PRMT4 is a type I protein arginine methyltransferase and plays important roles in various cellular processes. Overexpression of PRMT4 has been found to be involved in several types of cancers. Selective and in vivo effective PRMT4 inhibitors are needed for demonstrating PRMT4 as a promising therapeutic target. On the basis of compound 6, a weak dual PRMT4/6 inhibitor, we constructed a tetrahydroisoquinoline scaffold through a cut-and-sew scaffold hopping strategy. The subsequent SAR optimization efforts employed structure-based approach led to the identification of a novel PRMT4 inhibitor 49. Compound 49 exhibited prominently high potency and selectivity, moderate pharmacokinetic profiles, and good antitumor efficacy in acute myeloid leukemia xenograft model via oral administration, thus demonstrating this compound as a useful pharmacological tool for further target validation and drug development in cancer therapy.

In soluble methane monooxygenase enzymes (sMMO), dioxygen (O-2) is activated at a diiron(II) center to form an oxodiiron(IV) intermediate Q that performs the challenging oxidation of methane to methanol. An analogous mechanism of O-2 activation at mono- or dinuclear iron centers is rare in the synthetic chemistry. Herein, we report a mononuclear non-heme iron(II)cyclam complex, 1-trans, that activates O-2 to form the corresponding iron(IV)-oxo complex, 2-trans, via a mechanism reminiscent of the O-2 activation process in sMMO. The conversion of 1-trans to 2-trans proceeds via the intermediate formation of an iron(III)-superoxide species 3, which could be trapped and spectroscopically characterized at -50 degrees C. Surprisingly, 3 is a stronger oxygen atom transfer (OAT) agent than 2-trans; 3 performs OAT to 1-trans or PPh 3 to yield 2-trans quantitatively. Furthermore, 2-trans oxidizes the aromatic C-H bonds of 2,6-di-tert-butylphenol, which, together with the strong OAT ability of 3, represents new domains of oxoiron(IV) and superoxoiron(III) reactivities.

A Rh-catalyzed pot and step economic synthesis of aza-polycyclic aromatic hydrocarbons (N-PAHs) from readily available aryl ketones and alkynes has been disclosed. Additionally, a novel synthetic application of the well-known aminating reagent hydroxylamine-O-sulfonic acid (HOSA) has been explored as an in situ redox-neutral directing group for the formation of N-PAHs via isoquinoline. Lymphomas (cancers of the lymphatic system) account for 12% of malignant diseases worldwide. Burkitt’s lymphoma (BL) is a rare form of non-Hodgkin’s lymphoma in which the cancer starts in the immune B-cells. We report the synthesis and preliminary studies on the antiproliferative activity of a library of 9,10-dihydro-9,10-ethanoanthracene based compounds structurally related to the antidepressant drug maprotiline against BL cell lines MUTU-1 and DG-75. Structural modifications were achieved by Diels-Alder reaction of the core 9-(2-nitrovinyl)anthracene with number of dienophiles including maleic anhydride, maleimides, acrylonitrile and benzyne. The antiproliferative activity of these compounds was evaluated in BL cell lines EBV- MUTU-1 and EBV+ DG-75 (chemoresistant). The most potent compounds 13j, 15, 16a, 16b, 16c, 16d and 19a displayed IC50 values in the range 0.17-0.38 mu M against the BL cell line EBV- MUTU-1 and IC50 values in the range 0.45-0.78 mu M against the chemoresistant BL cell line EBV+ DG-75. Compounds 15, 16b and 16c demonstrated potent ROS dependent apoptotic effects on the BL cell lines which were superior to the control drug taxol and showed minimal cytotoxicity to peripheral blood mononuclear cells (PBMCs). The results suggest that this class of compounds merits further investigation as antiproliferative agents for BL.

A novel ionic liquid modified graphene oxide supported Mo-complex [GO@IL/MoO2(acae)(2)] was successfully prepared, characterized and its catalytic application was investigated. The GO@IL/MoO2(acae)(2) catalyst was prepared through the surface modification of graphene oxide with alkyl imidazolium chloride followed by treatment with molybdenyl acetylacetonate. The GO@IL/MoO2(acae)(2) catalyst was characterized by using FT-IR, PXRD, TGA, EDX, and SEM analyses. This catalyst showed high activity in the green production of pyrazolo[b] phthalazine dione derivatives under ultrasonic conditions. The designed catalyst was effectively recovered and successfully reused at least eight times without significant loss in its efficiency.

Two new naphthalenone derivatives, 5-hydroxy-4-oxo-2-(2-oxopropyl)-1,2,3,4-tetrahydronaphthalen-1-yl acetate (1) and 5-hydroxy-2-(2-hydroxypropyl)naphthalene-1,4-dione (2), together with two new anthrone derivatives, (S)-2,5-dihydroxy-2-methyl-1,2,3,4-tetrahydroanthracene-9,10-dione (3) and 4,5-dihydroxy-2-methyl-9H-xanthen-9-one (4), were isolated from the fermentation broth of endophytic Micromonospora sp. A new aromatic diamine, 10,10-bis [4-(4-amino-2-pyridinoxy)phenyl]-9(10H)-anthracenone (BAPPA) was synthesized from anthrone as the starting material, and the corresponding pyridine- and anthrone-containing poly(ether amide)s (PEAs) were prepared from BAPPA with various aromatic diacid chlorides via a low-temperature solution polycondensation. The resulting PEAs having number-average molecular weights (M-n) of 29,500-33,400 g/mol with the polydispersity index ranged from 1.75 to 1.84, were all amorphous, and displayed excellent thermal stability with the glass transition temperatures of 263-355 degrees C, the temperatures of 415-441 degrees C at 5% weight loss and the higher than 50% residues at 800 degrees C in nitrogen. These new PEAs were soluble in polar solvents such as N-methyl-2-pyrrolidone, N,N-dimethylacetamide and pyridine, even almost in tetrahydrofuran at room temperature and could form transparent, strong and flexible films with tensile strengths of 84-97 MPa, tensile moduli of 2.1-3.2 GPa and elongations at break of 4-13%. Also, the present PEAs exhibited high optical transparence with the UV cutoff wavelength in the 342-362 nm range, and 83-90% transmittance at 500 nm. These results indicated the introduction of anthrone and pyridine groups into the backbone could improve the solubility and transparence of polyamides.

Polycyclic aromatic hydrocarbons are difficult substrates for hydrogenation because of the thermodynamic stability caused by aromaticity. We report here the first chromium- and cobalt-catalyzed, regiocontrolled hydrogenation of polycyclic aromatic hydrocarbons at ambient temperature. These reactions were promoted by low-cost chromium or cobalt salts combined with diimino/carbene ligand and methylmagnesium bromide and are characterized by high regioselectivity and expanded substrate scope that includes tetracene, tetraphene, pentacene, and perylene, which have rarely been reduced. The approach provides a cost-effective catalytic protocol for hydrogenation, is scalable, and can be utilized in the synthesis of tetrabromo- and carboxyl-substituted motifs through functionalization of the hydrogenation product. The systematic theoretical mechanistic modelings suggest that low-valent Cr and Co monohydride species, most likely from zerovalent transition metals, are capable of mediating these hydrogenations of fused PAHs.
