)O}B(H)(NR2)](2) (NR2=TMP or HMDS) form dimers with an (AlO)(2) core and terminal B-N bonds. Since the bond retention (B-H) and bond breaking (B-O) in these transformations seemed surprising, DFT calculations run using M11/6-31G(d,p) gave an energy profile consistent with a sigma-bond metathesis mechanism where London dispersion interactions between iBu and (amide) Me groups play an important stabilising role in the final outcome.

An easy green synthetic approach for fused isoindoloquinazolinones has been developed under neat reaction (yields up to 91%) conditions. This new one-pot tandem methodology involves condensation of readily available anthranilamide with 3-(2-formylcycloalkenyl)-acrylic ester under solvent- and catalyst-free conditions. This strategy avoids the use of oxidant, and heavy metal catalysts and also is free from work-up and generation of toxic by-products. A dramatic change of photophysical properties of dihydroisoindoloquinazolinones in basic and aqueous media has also been documented in our study. Moreover, our model synthetic compound shows cytotoxic activity towards metastatic HepG2 and PC3 cancer cell lines.

Actinobacteria are a major source of novel bioactive natural products. A challenge in the screening of these microorganisms lies in finding the favorable growth conditions for secondary metabolite production and dereplication of known molecules. Here, we report that Streptomyces sp. MBT27 produces 4-quinazolinone alkaloids in response to elevated levels of glycerol, whereby quinazolinones A (1) and B (2) form a new sub-class of this interesting family of natural products. Global Natural Product Social molecular networking (GNPS) resulted in a quinazolinone-related network that included anthranilic acid (3), anthranilamide (4), 4(3H)-quinazolinone (5), and 2,2-dimethyl-1,2-dihydroquinazolin-4(3H)-one (6). Actinomycins D (7) and X2 (8) were also identified in the extracts of Streptomyces sp. MBT27. The induction of quinazolinone production by glycerol combined with biosynthetic insights provide evidence that glycerol is integrated into the chemical scaffold. The unprecedented 1,4-dioxepane ring, that is spiro-fused into the quinazolinone backbone, is most likely formed by intermolecular etherification of two units of glycerol. Our work underlines the importance of varying the growth conditions for the discovery of novel natural products and for understanding their biosynthesis.

The aromatic substrate profile of the cobalt nitrile hydratase from Rhodococcus rhodochrous ATCC BAA 870 was evaluated against a wide range of nitrile containing compounds (>60). To determine the substrate limits of this enzyme, compounds ranging in size from small (90 Da) to large (325 Da) were evaluated. Larger compounds included those with a bi-aryl axis, prepared by the Suzuki coupling reaction, Morita-Baylis-Hillman adducts, heteroatom-linked diarylpyridines prepared by Buchwald-Hartwig cross-coupling reactions and imidazo[1,2-a]pyridines prepared by the Groebke-Blackburn-Bienayme multicomponent reaction. The enzyme active site was moderately accommodating, accepting almost all of the small aromatic nitriles, the diarylpyridines and most of the bi-aryl compounds and Morita-Baylis-Hillman products but not the Groebke-Blackburn-Bienayme products. Nitrile conversion was influenced by steric hindrance around the cyano group, the presence of electron donating groups (e.g., methoxy) on the aromatic ring, and the overall size of the compound.

A sp(3) C-H bond functionalization and C – C bond cleavage were realized by AIBN/0 2 catalyst system, providing a series of benzophenones under mild reaction conditions. The mechanistic study shows that a peroxide intermediate is involved in this transformation, and in the case of diphenylmethanes, the sp(3) C-C bond is cleaved through the peroxide rearrangement, which might provides a new way to cleave relatively strong C-C bond and be applied to more general C-C bond activation. The tumor suppressor transcription factor CCAAT enhancer-binding protein alpha (C/EBP alpha) expression is down-regulated in myeloid leukemias and enhancement of C/EBP alpha expression induces granulocytic differentiation in leukemic cells. Previously we reported that Styryl quinazolinones induce myeloid differentiation in HL-60 cells by upregulating C/EBP alpha expression. To identify more potent molecule that can induce leukemic cell differentiation we synthesized and evaluated new series of styryl quinazolinones, ethynyl styryl quinazolinones, styryl quinolinones and thienopyrimidinones. Thienopyrimidinones were found toxic and styryl quinolinones were found inactive. Ethynyl styryl quinazolinone 39 and styryl quinazolinone 5 were found active on par with the earlier reported analogues 1 and 2 suggesting that the 5-nitro furan-2-yl styryl quinazolinones find a real promise in leukemic cell differentiation. The improved potency of 5 suggested that further modifications in the 5-nitro furan-2-yl styryl quinazolinones can be at the phenyl substitution at the 3-position of the quinazolinone ring apart from the 5-position of the heteroaryl ring.

Some antagonistic bacteria contribute to the management of plant diseases by stimulating the host natural defense and/or by providing direct biocontrol of pathogens. The objective of this study was to evaluate the biocontrol performances and the ability to induce a systemic response to crown gall of four strains belonging to Bacillus and Pseudomonas genera. Evaluation of in vitro antagonistic ability showed that the four bacteria had a different spectrum of activity against the three tested strains of Agrobacterium tumefaciens. A delay in time of appearance and a significant reduction of the tumor size were observed in tomato plants obtained by bio-primed seeds and stem inoculated with A. tumefaciens. Seed priming with antagonistic strains stimulated some systemic defense mechanisms in tomato plants that can be related to the reduction of disease symptoms induced by A. tumefaciens. Both antagonists and A. tumefaciens strains induced variations in phenol content and peroxidase activity while polyphenol oxidase activity was mainly affected by the single A. tumefaciens strain. The response was different in relation to the various combinations of antagonist/pathogen, however Pseudomonas brassicacearum EPR3 strain was the most effective strain.

A series of novel quinazolines as tubulin inhibitors occupying three zones of colchicine domain have been designed and synthesized inspired by the recently disclosed crystal structure of verubulin analogue 6 with tubulin. Among the newly synthesized compounds, 19c showed noteworthy potency against K562, HepG2, KB, HCT-8 and MDB-MB-231 cancer cells. In vitro microtubule polymerization assays identified 19c as a potent tubulin assembly inhibitor, the binding mode of which with tubulin was confirmed by molecular modeling studies to occupy three zones of tubulin domain. Furthermore, 19c disrupted the intracellular microtubule network, caused G2/M phase arrest, induced cell apoptosis and depolarized mitochondria of K562 cells. 19c also reduced the cell migration and disrupted the capillary-like tube formation of human umbilical vein endothelial cells (HUVECs). Importantly, 19c significantly and dose dependently inhibited tumor growth in H22 liver cancer xenograft mouse model. All these results suggested that 19c deserves further research as a novel and potential anti-tubulin agent for the treatment of cancers.

We report detailed Rainan spectra for the neutral and radical anion forms of benzophenone, fluorenone, 2,2′-bipyridyl, 4,4′-di-tert-butyl-2,2′-dipyridyl, and anthracene. Density functional theory (DFT) predictions for the Raman spectra of these molecules give additional insight into the assignment of each vibrational mode. While the use of DFT has been problematic in quantifying the thermochemistry of highly delocalized radicals, we find that DFT-predicted spectra using the popular B3LYP functional are in excellent agreement with the observed Raman spectra. In the case of the two bipyridyl compounds, the Raman spectra allowed us to conclude that the cis form of the radical anion complexed to a sodium cation was the preferred configuration. Benzophenone and fluorenone radical anions gave a significantly weakened C=O bond stretching vibrational frequency as expected from the population of an antibonding pi* orbital. For benzophenone, the C=O vibration dropped from 1659 to 1403 cm(-1) upon reduction. Similarly, fluorenone showed a C=O vibration observed at 1719 cm(-1) for the neutral form that decreased to 1522 cm(-1) for the radical anion. The structurally rigid anthracene showed relatively smaller Raman band shifts upon single-electron reduction as the pi* orbital is more equally delocalized on the entire structure. In total, we correlated 65 DFT-predicted vibrational modes for the neutral molecules with an overall error of 7.1 cm(-1) (root-mean-square errors (RMSEs)) and 67 DFT-predicted vibrational modes for radical anions with an overall error of 9.9 cm(-1). These comparisons between theory and experiment are another example to demonstrate the power of DFT in predicting the identity and geometry of molecules using Raman spectroscopy.
